    Notizen: Abstract Rats were implanted with stimulating electrodes aimed either at the medial forebrain bundle-lateral hypothalamus (MFB-LH) or the midbrain-central gray (MID-GG), and were trained to lever-press for brain self-stimulation on a fixed ratio: 15 schedule of reinforcement. The dose-dependent effects of morphine (0.1–3.0 mg/kg), naloxone (0.1–30 mg/kg), and naltrexone (0.1–30 mg/kg) were then determined during 1 h test sessions. Both naloxone and naltrexone decreased the rate of responding in the MFB-LH as well as in the MID-CG. However, decrements in response rates were produced in the MID-CG by both naloxone and naltrexone at one tenth the doses required to produce similar decrements with electrodes in the MFB-LH. Dose-dependent decreases in response rates produced by morphine occurred at the same doses in the two electrode sites. At both sites, the decreases in response rates produced by the highest dose of morphine were antagonized completely by a low dose of naloxone (0.1 mg/kg). At an intermediate dose of naloxone (1.0 mg/kg), antagonism occurred in the MFB-LH but not in the MID-CG. At a high dose of naloxone (10 mg/kg), a depression in lever-pressing occurred at both sites in the morphine-treated animal indicating that the depressive action predominated over antagonism. These data explain the lack of consistency of the effects of naloxone on brain self-stimulation previously reported by different laboratories, and demonstrate that the use of partial reinforcement schedules in a rational approach to the evaluation of opioid effects on brain self-stimulation behavior.

    Notizen: Abstract The acute effects of 5 neuroleptic drugs were tested in rats implanted with stimulating electrodes in the medial forebrain bundle and trained in a brain selfstimulation threshold procedure. Haloperidol (0.01–0.10 mg/kg) and loxapine (0.03–0.56 mg/kg) produced increases in reinforcement thresholds accompanied by reductions in response rates. Chlorpromazine (0.10–3.0 mg/kg) did not significantly alter reinforcement thresholds, but did produce dose-dependent reductions in response rates. Pimozide (0.1–1.75 mg/kg) was similar to chlorpromazine and significantly increased the reinforcement threshold only at the highest dose, although a graded decrease in response rates occurred over a wide dose-range. Clozapine (0.1–1.75 mg/kg) increased reinforcement thresholds without producing any significant changes in response rates, but when 3.0 mg/kg was administered, a marked disruption of behavior occurred. The results suggested that a distinction can be made between the effects of neuroleptics on motor behavior and on central reinforcement thresholds, and this may help in the interpretation of the relation between the chemical and clinical potency of antipsychotic drugs.

    Notizen: The rearrangements of 4′-methoxy-N-methyl- and N-methyl-4′-nitro-diatzoaminobenzene have been studied in 20% acetonitrile/aqueous buffers. The reactions are specifically acid catalyzed and involve pre-equilibrium formation of amine and diazonium salt followed by rate-limiting attack of the diazonium ion at a C-atom (C-coupling) to give the corresponding aminoazo compounds. There is no evidence to suggest that, under the present conditions, mechanisms other than the established Friswell-Green mechanism occur. The traditional two-stage synthesis of aminoazo compounds via isolated diazoamino compounds can therefore be replaced by a one stage process for amines which undergo initial attack at a N-atom by diazonium ion (N-coupling).

    Notizen: The parotid and the principal and accessory submandibular glands of the little brown bat, Myotis lucifugus (Vespertilionidae), were examined using light microscopy and staining methods for mucosubstances. The parotid gland is a compound tubuloacinar seromucous gland. Parotid gland secretory cells contain both neutral and nonsulfated acidic mucosubstances. The principal and accessory submandibular glands are compound tubuloacinar mucus-secreting glands. They contain somewhat atypical mucus-secreting demilunar cells that often appear to be interspersed between mucous tubule cells. The mucous tubule cells in both the principal and accessory submandibular glands contain sulfomucins. Demilunar cells of the principal submandibular gland contain moderate amounts of nonsulfated acidic mucosubstances, but the corresponding cells of the accessory submandibular gland contain considerable neutral mucosubstance with very little acid mucosubstance. Intercalated ducts composed of cuboidal or low columnar epithelial cells are present in all three glands. Striated ducts in all glands are composed of columnar cells whose apices bulge into the ductal lumina. Excretory ducts are composed of simple columnar epithelium, with occasional basal cells that suggest a possible pseudostratified nature. The cells of the excretory ducts also have bulging apices. All duct types contain apical cytoplasmic secretory material that is a periodic acid-Schiff positive, neutral mucosubstance. Ductal apical secretory material is more evident in intercalated and striated ducts than in excretory ducts.

