    Notes: Integument from blue and green areas of the variant blue frog were analyzed biochemically for pteridines and carotenoids. Solvent extraction and absorption spectrophotometry indicated that β carotene was greatly reduced in the blue skin, and present in high quantities in the green skin of the blue frog. Thin layer and paper chromatography indicated that the pteridines were almost totally lacking in the blue skin, and present in normal quantities in the green skin of the blue frog.Light and electron microscopy indicated that the xanthophore pigment cells were either greatly altered or absent from the blue integument and present in the green integument. The fine structure of the xanthphores of the green integument contained the normal ultrastructural components of xanthopores found in regular green integument. The blue integument contained an abnormal cell type that occupied the position in the dermal chromatophore unit normally held by the xanthophores. The possibility of these cells being abnormal xanthophores or some other cell type is discussed.

    Notes: This manuscript describes in precise detail the ultrastructural alterations produced as a result of laser microirradiation of nucleoli and nucleoplasm of tissue culture cells. Because of the general difficulty of single cell recovery, flat embedding, and serial sectioning, very few studies have ever been conducted on microbeam irradiated cells; yet the use of the microbeam technique has become widespread in functional studies of the nucleus. The results presented here demonstrate two classes of lesion material: small spherical electron dense bodies 0.05-0.02 μm in diameter and a larger, more irregular electron dense material up to 1 μm in length. The occurrence of these different types of lesion materials is described in control irradiated nucleoli and nucleoplasm, irradiated nucleoli and nucleoplasm in quinacrine treated cells, irradiated nucleoli and nucleoplasm in actinomycin D treated cells, and irradiated nucleoli and nucleoplasm in combined actinomycin D and quinacrine treated cells. In all the cells in which actinomycin D was employed, nucleoli were selectively irradiated in either their granular or fibrillar zones. The results of the ultrastructural studies are discussed in light of earlier functional studies.

    Notes: Ethidium bromide (l0 μg/ml) and bromodeoxyuridine (25 μg/ml) were used to sensitize selective cell organelles to visible wavelengths of an argon ion Her (488 and 514 nanometers). Ethidium bromide was shown to be salabtlve In sensitizing nucleoli, chromosomes, and the centriolar region of PTK2 cells to the laser microbeam. Similarly, BrDU sensitized chromosomes to the microbeam irradiation. The lesions produced on the chromosomes when either agent was used appeared as a phase paling of the irradiated segment. Nucleolar lesions also appeared as a phase paling, and the centriolar region alteration appeared either as a phase paling or a phase darkening.

    Notes: In the spiroboloid millipeds the male gonopods develop from rudiments of both pairs of legs of the seventh body segment. They are first evident as “lumps” on the sternum of the seventh segment following the molt to instar 5; they replace the functional walking legs of the seventh segment of earlier instars. These “lumps” go through progressive morphological changes in each instar until they attain the adult form. It is evident that in the spirobolid millipeds the gonopods do not develop as a gradual modification of functional walking legs; rather they pass through a progressive growth and differentiation of their own.

    Notes: Three-dimensional, histological, and x-ray techniques provide a picture of body segment and limb morphogenesis. Cell multiplication begins in the proliferation region (“meristem”) during the molt from the preceding instar. By four days post-molt, the cells that will form the new segments are well under way in their anterior, lateral, and dorsal migration. It is suggested that after the anlagen for all the new segments are estabilshed, a mitotic wave commences in the most anterior anlagen and moves posteriorly during the remainder of the instar. When cell proliferation is complete, final differentiation of the segments takes place.The process of limb formation is one of cell proliferation and perhaps migration. Each limb develops in a membranous pocket during the instar following the one in which its respective body segment was formed.

