    Notes: Abstract Synthesis of growth-limiting proteins (GLP) is required for continued auxin-induced elongation of oat (Avena sativa L.) coleoptiles. In order to determine whether GLP synthesis is dependent or independent of auxin, a double-labeling ratio technique, coupled with disc-gel electrophoresis, has been used to assess the effect of auxin on the pattern of protein synthesis. Sections were peeled to enhance amino-acid uptake; proteins were labeled with [14C]- or [3H] leucine in the presence or absence of indole-3-acetic acid for 40 min to 6 h, and were separated into soluble, membrane-associated, and wall-associated fractions. Regardless of the conditions used, or the protein fraction examined, no changes in response to auxin were detected in the pattern of protein synthesis. In order to escape detection by this technique an auxin-induced protein would have to comprise less than 0.75% of the total newly synthesized protein. Thus the synthesis of GLP appears to be independent of auxin. The same technique has been used to follow protein turnover. During the chase, proteins are initially degraded at an average rate of 8% h−1, and some protein bands showed as much as 14% h−1 degradation. No protein was detected which had a turnover rate as rapid as the GLP.

    Notes: Abstract The cytoplasm of subepidermal parenchyma cells of Avena sativa L. coleoptiles was collected at one end of the cell by centrifugation. The electrical properties of both plasmalemma and tonoplast were then examined with microelectrodes inserted into both cytoplasm and vacuole of the same cell. The input resistance of the cytoplasm measured with either electrode was 7.5±0.8 MΩ while that of the vacuole measured with the single vacuolar electrode and a bridge circuit was 29.2±3.1 MΩ. The latter value was not significantly different from that of control, uncentrifuged cells. The resistance of the tonoplast is therefore several times larger than the input resistance of the cytoplasm, but the specific resistance of the plasma membrane cannot be calculated without knowledge of the extent and pattern of intercellular coupling. Electrical coupling of the cytoplasms of adjacent cells was observed in only two out of eight experiments. The mean potential of the vacuoles,-77.8±6.4 mV, was not significantly different from that of the cytoplasm; however, all the available evidence indicates that variable tip potentials in impaled cells made absolute determination of the membrane potential uncertain. In fusicoccin, the cells hyperpolarized by 20 mV within 10 min. This reponse occurred entirely at the plasmalemma.

    Notes: Abstract We have compared the effects of cycloheximide (CHI) and two other rapid and effective inhibitors of protein synthesis, pactamycin and 2-(4-methyl-2,6-dinitroanilino)-N-methyl proprionamide (MDMP), on protein synthesis, respiration, auxin-induced growth and H+-excreation of Avena sativa L. coleoptiles. All three compounds inhibit protein synthesis without affecting respiration. The effectiveness of the inhibitors against H+-excretion and growth correlates with their ability to inhibit protein synthesis. Both CHI and MDMP inhibit auxin-induced H+-excretion after a latent period of 5–8 min, and inhibit growth after a 8–10-min lag. These results support the idea that continued protein synthesis is required in the initial stages of the growth-promoting action of auxin.

    Notes: Abstract The phytotoxin fusicoccin (FC) causes rapid synthesis of malate in coleoptile tissues, presumably via phosphoenolpyruvate (PEP) carboxylase coupled with malate dehydrogenase. The possibility that FC directly affects PEP carboxylase in Avena sativa L. and Zea mays L. coleoptiles was studied and rejected. The activity of this enzyme is unaffected by FC whether FC is added in vitro or a pretreatment to the live material. FC does not change the sensitivity of the enzyme to bicarbonate or malate. The activity of FC, instead, appears to be indirect. The pH sensitivity of PEP carboxylase is such that its activity, and thus the rate of malate synthesis, may be enhanced by an increase in cytoplasmic pH accompanying FC-induced H+ excretion. Since the enzyme is also particularily sensitive to bicarbonate levels, malate synthesis may also be enhanced by FC-induced uptake or generation of CO2.

    Notes: Abstract Variability exists among worker honey bees for components of division of labor. These components are of two types, those that affect foraging behavior and those that affect life-history characteristics of workers. Variable foraging behavior components are: the probability that foraging workers collect (1) pollen only; (2) nectar only; and (3) pollen and nectar on the same trip. Life history components are: (1) the age the workers initiate foraging behavior; (2) the length of the foraging life of a worker; and (3) worker length of life. We show how these components may interact to change the social organization of honey bee colonies and the lifetime foraging productivity of individual workers. Selection acting on foraging behavior components may result in changes in the proportion of workers collecting pollen and nectar. Selection acting on life-history components may affect the size of the foraging population and the distribution of workers between within nest and foraging activities. We suggest that these components define possible sociogenic “pathways” through which colony-level natural selection can change social organization. These pathways may be analogous to developmental pathways in the morphogenesis of individual organisms because small changes in behavioral or life history components of individual workers may lead to major changes in the organizational structure of colonies.

