    Notes: Summary Electrophysiological and anatomical studies have suggested the existence of a pathway between the caudal raphé nuclei and regions of the spinal cord containing the sympathetic preganglionic neurons. However synaptic connections between cells in the raphé nuclei and identified sympathetic preganglionic neurons have not yet been shown. We have used a combination of anterograde tracing using Phaseolus vulgaris leucoagglutinin (PHA-L), retrograde tracing using a conjugate of cholera B chain and HRP and electron microscopy to look for such a pathway in rats. When PHA-L had been injected into the regions mainly restricted to the raphé pallidus and raphé magnus, synaptic contacts were found between PHA-L containing terminals and preganglionic neurons retrogradely labelled from the adrenal medulla. Out of the 43 synaptic contacts analysed, 26 were onto somata and 14 onto dendrites. 75% of the total appeared to have symmetric membrane specialisations, 20% asymmetric and the remainder could not be classified. Synaptic contacts were not seen in an animal in which the PHA-L injection site involved cells in the ventral raphé obscurus and surrounding gigantocellular reticular formation. These findings provide evidence of the existence of a direct monosynaptic pathway between cells in the raphé pallidus and/or caudal raphé magnus, and identified sympathetic preganglionic neurons and give further support for a role for the caudal raphé nuclei in sympathetic autonomic regulation.

    Notes: Summary Seedlings of nine tropical species varying in growth and carbon metabolism were exposed to twice the current atmospheric level of CO2 for a 3 month period on Barro Colorado Island, Panama. A doubling of the CO2 concentration resulted in increases in photosynthesis and greater water use efficiency (WUE) for all species possessing C3 metabolism, when compared to the ambient condition. No desensitization of photosynthesis to increased CO2 was observed during the 3 month period. Significant increases in total plant dry weight were also noted for 4 out of the 5 C3 species tested and in one CAM species, Aechmea magdalenae at high CO2. In contrast, no significant increases in either photosynthesis or total plant dry weight were noted for the C4 grass, Paspallum conjugatum. Increases in the apparent quantum efficiency (AQE) for all C3 species suggest that elevated CO2 may increase photosynthetic rate relative to ambient CO2 over a wide range of light conditions. The response of CO2 assimilation to internal Ci suggested a reduction in either the RuBP and/or Pi regeneration limitation with long term exposure to elevated CO2. This experiment suggests that: (1) a global rise in CO2 may have significant effects on photosynthesis and productivity in a wide variety of tropical species, and (2) increases in productivity and photosynthesis may be related to physiological adaptation(s) to increased CO2.

    Notes: Summary Intravenous infusion of the nucleoside adenosine stimulates respiration, probably at least partly by an action in the carotid bodies, and also potentiates the ventilatory response to hypoxia, suggesting that it might be involved in the control of breathing. Whether the effects of adenosine occur at concentrations likely to be achieved in vivo is unknown and was investigated in 7 patients with arterial catheters inserted for diagnostic purposes. During intravenous infusion of adenosine (Maximum dose per min: mean 130 μg kg−1) mean minute ventilation increased from 5.5 to 10.9 l min−1 while mean plasma adenosine concentration in the aortic arch increased from 0.07 to 1.2 μM. In 3 patients ventilation first increased without a detectable increase in aortic adenosine concentration, suggesting a possible intra-pulmonary effect of adenosine, although increased concentrations were apparent at higher doses. Micromolar concentrations of adenosine are probably achieved in vivo in tissues during hypoxia. The present results show that at such concentrations adenosine stimulates respiration and are consistent with the suggestion that adenosine release may mediate or modulate the ventilatory response to hypoxia. A possible intra-pulmonary effect of adenosine merits further study.

    Notes: A knowledge of flow behavior is important in the study of laminar flow in twin-screw extrusion processes to predict the velocity distribution and to understand the mixing process. The flow of a power law fluid in self-wiping twin-screw extruders is examined using a two-dimensional finite element analysis of a mid-channel section of intermeshing screws. Theory is compared with experiment using food biopolymer and plastic materials. Comparisons showing overprediction of throughputs, but similarities in behavior, suggest that this model could provide an upper limit for melt conveying. For most of the throughput range examined, pumping of intermeshing self-wiping screws appears to be almost independent of the power law flow index of the melt extruded, but the value of the flow index determines the degree of influence intermeshing has on the overall pressure gradient generated in the extruder.

    Notes: The scratch test is widely used to assess the adhesion of ceramic coatings deposited on cermet and alloy substrates by both physical vapour deposition and chemical vapour deposition (CVD). It has been observed during such tests that, depending on the coating type, complete coating removal from the scratch channels can be preceded by flaking of the coating at the edges of the channels. An attempt has been made to ascertain, for CVD coated powder metallurgy (PM) high speed steel (HSS) cutting tool inserts, whether a relationship exists between the presence/absence and type of pre-critical load coating flaking observed during scratch testing and interfacial compositional variations determined by Auger electron spectroscopy (AES). Scratch test and AES results are presented for the following CVD coated PM HSS inserts: titanium nitride (TiN) coated, where no flaking has been observed; titanium carbide (TiC) coated, where mixed adhesive/cohesive flaking of the TiC coating has been observed and multilayer aluminium oxide (Al2O3) coated, where adhesive flaking of the Al2O3coating has been observed. In the case of the TiN and TiC coated inserts it is not considered that differences in coating/substrate interdiffusion zone thickness alone are responsible for the observed divergence in scratch test behaviour. Differences in coating microhardness are also suggested to be a significant factor. The behaviour of the multilayer Al2O3 coated insert during scratch testing is thought more likely to be singularly associated with interfacial chemistry.
