    Notes: S-nitroso-cysteinyl-glycine, a novel nitric oxide-adduct thiol compound, can be detected in the brain (2.3 ± 0.6 pmol/mg protein), and released following stimulation of sensory afferents to the rat ventrobasal thalamus in vivo (resting conditions 17 n m; stimulation: 186 n m). Iontophoretic application of CysNOGly (20–80 nA) onto thalamic neurons in vivo resulted in enhancements of excitatory responses to either NMDA or AMPA (182 ± 13.6% and 244 ± 27.8% of control values, n = 15). CysNOGly enhanced responses to stimulation of vibrissal afferents to 132 ± 2.2% (n = 7) of control values. In contrast, the dipeptide CysGly reduced responses of ventrobasal neurons to NMDA and AMPA (54 ± 8.4% and 55 ± 10.8% of control, n = 5). CysNOGly was also a potent activator of soluble guanylate cyclase in vitro. Moreover, we found that NMDA elevated CysNOGly levels in vitro and this stimulatory effect was reduced by inhibitors of the neuronal NO synthase and of the γ-glutamyl transpeptidase, suggesting that production of NO and CysGly is a prelude to CysNOGly synthesis. These findings suggest that the nitrosothiol CysNOGly plays a role in synaptic transmission in the ventrobasal thalamus. We propose a novel synaptic buffering mechanism where S-nitroso-cysteinyl-glycine serves to restrict the locus of action of nitric oxide and so increase its local availability for target delivery. This could lead to a change in neuronal responses favouring sensory transmission similar to that seen in wakefulness or arousal in order to locally enhance transmission of persistent sensory stimuli.

    Notes: The aim of the present investigation was to study the effect of saliva flow on chewing efficiency by means of measurements of salivary flow rates as well as of parameters of masticatory performance. For this reason, 26 patients (13 xerostomic and 13 controls) with similar individual characteristics (14 females aged 41·3 ± 12·2 years; 12 males aged 38·0 ± 10·9 years) without CMD were encouraged to chew different foods (jelly bears, peanuts and rusk). Resting and stimulated flow rates of whole saliva (WHL) were ascertained after the method of Dawes (1987); resting and stimulated flow rates of palatal saliva (PAL) were determined with the help of the method of Niedermeier and Huber (1989); time of mastication (TM) as well as the number (NC) and duration (DC) of chewing cycles and time for swallowing (TS) were assessed using video recording. The mean resting WHL was 0·12 ± 0·05 mL min−1; the mean stimulated WHL was 0·24 ± 0·09 mL min−1; the mean resting PAL was 8·29 ± 7·88 µL cm−2 min−1 and the mean stimulated PAL was 8·96 ± 8·34 μL cm−2 min−1. Statistical analysis (Pearson method) revealed a strong correlation between resting or stimulated PAL and TM (R=0·808 or 0·834 for rubber bears; R=0·877 or 0·900 for peanuts; R=0·840 or 0·882 for rusk), DC (R=0·763 or 0·785 for rubber bears; R=0·836 or 0·867 for peanuts; R=0·749 or 0·779 for rusk) and TS (R=0·877 or 0·902 for rubber bears; R=0·860 or 0·918 for peanuts; R=0·822 or 0·864 for rusk). Only for rusk, resting and stimulated PAL correlated fairly with NC (R=0·697 and 0·736). No sufficient correlation was found between resting or stimulated WHL and parameters of mastication. The results clearly demonstrate that mucous saliva flow rates significantly bias measurements of masticatory performance.

    Notes: Abstract In isolated guinea-pig parietal cells pretreated for 60 min with the H2-antagonist ranitidine, the antimitotic agents colchicine and vinblastine, the microfilament-disrupting agent cytochalasin B resulted in a concentration-dependent inhibition of histamine-stimulated acid secretion up to 80%. Ranitidine reduced histamine binding to the membrane located H2-receptor. The anti-cytoskeletal agents inhibited the cellular histamine uptake but did not effect the histamine methyltransferase activity which was significantly reduced by ranitidine. The data suggest that cytoskeletal elements like microtubules and microfilaments are of very specific functional significance not only in the secretory process of the parietal cell but also for cellular transport mechanisms.

    Description / Table of Contents: Zusammenfassung Es wird über statistische und faktorenanalytische Untersuchungen an 182 HAWIE-Protokollen männlicher Patienten von vier Krankheits-gruppen (Commotio cerebri, Contusio cerebri, symptomatische Epilepsie, sonstige hirnorganische Erkrankungen) und drei Altersgruppen (20–34 Jahre, 35–49 Jahre, 50–64 Jahre) berichtet. Zur Auswertung kamen sowohl die einzelnen Untertests als auch die Intelligenzquotienten des HAWIE. Als Vergleichsgruppe wurden die Standardisierungsstichproben von Wechsler u. Riegel herangezogen. Die Untersuchung ergab: 1. Der HAWIE vermag Beeinträchtigungen intellektueller Leistungs-funktionen zu objektivieren und zu qualifizieren, erlaubt aber keine Aussage über die Art der zugrundeliegenden hirnorganischen Störung und Schädigung. 2. Die sogenannte Hirnleistungsschwäche mit Verlangsamung und Umstellungserschwerung der Denk- und Handlungsabläufe scheint weniger in einer generellen Intelligenzminderung oder einer Störung in bestimmten Intelligenzbereichen als vielmehr in einer Störung der Integration mit Umstrukturierung des Funktionsgefüges und stärkerer Trennung einzelner Leistungsfunktionen bedingt zu sein. Weitere Untersuchungen zu dieser Frage sind angelaufen.
    Notes: Summary We discuss the statistical and factor-analytical evaluation of 182 HAWIE records obtained from patients suffering from 4 different cerebral disorders (Commotio cerebri, contusio cerebri, symptomatic epilepsy, and other organic diseases of the brain). These patients are divided into 3 different age groups (20 to 34 years, 35 to 49 years, 50 to 64 years). The results of 10 subtests and of the HAWIE intelligence quotient were evaluated. Conclusion: 1. HAWIE may verify and substantiate impairments of functions related to intellectual performance, but it does not permit any statement about the nature of the underlying organic lesion or injury of the brain. 2. The so-called insufficiency of mental performance comprises sluggish thinking and acting as well as adaptive difficulties. This insufficiency appears to be caused by an impaired integration resulting in altered functional structures and a stricter separation of individual functions maintaining mental performance rather than by a general decrease in intelligence. Further studies on this subject are in progress.

    Notes: A method for measurement of PGF2α, PGE1, PGE2, 6-keto-PGF1α, TXB2, 2,3-dinor-TXB2 as well as 5-, 8-, 9-, 11-, 12-, 15-HETE and HHT, utilizing negative ion chemical ionization GC/MS is presented. A highly efficient separation and purification procedure prior to the derivatization sequence allows quantification of the arachidonic acid metabolites described in two GC/MS runs. The detection limit was in the femtomole range. Application of the method to the quantitative profiling of arachidonic acid metabolites in various tissues and incubation media is demonstrated.
