    Notes: Summary Formation of the 2-methoxymethyl-pyrrolidine-1-dithiocarboxylates2–4 and alkylation of2 and3 were studied. Enantiomeric and diastereomeric derivatives of4, the preparation of diastereomeric mixtures of4 by alkylation of3 in the presence of strong bases, and formation of6 by phase transfer alkylation of2 are described. The two enantiomers of 2-(4-bromophenyl)-2-oxo-ethyl 2-methoxymethylpyrrolidine-1-dithiocarboxylate2 have been characterized by X-ray analysis.

    Notes: This article treats the basic problem of selection of experimental conditions for microbiological experiments for evaluation of newly isolated bacterial strains, mutants, or plasmid/strain combinations. For this purpose shake flask experiments in a 210-4confounded factorial design at resolution IV with four blocks of 16 flasks were used. The design was used for testing of two new strain/plasmid combinations (E. coli MT 102/403-SD2 and W 3110/403-SD2) i.e., both strains with the same plasmid 403-SD2. Both strains were integrated in the design, so both strains were tested with nine factors (temperature, aeration, glucose, initial pH, pH regulation, reduced aeration, chloramphenicol, acetate, and glycerol). With both strains the interaction between initial pH and reduced aeration had a significant influence on the yield of the recombinant-DNA product nuclease. There was more than a factor of 10 between lowest and highest yield of product. In this interactive system the strains reacted differently. MT 102/403-SD2 had highest yields at high initial pH (8.4) and no reduction in aeration, whereas W 3110/403-SD2 had highest yields of nuclease at low initial pH (7.4) and reduced aeration (rubber stopper inserted after cultivation for 12 h). These data (and previous work) clearly demonstrate that it is impossible to suggest a simple set of experimental conditions for testing of new plasmid/strain combinations. It is clear that the exclusive application of a standardized growth technique e.g., LB-medium at 37°C at an unspecified and uncontrolled aeration level, may lead to wrong conclusions on properties and potentials of now plasmid/strain combinations and may lead to rejection of useful strains or plasmids.

    Notes: A new synthesis of pyrido[3,2-f][1,4]thiazepine derivatives 3 starting with 2-chloronicotinic acid (1), methylaminoacetonitrile hydrochloride and carbon disulfide is described. As proved by a crystal structure determination, a boat conformation with approximated mirror symmetry can be assigned to the 1,4-thiazepine ring in 3b. 2-Chloro-N-cyanomethyl-N-methyl-5-nitrobenzamide (5) reacts with carbon disulfide in presence of a strong base in DMF or DMSO depending on the temperature to either the benzothiopyran compound 6 or by intramolecular aromatic nucleophilic substitution to a seven-membered ring system as thiolate anion which can be alkylated to give the 1,4-benzothiazepine derivative 7, or to an open-chain amido ketene dithioacetal 8.

    Notes: A simple and time-saving electrophoretic method for the determination of the 6 common phenotypes of red cell esterase D (ESD) is presented. Using a continuous buffer system composed of malic acid, the separation of the allozymes is accomplished at pH-values between 5.35-5.45 by thin-layer agarose gel electrophoresis in 4 h. Until now, a combination of electrophoresis and isoelectric focusing was required to obtain the same results. The economical advantage as well as the modest requirements concerning laboratory equipment are evident.
