    Notizen: Summary Exogenous glucagon or epinephrine were infused into normal overnight fasted dogs to raise circulating hormone levels to concentrations within the physiologic range. Plasma levels of glycerol and free fatty acids remained unchanged during the glucagon infusion, but rose significantly during the administration of epinephrine. Plasma insulin in the systemic circulation remained unchanged during the glucagon infusion and increased slightly during the infusion of the catecholamine. The data suggest that in normal dogs glucagon in physiological amounts has no lipolytic effect. The importance of the sympathetic nervous system in regulating lipolysis in normal mammals is stressed.

    Notizen: Summary In the isolated rat vas deferens, various 2-substituted adenosine derivatives and adenosine inhibited contractions elicited by field stimulation but had little effect on responses to exogenous noradrenaline. 2-Chloroadenosine, 2-bromoadenosine, 2-hydroxyadenosine and 2-fluoroadenosine were all more potent than adenosine. Theophylline antagonized the action of the 2-substituted derivatives. The inhibitory action of adenosine was potentiated by dipyridamole, 2-amino-6-[(2-hydroxy-5-nitro)benzylthio]-9-β-d-ribofuranosylpurine (HNBTG) or 2′-deoxycoformycin while that of 2-chloroadenosine was not altered by any of these drugs or by phenoxybenzamine, atropine or indomethacin. Pretreatment of the vas deferens with both HNBTG and 2′-deoxycoformycin eliminated the difference in inhibitory potency between adenosine and 2-chloroadenosine. These results indicate that 2-substituted adenosine derivatives, like adenosine, produce inhibition of transmission by acting on a presynaptic site which can be blocked by theophylline. The high apparent potency of 2-chloroadenosine compared to adenosine appears to be duc to the former being neither taken up nor deaminated, while the apparent potency of adenosine is masked by uptake and deamination in this tissue.

    Notizen: Abstract A method for isolation and purification of intact nuclei from barley leaves was developed and several properties of the chromatin were studied. The dense structure of the main part of the chromatin does not alter the accessibility of the DNA to nucleases. 60% of the nuclear DNA can be degraded by micrococcal endonuclease. Nevertheless the solubility of the chromatin fragments depends on the extent of nuclease digestion; solubilisation occurring only when the major part of the internucleosomal DNA was degraded (≃30% of digestion). Electron microscopic observations suggest that this was due to particularly dense organization of the chromatin “in situ”. The possible physiological meaning of some of these properties are discussed.

    Notizen: Abstract Ab initio calculations have been carried out on CO and N2 and the relevant core hole states with different basis sets to investigate differences in geometries and force constants. From these calculations vibrational band profiles of the core level ESCA spectra for these molecules have been interpreted, obviating the need to rely on data pertaining to the equivalent core species. The agreement with experimental profiles is excellent. The O1s level of CO which has not been subjected to detailed theoretical analysis previously, is predicted to show substantial vibrational structure in excellent agreement with recently acquired experimental data. The effect of temperature on the band profiles has also been considered. Theoretically derived core binding and relaxation energies of these systems have been investigated both as a function of basis set, and of internuclear distance. Density difference contours have been computed and give a straightforward pictorial representation of the substantial electron reorganizations accompanying core ionizations. Small basis sets with valence exponents appropriate to the equivalent core species when used in hole state calculations describe bond lengths, force constants, core binding energies and relaxation energies with an accuracy comparable to that appropriate to the corresponding extended basis set calculations.

    Notizen: Abstract Calculations have been carried out on an extensive series of molecules for both the neutral species and core ionized states. Substituent effects on C1s , N1s , O1s , and F1s levels have been investigated and where available comparison has been drawn with experiment. Comparison with Koopmans' theorem has allowed a relatively detailed study of change in relaxation energies as a function of substituent effect on a given core level. Whilst for C1s levels the computed shifts in core binding energies are approximately linearly related to differences in relaxation energies for the N1s , O1s , and F1s levels, the relative electronegativity of the substituent can invert this correlation. The empirical correction of Koopmans' theorem for differences in relaxation energies at different sites has been investigated for large molecules. The results compare well with direct hole state calculations.

    Notizen: During the process of chromatin cndensation in the spermiogenesis of the neogastropod mollusc Murex brandaris, the nuclear protein complement undergoes a complex series of changes. These changes lead to the appearance of three small protamines in the ripe sperm nuclei. We have characterized this system electrophoretically and at the compositions with antibodies elicited against a specific spermatozoan protamine. Our results indicate that the complex pattern of chromatin condensation during spermiogenesis in this species (M. brandaris) may be modulated by a series of post-translational (and intranuclear) modifications of DNA-interacting proteins, such as precursors to the sperm protamines. The amino acid composition of each sperm protamine is remarkably simple (lys + arg + gly ≥96 mol%). This system of spermiogenic/spermatozoal proteins in the neogastropod M. brandaris clearly differs from that in patellogastropods and archaeogastropods, and it may be helpful in understanding evolutionary changes in the chromatin condensation pattern during the spermiogenesis of gastropod molluscs. © 1994 Wiley-Liss, Inc.