    Notizen: Previous studies in this laboratory have shown that membranes derived from dorsal root ganglia (DRG) neurites are mitogenic for cultured Schwann cells derived from the same source [Salzer et al (1980): J Cell Biol 84:767-778]. Improved procedures are described for preparing Schwann cells derived from dorsal root ganglia that are highly responsive to various mitogens. Under these conditions, the cells respond not only to the neurite mitogen but also to pituitary extracts, dibutyryl cyclic AMP, and cholera toxin that have been shown previously to be good mitogens for Schwannn cells derived from sciatic nerve [Raff et al (1978): Cell 15:813-822], thus reconciling discrepancies in the response of these different Schwann cell preparations to mitogens. Searching for a source of membranes more suitable for biochemical characterization of the neurite mitogen, we found that bovine brain axolemma, prepared by the method of DeVries et al [(1977): Brain Res 147:339-352] is highly mitogenic for Schwann cells. The milotic index of Schwann cells was increased by the addition of axolemma from 0.5%-2% to 30%-50% during 24-h incubation with [3H]thymidine. Half maximal effect was obtained at about 0.4 μg axolemma protein per microwell containing 2-4 × 10 3 cells. The axolemma mitogen appears to be an integral membrane protein that remains bound to the membrane under various ionic conditions but can be extracted in a partially active form with deoxycholate. Like the DRG neurite mitogen, the mitogenic activity of axolemma was abolished by trypsin treatment. Unlike the neurite preparation, however, the mitogenic activity of axolemma was only partially inactivated by heat treatment (60%-70% inactivation). A significant difference between the mitogenic activity of axolemma membranes and neurite membranes is the fact that axolemma membranes fail to stimulate Schwann cell proliferation in a defined, serum-free medium (N-2), whereas neurites show significant mitogenic activity in this medium. These findings indicate a possible difference between DRG neurites and brain axolemma either in the mitogen itself or surface components responsible for recognition between the membranes and the cells.

    Notizen: The ultrastructure of the parotid gland was examined in the little brown bat. The seromucous acinar cells contained abundant granules of variable morphology. These granules were characterized by a submembranous dense layer consisting of fine parallel slats. In some bats, the matrix of the granules was structureless, whereas in others it consisted of closely packed but randomly arranged bundles of tubules. The intercalated ducts had a highly developed rough endoplasmic reticulum, often containing large numbers of intracisternal granules. In contrast, only a few secretory granules were present in the supranuclear cytoplasm. The striated ducts, which exhibited the characteristic basal striations consisting of vertically oriented mitochondria and highly folded plasmalemmas, contained numerous small dense granules in a subluminal band. These granules had a paracrystalline substructure with a periodicity of 8 nm. Excretory ducts strongly resembled striated ducts. They showed the same kind of basal striations and about half their constituent cells contained small paracrystalline granules.

    Notizen: This paper is concerned with detecting crosslinking and changes in crosslink densities in cured polyimides. The methods used include diffusional weight gain and dynamic mechanical measurements on thin films of the cured polymers. In this study changes in diffusivities (D) for dimethylformamide (DMF) sorption into polyimide films as well as dynamic mechanical moduli and loss tangents are shown to be related to differences in polyimide cure schedule and composition. In all cases studied there is a β loss dispersion in the range 120 to 150°C and a γ loss dispersion in the range -50 to -80°C. The position and intensity of the β and γ relaxation processes vary primarily with cure cycle. There is a significant correlation between the magnitude of the diffusion coefficient and β and γ transition temperatures (Tβ and Tγ). Tβ increases and Tγ decreases with higher values of D so that T*, the difference Tβ - Tγ, is directly proportional to the diffusivity. By relating these results to data in the literature and the known curing behavior of polyimides, this trend is shown to be consistent with the presence of crosslinking and to be related to the crosslink density of the polymers. The data of this study indicate that measurements of diffusion coefficients for solvent sorption can be used to study changes in crosslink densities in glassy thermosetting polymers. Since the method is best suited for studying one dimensional diffusion, it also is suggested as a possibility for determining relative crosslink densities in adhesives bonded between substrates.

    Notizen: The chemisorption of SO2 on a W and a Pt surface has been investigated by flash desorption mass spectrometry (FDMS) and AES. AES data indicate that the chemisorbed sulfur dioxide molecules oxidized the W surface, and produced a stable tungsten oxide during the heating (up to 1400 K) process. FDMS data indicate that the decomposition of SO2 yielded elemental and molecular sulfur during the desorption process. FDMS data show that SO2 exists in four binding states (α1, α2, α3 and α4) on a clean Pt surface and in three binding states (α5, α6 and α7) on an oxidized Pt surface. In addition, the coadsorption of SO2 and O2 was carried out on a Pt surface. The coadsorption process can produce SO3 which exists in two different binding states on a Pt surface. The data indicate that the oxidation of SO2 to form SO3 on a Pt surface occurs by means of a Langmuir-Hinshelwood mechanism.
