    Notes: Abstract In response to bacterial infection epithelial cells up-regulate expression and secretion of proinflammatory cytokines. Previous work from our laboratory showed that basolateral infection of polarized T84 cells with Yersinia enterocolitica induces interleukin-8 (IL-8) secretion in the absence of significant invasion. Here we studied Y. enterocolitica-induced IL-8 secretion by epithelial HeLa cells as a function of Yersinia invasion or adhesion. For this purpose we tried to separated induction of IL-8 secretion from invasion by treating HeLa cells with signal transduction inhibitors prior to infection. While staurosporin and genistein inhibited both Yersinia invasion and Yersinia-triggered IL-8 secretion, wortmannin, an inhibitor of the phosphatidylinositol-3-phosphate kinase (PI3-K), blocked invasion of Y. enterocolitica into HeLa cells but did not show any effect on IL-8 secretion. These results suggest that Yersinia adhesion might be sufficient to induce IL-8 secretion by epithelial cells. Further analysis demonstrated the requirement of the Yersinia invasion locus inv for adhesion-mediated induction of IL-8 secretion. Thus, HeLa cells infected with an E. coli strain expressing the Y. enterocolitica inv locus induced IL-8 secretion in the presence and absence of wortmannin. Reverse transcription-polymerase chain reaction analysis revealed that adhesion of inv-expressing Y. enterocolitica or E. coli results in the transcriptional activation of the IL-8 gene. These results suggest that Y. enterocolitica adhesion to host cells via Inv activates de novo synthesis and secretion of IL-8.

    Description / Table of Contents: Summary In the following pilot project, conducted from February to August 1995 the spatial organization of red deer in the north German mountain range Harz was investigated using satellite telemetry. The respective locations of two males and one female were ascertained at regular intervals using a GPS receiver. These data were then transferred via the experimental satellite TUBSAT-A to the ground station in Berlin. The purpose of this study was twofold: to provide data on the size of the deer's home ranges and seasonal migrations in this area as well as to test the practical applicability of this technically advanced system for wildlife research purposes. The preliminary investigations resulted in an average error in spatial location of ca. 100 m (n = 52) using the GPS receivers. A total of 373 valid spatial locations were registered for the 3 animals telemetrically investigated, whereby 298 locations were recorded for Animal Nr. 2, a juvenile red deer female. Both male red deer proved to be relatively sedentary; Animal 1 used the environs of the winter feeding range as its home range from April–June, Animal 4 used this area from February to May. From March 24 to April 23 the female deer used the area around its winter feeding area as home range, before wandering off to an area 8 km northwest where it still was found in August. The sizes of the home ranges of the animals, calculated according to the minimal-area-method, were between 3.9 km2 and 8.8 km2. In this study satellite telemetry proved to be a practical, ecologically sound, and cost effective method for investigating the spatial locations over time of red deer in difficult terrain.
    Abstract: Résumé Dans ce projet-pilote, l'organisation spatiale de cerfs des moyennes-montagnes du Harz (Allemagne du Nord) a été étudiée de février à août 1995 par télémétrie au moyen de satellites. A cet effet, les positions d'un cerf femelle et de deux cerfs mâles ont été communiquées à intervalles réguliers via un récepteur GPS et transmises via les satellites expérimentaux TUBSAT-A à la station terrestre de Berlin. L'étude visait d'une part à fournir des premiers résultats sur l'étendue du home range et des déplacements saisonniers du Cerf dans cette région et, d'autre part, à vérifier la faisabilité, quant à son application à la recherche en faune sauvage, de la technique coûteuse de la télémétrie par satellite. Les recherches préliminaires firent apparaître une erreur moyenne de localisation du récepteur GPS d'environ 100 m (n = 52). Pour les trois sujets suivis à distance, on a recueilli 373 positionnements valables, dont 298 provenaient du cerf n° 2, un sujet femelle juvénile. Les deux cerfs mâles se révélèrent relativement sédentaires: le home range du cerf n° 1, d'avril à juin, et du cerf n° 4, de février à mai, concernait une zone proche d'un point d'affouragement hivernal. De son côté, le cerf femelle se tenait, du 24 mars au 23 avril, dans un secteur situé à proximité d'un point d'affouragement puis gagnait un secteur situé au Nord-Ouest, à quelque 8 km de là, où il se tenait encore au mois d'acoût. Les étendues du home range des 3 animaux atteignent, selon la méthode des surfaces minimales, entre 3,9 km2 et 8,8 km2. La télémétrie par satellite s'est révélée, dans la présente étude, d'une application écologiquement et économiquement intéressante en vue d'étudier le comportement spatio-temporal du Cerf dans un milieu d'accès difficile.
    Notes: Zusammenfassung In vorliegendem Pilotprojekt wurde von Februar bis August 1995 die Raumorganisation von Rothirschen in dem norddeutschen Mittelgebirge Harz mittels Satellitentelemetrie untersucht. Dazu wurden in regelmäßigen Abständen die Positionen von einem weiblichen und zwei männlichen Tieren über GPS-Empfänger ermittelt und über den Experimentalsatelliten TUBSAT-A an die Bodenstation in Berlin weitergeleitet. Die Untersuchung sollte einerseits erste Erkenntnisse über die Größe der Streifgebiete und saisonalen Wanderungen von Rothirschen in diesem Gebiet liefern, andererseits die technisch aufwendige Satellitentelemetrie auf Praxistauglichkeit in der Wildtierforschung prüfen. Die Voruntersuchungen ergaben einen mittleren Fehler in der Ortsbestimmung der GPS-Empfänger von rund 100 m (n = 52). Von den 3 telemetrierten Tieren liegen insgesamt 373 gültige Ortsbestimmungen vor, wobei 298 Positionen von Tier Nr. 2, einem juvenilen Rothirschweibchen, stammten. Die beiden Rothirsche waren relativ standorttreu, Tier 1 nutzte von April–Juni, Tier 4 von Februar–Mai als Streifgebiet (home range) die Umgebung der Winterfütterung. Auch das Rothirschweibchen bewegte sich vom 24. März bis 23. April in dem Gebiet um seine Winterfütterung, wanderte dann in ein anderes Gebiet 8 km nordwestlich, wo es sich noch im August aufhielt. Die Größen der home ranges der 3 Tiere betragen, nach der Minimal-Area-Methode berechnet, zwischen 3,9 km2 und 8,8 km2. Die Satellitentelemetrie hat sich in der vorliegenden Untersuchung als eine praxistaugliche, ökologisch schonende und kostengünstige Methode erwiesen, das Raum-Zeitverhalten von Rothirschen in schwierigem Gelände zu untersuchen.
