    Notes: Summary Although the absorption of verapamil is almost complete after oral administration, its bioavailability is low due to extensive hepatic first-pass metabolism. Besides large interindividual differences in first-pass metabolism, pronounced day-to-day intraindividual variations in first-pass metabolism are observed, leading to erroneous results in relative bioavailability studies. Stable isotope techniques, which permit simultaneous administration of a solution and a tablet, can successfully be used to overcome these difficulties. The method has the advantage that two experiments can be carried out in a single test. Furthermore, the number of subjects required in bioavailability studies can be greatly reduced. Using this technique the bioavailability of verapamil tablets (Isoptin® 80) relative to a stable labelled solution of verapamil was found to be 108.1%, with a 95% confidence interval between 89.1 and 127.1%.

    Notes: Summary The pharmacokinetics of verapamil in five healthy volunteers were investigated on 4 occasions during chronic administration of deuterated verapamil. There was no statistically significant difference in oral clearance, terminal half-life, bioavailability, morning trough level and peak concentration or in the time of their occurrence on the four occasions. The plasma clearance, however, exhibited considerable inter- and intra-individual variation, ranging between 26.3% and 85.4% and 12.0% and 48.0%, respectively. Comparison of these pharmacokinetic parameters with data from previous single dose studies in the same subjects revealed a significant (p〈0.05) decrease in the clearance and an increase in the apparent bioavailability of verapamil during chronic administration, although no difference in the half-life was found. Due to the considerable variation in the oral clearance of verapamil during chronic dosing, steady-state conditions in a strict pharmacokinetic sense may never be attained, and pharmacokinetic data obtained in single dose studies will be of limited value in predicting steady-state plasma concentrations.

    Notes: The gas-phase, ion molecule reaction between C3O2+. and C3O2 has been studied by both double-focusing and ion trap mass spectrometry, rationalized by the formation of a dimeric, odd electron cation [C6O4]+. which decomposes extensively through sequential CO losses giving rise to [C5O3]+. and [C4O2]+. ions. The thermodynamics of this process have been investigated by means of ab initio calculation performed on the above species using different basis sets (STO-3G, 3-21G and 6-31G*).

    Notes: We report here the spectra that result when singly and doubly charged buckminsterfullerene ions, C60+· and C602+, collide with self-assembled monolayer films at collison energies ranging from 125 to 320 eV. The surfaces were prepared by the spontaneous assembly of octadecanethiol (CH3(CH2)17SH), perdeuteroeicosanethiol (CD3(CD2)19SH), and 2-(perfluorooctyl)-ethanethiol (CF3(CF2)7CH17CH2SH) on vapor-deposited gold. The fluorinated surface causes the greatest conversion of kinetic energy into internal energy, more extensive fragmentation of doubly charged buckminsterfullerene C602+ is detected in collisions with this surface than in collisions with the alkanethiolate surfaces.

    Notes: Three different methods were used for the determination of the effective incident angle related to EDXRF when radioisotope annular sources are involved: weighted averaging, Compton peak method and minimization of the difference between the certificated and measured concetrations of six international standard samples. By measuring the Compton peak energies of various analytical reagent grade elements and compounds, it was found that the incoherent peak energy depends on the mean atomic number of the matrix, so the effective incident angle is also matrix dependent.

    Notes: Two widely used concentration calculation techniques, emission-transmission and elemental sensitivity methods, were compared. In the determination of the trace element content of environmental materials (e. g. plants, soil), the mass absorption coefficients for the exciting and characteristic x-rays are dominated of the ‘non-detectable’, light major components. When using the elemental sensitivity method, these mass absorption coefficients must be a known a priori. When using the emission-transmission method, the combined attenuation of the incident and fluorescence x-rays within the specimen for the ith element is determined experimentally. The trace element content of biological materials was determined with both methods and the concentrations obtained were compared by t-test and one-way analysis of variance methods. In the case of available reference materials the concentration values obtained with these methods were compared with the certified values.
