    Notes: This study investigates the influence of the source of R&D funds and management ownership on R&D productivity. The lagged effect of the source of R&D funds on R&D output is investigated for a sample of US manufacturing firms in five industries over the 1996–99 period. Estimates based on 779 firm-years show that R&D productivity increases with the proportion of stock held by managers and directors of firms primarily in the Other Electronics industry. The estimates also show that recipients of government-sponsored R&D funds in the Chemicals industry have lower levels of output (sales) for each dollar committed to R&D. In addition, output for firms in the Chemicals industry worsens as management stockholding increases, implying an agency cost rationale for the observed difference in output. The implication is that firms with high manager-owner content are less productive with government-sponsored R&D than with company-financed R&D. The reported results suggest that potential agency costs should be incorporated in government-sponsored R&D contracts. It also suggests that the source of R&D funds should be disclosed and incorporated into the valuation of intangible assets attributable to research and development.

    Notes: Cyclin-dependent kinase 5 (Cdk5) is emerging as a neuronal protein kinase involved in multiple aspects of neurotransmission in both post- and presynaptic compartments. Within the reward/motor circuitry of the basal ganglia, Cdk5 regulates dopamine neurotransmission via phosphorylation of the postsynaptic signal transduction pathway integrator, DARPP-32 (dopamine- and cyclic AMP-regulated phosphoprotein, Mr 32 000). Cdk5 has also been implicated in regulating various steps in the presynaptic vesicle cycle. Here we report that Cdk5 phosphorylates tyrosine hydroxylase (TH), the key enzyme for synthesis of dopamine. Using phosphopeptide mapping, site-directed mutagenesis, and phosphorylation state-specific antibodies, the site was identified as Ser31, a previously defined extracellular signal-regulated kinases 1/2 (ERK1/2) site. The phosphorylation of Ser31 by Cdk5 versus ERK1/2 was investigated in intact mouse striatal tissue using a pharmacological approach. The results indicated that Cdk5 phosphorylates TH directly and also regulates ERK1/2-dependent phosphorylation of TH through the phosphorylation of mitogen-activated protein kinase kinase 1 (MEK1). Finally, phospho-Ser31 TH levels were increased in dopaminergic neurons of rats trained to chronically self-administer cocaine. These results demonstrate direct and indirect regulation of the phosphorylation state of a Cdk5/ERK1/2 site on TH and suggest a role for these pathways in the neuroadaptive changes associated with chronic cocaine exposure.

    Notes: Glial cell line-derived neurotrophic factor (GDNF) improves motor dysfunction associated with aging in rats and non-human primates, in animal models of Parkinson's disease, and may improve motoric function in patients with advanced Parkinson's disease. These improvements are associated with increased dopamine function in the nigrostriatal system, but the molecular events associated with this increase are unknown. In these studies, 100 µg of GDNF was injected into the striatum of normal aged (24-month-old) male Fischer 344 rats. The protein levels and phosphorylation of TH, ERK1/2, and related proteins were determined by blot-immunolabeling of striatum and substantia nigra harvested 30 days after injection. In GDNF-treated rats, TH phosphorylation at Ser31 increased ∼40% in striatum and ∼250% in the substantia nigra. In the substantia nigra, there was a significant increase in ERK1 phosphorylation. In striatum, there was a significant increase in ERK2 phosphorylation. Microdialysis studies in striatum showed that both amphetamine- and potassium-evoked dopamine release in GDNF recipients were significantly increased. These data show that GDNF-induced increases in dopamine function are associated with a sustained increase in TH phosphorylation at Ser31, which is greatest in the substantia nigra and maintained for at least one month following a single striatal administration of GDNF. These findings, taken from the nigrostriatal system of normal aged rats, may help explain the long lasting effects of GDNF on dopamine function and prior studies supporting that a major effect of GDNF involves its effects on dopamine storage and somatodendritic release of dopamine in the substantia nigra.

    Notes: The machinability of a thermoplastic compound that consisted of 52 vol% ceramic particles and 48 vol% thermoplastic binders was investigated. To investigate the machining mechanisms, various blends of a ductile polymer (ethylene ethyl acrylate (EEA)) and a brittle polymer (isobutyl methacrylate (IBMA)) were used as thermoplastic binders. The fraction of IBMA to EEA in the blend was increased from 0 to 50 vol%. As the IBMA content was increased, the thermoplastic compound exhibited a stiffer stress versus strain response under compression because of the brittle nature of the IBMA polymer. The machinability of the thermoplastic compound was remarkably improved with increased IBMA content because of the mitigation of the extensive deformation of the thermoplastic compound.

    Notes: A new ceramic freeze-casting technique capable of manufacturing near room temperature with a sublimable vehicle was accomplished. Fluid-concentrated slurries of Al2O3 powder in molten camphene (C10H16) were prepared at 55°C. These slurries were quickly solidified (frozen) at room temperature to yield rigid solid green bodies, followed by frozen camphene removal by sublimation (freeze-drying) with negligible shrinkage. Sintering without any special binder burnout process yielded sintered bodies with over 98% theoretical density. The proposed advantages include (1) elimination of extremely cold temperatures, (2) elimination of troublesome binder burnout process, and (3) fast manufacturing cycle due to quick solidification.

    Notes: Structure and formation of aluminosilicate hydrogels were investigated as a function of chemical composition. Oscillatory rheometry was used to measure the dynamic moduli, G‘ and G“, of the gels during setting. The formation of aluminosilicate hydrogels was monitored over time as a function of the storage modulus for gels of low solids volumes, φ 〈 0.030. Overall gel strength correlated well to the fractal dimension, D, observed by neutron scattering. Scattering data were interpreted using the power law, and the Guinier relationship was used to determine the size of the constructing colloidal/gel particles. Aluminosilicate hydrogels were observed to consist of large fractal aggregates of indeterminable size constructed from smaller subunits of ∼45 Å in dimension. Structure and formation of the gels were highly dependent on [Al2O3] and [Na2O] content.