    Notes: Abstract Previously we reported that there are subfamily differences in drone production in queenless honey bee colonies, but these biases are not always explained by subfamily differences in oviposition behavior. Here we determine whether these puzzling results are best explained by either inadequate sampling of the laying worker population or reproductive conflict among workers resulting in differential treatment of eggs and larvae. Using colonies composed of workers from electrophoretically distinct subfamilies, we collected samples of adult bees engaged in the following behavior: “true” egg laying, “false” egg laying, indeterminate egg laying, egg cannibalism, or nursing (contact with larvae). We also collected samples of drone brood at four different ages: 0 to 2.5-h-old eggs, 0 to 24-h-old eggs, 3 to 8-day-old larvae, and 9 to 14-day-old larvae and pupae. Allozyme analyses revealed significant subfamily differences in the likelihood of exhibiting egg laying, egg cannibalism, and nursing behavior, as well as significant subfamily differences in drone production. There were no subfamily differences among the different types of laying workers collected from each colony, suggesting that discrepancies between subfamily biases in egg-laying behavior and drone production are not due to inadequate sampling of the laying worker population. Subfamily biases in drone brood production within a colony changed significantly with brood age. Laying workers had significantly more developed ovaries than either egg cannibals or nurses, establishing a physiological correlate for the observed behavioral genetic differences. These results suggest there is reproductive conflict among subfamilies and individuals within queenless colonies of honey bees. The implications of these results for the evolution of reproductive conflict, in both queenright and queenless contexts, are discussed.

    Notes: Summary Three experiments were performed to determine whether brood care in honey bee colonies is influenced by colony genetic structure and by social context. In experiment 1, there were significant genotypic biases in the relative likelihood of rearing queens or workers, based on observations of individually labeled workers of known age belonging to two visually distinguishable subfamilies. In experiment 2, no genotypic biases in the relative likelihood of rearing drones or workers was detected, in the same colonies that were used in experiment 1. In experiment 3, there again were significant genotypic differences in the likelihood of rearing queens or workers, based on electrophoretic analyses of workers from a set of colonies with allozyme subfamily markers. There also was an overall significant trend for colonies to show greater subfamily differences in queen rearing when the queens were sisters (half- and super-sisters) rather than unrelated, but these differences were not consistent from trial to trial for some colonies. Results of experiments 1 and 3 demonstrate genotypic differences in queen rearing, which has been reported previously based on more limited behavioral observations. Results from all three experiments suggest that genotypic differences in brood care are influenced by social context and may be more pronounced when workers have a theoretical opportunity to practice nepotism. Finally, we failed to detect persistent interindividual differences in bees from either subfamily in the tendency to rear queen brood, using two different statistical tests. This indicates that the probability of queen rearing was influenced by genotypic differences but not by the effect of prior queen-rearing experience. These results suggest that subfamilies within a colony can specialize on a particular task, such as queen rearing, without individual workers performing that task for extended periods of time.

    Notes: Concanavalin A (Con A) binding and Con A-mediated hemadsorption to LM cells were found to decrease significantly at both 5-7°C and 15-19°C. The higher of these critical temperatures responds to a change in state of the membrane lipids and can be increased or decreased in cells where the membrane phospholipids contain less or more double bonds, respectively. The lower critical temperature for Con A binding or Con A-mediated hemadsorption does not respond to these changes in membrane lipid composition. Though the amount of Con A bound to the cell surface is a determinant of Con A-mediated agglutinability, the major components of the decreases in Con A-mediated hemadsorption which occur at both these critical temperatures do not have their origin in the decreases in Con A binding which occur over these same temperature ranges - that is 5-7°C and 15-19°C.Con A-mediated hemadsorption measured at 22°C was dramatically inhibited when LM cells were first incubated at 7°C or less. Reversal of this inhibition required 20-30 min of subsequent incubation at 22°C, indicating that factors other than membrane lipid “fluidity” are determinants of agglutinability. LM cells treated with the microtubule-disrupting alkaloids colchicine, colcemid, or vinblastine at concentrations as low as 10-6 M were as much as fourfold more agglutinable with Con A. By contrast, lumicolchicine, an inactive derivative of colchicine, had a slight inhibitory effect on Con A-mediated hemadsorption. Colchicine, vinblastine, or lumicolchicine treatment of LM cells did not alter the quantitative binding of labeled lectin. The results suggest that membrane lipid “fluidity” and the cell cytoskeleton (microtubule/microfilament system) are important determinants of lectin interactions with cell surfaces. The results are interpreted in terms of a model of cell-cell and cell-lectin interactions which assigns a central role to the Con A receptor.
