    Notes: The calcineurin inhibitors cyclosporine and tacrolimus are the cornerstone immunosuppressants used in solid organ transplantation. Studies investigating calcineurin (CaN) activity in renal transplanted patients have been published, but basic properties of the enzyme activity in healthy subjects remain to be described. The aim of this study was to investigate whether CaN displays circadian variation or sex difference is present in healthy subjects. Twenty subjects had blood samples drawn every 4 h for a 24-h period. CaN activity was determined in whole blood as the release of 32P from a phosphorylated peptide. Activity of the 32P was quantitated by liquid scintillation and results converted to units CaN utilizing a calibration curve. We found no circadian variation in CaN activity and no difference between the two sexes. The clinical importance of these findings is that blood samples for calcineurin activity can be drawn without taking the exact time of day into consideration, but only considering the time of drug intake.

    Notes: A 27-year-old female, after a fracture of the mandible treated with osteosynthesis and after a difficult extraction, developed osteomyelitis with fistulation and sequestration. Sequestrectomy and extraction of the teeth in the osleomyelitic area did not clear the patient of symptoms. A patch test to nickel turned out to be positive. After removal of the stainless steel cerclage in the mandible, clinical and radiographic changes returned to normal within 2 and 8 weeks, respectively. It is concluded that, whenever acute treatment docs not lake priority, patch tests comprising nickel, cobalt and chromium should he performed in patterns who are candidates for metal osteosynthesis.

    Notes: Intraperitoneal injection of nononcogenic lymphocytic choriomeningitis (LCM) virus in adult C3H mice causes a symptomless infection but stimulates specific cell-mediated and humoral immune responses. However, median survival time of virus-infected mice inoculated with syngeneic tumor cells was significantly shortened, and growth of semiallogeneic tumors was significantly enhanced. Cell-mediated cytotoxicity measured as chromium-51 release from labeled tumor cells was significantly suppressed but was recovered within 55 days after infection. The suppressed immune responsiveness could be conferred on a normal spleen cell population when activated in virus-infected recipients. Chronically LCM virus-infected mice showed an unimpaired cell-mediated immune response to tumor allografts.

    Notes: Summary Swelling of the left foot and changes in its vascular volume (VV) were studied in seven healthy subjects during 8 h of seated work without leg movements. Changes in total plasma volume (PV) were calculated from hematocrit values. Reference values (r.v.) were obtained during a working day requiring intermittent physical activity (walking). Significant changes during the first 4 h: the foot swelled by 3.5% (r.v.: 2.2%) and VV was reduced by 0.5% of the foot volume (r.v.: increased by 0.3%). Accordingly, the interstitial fluid volume (IFV) of the foot increased by 4.0% (r.v.: 1.9%). The loss of PV was 6.3%. During the last 4 h the only significant change was an increase in foot volume by 1.9%. It is concluded that (1) foot swelling should be corrected for changes in VV to obtain an exact measure of the change in IFV, (2) prolonged elevated pressure, assumed to occur in the feet during relaxed sitting, does not imply distension (“delayed compliance“) of the vascular system as previously suggested, (3) hemoconcentration seems to reach complete stability during the initial period of quiet sitting, (4) loss of PV during sedentary work may be avoided by a modest increase in leg activity.