    Notes: The indirect solid freeform fabrication process is used to fabricate low-loss complex geometry periodic dielectrics for use in the microwave range. Relevant processing details, such as mold fabrication, slurry rheology, curing, and binder burnout, are discussed. Woodpile electromagnetic bandgap structures exhibiting a −40 dB well were shown to be operational at frequencies 〉110 GHz. Novel designs of planar periodic dielectric resonators with Q= 965 at 33 GHz (Q= ratio of the power stored to power lost in the resonator) were realized in alumina. Metallo-dielectric filters with low-insertion loss were also created via ISFF; modeling of these structures using resistor-inductor-capacitor-equivalent circuit analysis was performed to calculate a Q of 645.

    Notes: Freeze casting for Al2O3 was accomplished at room temperature with nonaqueous sublimable vehicles in the naphthalene–camphor eutectic system with a eutectic temperature of 31°C. A fully dense sintered body (〉99.5% of theoretical density (T.D.)) was obtained with a eutectic composition vehicle, whereas at most 90% T.D. was obtained with an off-eutectic (i.e., hypo- or hypereutectic) composition vehicle due to formation of large uniquely shaped voids. Microstructural observation suggested that growing pro-eutectic crystals rejected the suspended Al2O3 particles to form large voids during the solidification process. At the eutectic composition, formation of fine lamellar microstructure in a solidified vehicle is considered to inhibit particle rejection resulting in large voids.

    Notes: Objective : Our aim was to evaluate the utility of transcutaneous CO2 (TC-CO2) monitoring in following the response to therapy in children and adolescents in diabetic ketoacidosis (DKA) requiring ICU admission.Methods : During therapy for DKA, TC-CO2 was monitored and compared with the serum bicarbonate values (HCO3−). As clinically indicated, serum electrolytes including serum HCO3− were drawn and at the same time, the TC-CO2 value was recorded. Using the TC-CO2 value and the equation (PaCO2 = 1.5 × HCO3− + 8), a calculated HCO3− value was determined. The measured serum HCO3− value was subtracted from the calculated HCO3− value and the absolute difference was recorded. The difference between the measured serum HCO3− and the calculated HCO3− was analysed using analysis of variance after stratification for serum HCO3− values and patient age ranges. Calculated to serum HCO3− values were compared using linear regression analysis. In addition, serum HCO3− values were plotted against TC-CO2 values to derive an equation from these data which would relate the TC-CO2 values to the measured serum HCO3−.Results : The cohort included 28 patients ranging in age from 1.6 to 21 years (9.4 ± 4.7 years) and in weight from 13 to 96 kg (35.6 ± 18.2 kg). Two additional patients were not included as the initial TC-CO2 values were inaccurate because of tissue hypoperfusion. In the remaining 28 patients, the absolute difference between the calculated bicarbonate and serum bicarbonate values was 1.5 ± 1.2 mmol·l−1. The difference was ≤2 mol·l in 74.4% of samples and ≤5 mmol·l−1 in 99.2%. No difference in the value between the calculated and measured serum HCO3− was present according to serum HCO3− value or patient's age. Linear regression analysis of calculated HCO3− against serum HCO3− revealed a slope of 0.95 and an r2 value of 0.88. Linear regression analysis using serum HCO3− against TC-CO2 resulted in the following equations: 〈displayedItem type="mathematics" xml:id="di-fml-1" numbered="no"〉〈mediaResource alt="image" href="urn:x-wiley:11555645:PAN1176:PAN_1176_mu1"/〉 Conclusions : TC-CO2 correlates with serum HCO3− and can be used to follow therapy in children and adolescents in DKA.

    Notes: Background : Flexible fiberoptic bronchoscopy is an important diagnostic tool for pediatric pulmonologists. Because of its favorable respiratory profile, ketamine has become a popular sedative for this procedure, but may be associated with unpleasant emergence reactions in the older child. Remifentanil is a newer, ultra-short acting opioid that has been shown to provide effective sedation and cough suppression for fiberoptic bronchoscopy when combined with intermittent propofol boluses. However, delivery of these agents as a combined, single infusion has not been described.Methods : Children ≥2 years of age undergoing fiberoptic bronchoscopy were enrolled. Remifentanil was mixed in a single syringe with undiluted propofol giving final drug concentrations of 10 mg·ml−1 of propofol and 15–20 μg·ml−1 of remifentanil. Sedation was induced with a bolus of approximately 0.1 ml·kg−1 of this mixture and maintained by titrating the drip throughout the procedure. Vital signs, sedative effectiveness, recovery patterns, and complications were prospectively recorded.Results : Fifteen patients aged 9.0 ± 5.3 years were sedated. Sedation was induced with 1.2 ± 0.4 mg·kg−1 propofol (2.4 ± 0.8 μg·kg−1 remifentanil) and maintained with 4.1 ± 1.8 mg·kg−1·h−1 propofol (0.13 ± 0.06 μg·kg−1·min−1 remifentanil). Five patients received low-dose ketamine to augment sedation. The maximal decrease in respiratory rate was 6.1 ± 5.3 b·min−1 (27.6 ± 21%) and no patient became hypoxemic. All procedures were completed easily without significant complication. Patients recovered to baseline 13.3 ± 8.5 min following infusion discontinuation.Conclusions : A remifentanil/propofol mixture provided effective sedation and rapid recovery in pediatric patients undergoing fiberoptic bronchoscopy.
