    Notes: Summary Different levels of anaesthesia in cats surgically prepared under thiopentone sodium were induced by adding 0.2%, 0.8% and 2.2% halothane to 80% N2O, 19% O2 and 1% CO2 mixture. Properties of visual cortical neurones were investigated under each anaesthetic level. 80% N2O alone produced a combination of slow and fast wave components in the EEG (2nd stage of anaesthesia). Under this condition, many cortical neurones could be classified as “sustained” or “transient” on the basis of their firing pattern to a stationary bar, and sharp tuning curves for orientation and spatial frequency could be obtained, the cells having low contrast thresholds. As the halothane concentration was increased, the slow wave component of the EEG increased (3rd stage of anaesthesia), the tuning curves of cortical neurones to orientation and spatial frequency became less sharp, the contrast threshold was raised and classification into “sustained” and “transient” neurones became difficult. Finally the neurones became undrivable when the EEG showed continuous, large-amplitude slow waves (4th stage of anaesthesia). Thus findings made during the slow wave EEG patterns cannot reveal the full capabilities of cortical neurones.

    Notes: Summary Cell separation in abscission zones of explants of Phaseolus vulgaris L. is preceded by the enlargement of a single row of cells. Unlike the other cells surrounding them, the cells of this row enlarge in response to ethylene and not to auxin. Enlargement follows an increase in the activity of carboxymethyl 1: 4-glucanase in the abscission zone; also a response to ethylene and not to auxin. Eventual cell separation occurs between the enlarging cells and the non-enlarging tissue distal to them. It is proposed that the formation of cells at the abscission zone, with growth properties dissimilar to those of the rest of the pulvinus and petiole, is an example of precise positional differentiation. The presence and enlargement of such cells may be a general prerequisite for the abscission of organs or tissues distal to them.

    Notes: Synopsis A procedure is described for the histochemical demonstration of aniline hydroxylase activity in cryostat sections of rat liver. Tissue sections are incubated in a medium containing aniline; thep-aminophenol formed as a result of enzymatic action is coupledin situ with Fast Blue RR. The staining reaction is found to be confined to the cytoplasm of the hepatocytes. Confirmatory tests for true enzymatic staining reaction include the incubation of sections in medium from which aniline is omitted, and under conditions of enzyme inhibibition. A method for the quantitation of the histochemical staining reaction is also described. The histochemical reactions have been investigated on rat livers subjected to conditions eliciting microsomal enzyme stimulation and inhibition, bothin vitro andin vivo. A close correlation was found between the staining reactions observed and the results of the quantitative histochemical method and the biochemical estimations of aniline hydroxylase activity in liver microsomal fractions obtained by differential centrifugation.

    Notes: Synopsis The histochemical technique of Gangolli & Wright (1971) was used to detect aniline hydroxylase activity in a number of rat tissues. A high degree of activity was found in the adrenals (fasciculata and reticularis zones), the cortical tubules of the kidney, the epithelium of the small intestine, cardiac and ‘red’ striated muscle and the corpus luteum. Less activity was found in the placenta, the endometrium in pregnancy and the epithelium of the bronchi and large intestine. Weak activity was present in the anterior pituitary gland, smooth muscle of the intestine, uterus and bladder, skin and bladder epithelium and ‘white’ striated muscle. These was no activity in the thyroid and parathyroid, lymphoid tissue and endometrium. In all tissues the enzyme activity was denoted by a brownish deposit in the form of discrete small granules. The staining reaction was totally abolished when tissues were incubated in an atmosphere of a mixture of carbon monoxide and oxygen (80∶20 v/v), or in nitrogen, or by the omission of aniline from the incubation medium. Although it is not certain whether aniline hydroxylase activity in extrahepatic tissues has the same connotation and significance as microsomal hepatic aniline hydroxylase, its successful inhibition by carbon monoxide and nitrogen in a wide variety of tissues indicates that the histochemical demonstration of this enzyme may merit consideration in the study of chemically induced cellular damage in a variety of organs.

    Notes: Synopsis Methods have been developed for the analytical estimation and histochemical demonstration of carrageeman in the granuloma induced in rats and guinea-pigs by subcutaneous injection of degraded carrageenan. The analytical method for the determination of carrageenan in tissues involved a preliminary clean-up procedure. The tissues were defatted by solvent extraction and incubated with papain and trypsin to remove proteins. Carrageenan and naturally occurring acid mucopolysaccharides were isolated using cetyl pyridinium chloride. The subsequent separation and estimation of carrageenan was carried out by electrophoresis on cellulose acetate paper. Following electrophoresis the cellulose acetate strips were incubated with hyaluronidase to remove acid mucopolysaccharides, and stained with Toluidine Blue. The stained band corresponding to pure degraded carrageenan was quantitated on a scanning densitometer. The method was capable of detecting 0.25 μg of degraded carrageenan in tissue. The most suitable method for the histochemical demonstration of carrageenan in paraffin embedded tissues was found to be Alcian Blue at either pH 1 or a CEC (critical electrolyte concentration) of 1.0 M MgCl2 (pH 5.8). At this pH or CEC, both the carrageenan and the strongly acidic glycosaminoglycans were stained. Prior digestion with hyaluronidase and neuraminidase eliminated the staining of the tissue polysaccharides so that the carrageenan could be visualized within macrophages and in the extracellular space. Mast cell granules retained their staining properties after mucolytic digestion; but morphologically, mast cells could be distinguished from macrophages containing carrageenan.

    Description / Table of Contents: Zusammenfassung Die Oberflächenleitfähigkeit von Polystyrol-Latexpartikeln wurde in Abhängigkeit von der Konzentration in Kaliumchlorid, Bariumperchlorat, Aluminiumperchlorat und Hexolnitrat gemessen. Es besteht ein Anwachsen der Oberflächenleitfähigkeit mit wachsender Konzentration für die drei einfachen Elektrolyte. Dabei erreicht die Oberflächenleitfähigkeit anscheinend in jedem Elektrolyt denselben konstanten Wert. Die gemessene Oberflächenleitfähigkeit ist mehrere Größenordnungen höher als die für eineGouy-Schicht berechnete. Die Resultate wurden dahingehend interpretiert, daß sie auf einen großen Beitrag zur Gesamtleitfähigkeit durch eine fixierte Schicht und einen vernachlässigbaren Beitrag aus der diffusen Schicht hinweisen.
    Notes: Summary The surface conductivity of polystyrene latex particles has been measured in a range of concentrations of potassium chloride, barium perchlorate, aluminium perchlorate, and hexol nitrate. There was an increase of surface conductivity with increasing concentration for the three simple electrolytes; the surface conductivity eventually attained the same constant value in each electrolyte. The measured surface conductivity was several orders of magnitude larger than that calculated for theGouy layer; the results have been interpreted as indicative of a large contribution to the total surface conductivity from the fixed layer and a negligible contribution from the diffuse layer.

Die Beziehung F/ϱ ist unabhängig von der Natur des Elektrolyts und der Meßfrequenz im Bereich von 500 Hz zu 1,5 MHz.
    Notes: Summary The formation factor,F, required for the determination of the surface conductance at a solid/liquid interface, has been measured in solutions of potassium chloride, sodium chloride, sodium sulphate and barium perchlorate, some measurements were made at different frequencies. Provided electrode polarization is eliminated the variation ofF with the volume fraction of the solid, ϱ, is in complete agreement with that predicted theoretically byMaxwell. Further the F/ϱ relationship is independent of the nature of the suspending electrolyte and the frequency of measurement in the range 500 Hz to 1.5 MHz.