    Notes: Direct development in amphibians is an evolutionarily derived life-history mode that involves the loss of the free-living, aquatic larval stage. We examined embryos of the direct-developing anuran Eleutherodactylus coqui (Leptodactylidae) to evaluate how the biphasic pattern of cranial ontogeny of metamorphosing species has been modified in the evolution of direct development in this lineage. We employed whole-mount immunohistochemistry using a monoclonal antibody against the extracellular matrix component Type II collagen, which allows visualization of the morphology of cartilages earlier and more effectively than traditional histological procedures; these latter procedures were also used where appropriate. This represents the first time that initial chondrogenic stages of cranial development of any vertebrate have been depicted in whole-mounts.Many cranial cartilages typical of larval anurans, e.g., suprarostrals, cornua trabeculae, never form in Eleutherodactylus coqui. Consequently, many regions of the skull assume an adult, or postmetamorphic, morphology from the inception of their development. Other components, e.g., the lower jaw, jaw suspensorium, and the hyobranchial skeleton, initially assume a mid-metamorphic configuration, which is subsequently remodeled before hatching. Thirteen of the adult complement of 17 bones form in the embryo, beginning with two bones of the jaw and jaw suspensorium, the angulosplenial and squamosal. Precocious ossification of these and other jaw elements is an evolutionarily derived feature not found in metamorphosing anurans, but shared with some direct-developing caecilians. Thus, in Eleutherodactylus cranial development involves both recapitulation and repatterning of the ancestral metamorphic ontogeny. These modifications, however, are not associated with any fundamental change in adult morphology and cannot at this time be causally linked to the evolutionary success of this extraordinarily speciose genus.

    Notes: Platelet-derived growth factor (PDGF) and angiotensin II (All) are thought to mediate their biological effects in vascular smooth muscle cells (VSMCs) by causing alterations in cytosolic free calcium ([Ca2+]i). In this study we examine the pathways by which PDGF and All alter [Ca2 +]i in VSMCs. Addition of PDGF resulted in a rapid, transient, concentration-dependent increase in [Ca2+]i; this rise in [Ca2 +]i was blocked completely by preincubation of cells with ethylene glycol-bis (β-aminoethyl ether) N,N,N′,N′-tetraacetic acid (EGTA) or CoCl2, by the voltage-sensitive Ca2 +-channel antagonists verapamil or nifedipine, by 12-O-tetradecanoylphorbol-13-acetate (TPA), or by pertussis toxin. All also caused an increase in [Ca2 +]i; however, All-stimulated alterations in [Ca2+]i displayed different kinetics compared with those caused by PDGF. Pretreatment of cells with 8-(diethylamine)-octyl-3,4,5-trimethyoxybenzoate hydrochloride (TMB-8), almost totally inhibited All-induced increases in [Ca2+]i. EGTA or CoCl2 only slightly diminished All-stimulated increases in [Ca2+]i. Nifedipine, verapamil, TPA, and pertussis toxin pretreatment were without effect on All-induced increases in [Ca2+]i. PDGF and All both stimulated increases in total inositol phosphate accumulation, although the one-half maximal concentration (ED50) for alterations in [Ca2+]i and phosphoinisitide hydrolysis differed by a factor of 10 for PDGF (3 × 10-10 M for Ca2+ vs. 2.5 × 10-9 M for phosphoinositide hydrolysis), but they were essentially identical for All (7.5 × 10-9 M for Ca2+ vs. 5.0 × 10-9 M for phosphoinositide hydrolysis). PDGF stimulated mitogenesis (as measured by [3H]-thymidine incorporation into DNA) in VSMCs with an ED50 similar to that for PDGF-induced alterations in phosphoinositide hydrolysis. PDGF-stimulated mitogenesis was blocked by pretreatment of cells with voltage-sensitive Ca2 + channel blockers, TPA, or pertussis toxin. These results suggest that PDGF and All cause alterations in [Ca2+]i in VSMCs by at least quantitatively distinct mechanisms. PDGF binding activates a pertussis-toxin-sensitive Ca2+ influx into cells via voltage-sensitive Ca2+ channels (blocked by EGTA, verapamil, and nifedipine), as well as stimulating phosphoinositide hydrolysis leading to release of Ca2+ from intracellular stores. All-induced alterations in [Ca2+]i are mainly the result of phosphoinositide hydrolysis and consequent entry of Ca2+ into the cytoplasm from intracellular stores. Our data also suggest that changes in [Ca2 +]i caused by PDGF are required for PDGF-stimulated mitogenesis.

    Notes: P19 embryonal carcinoma (EC) cells can be induced in vitro to differentiate into cells resembling those normally formed in the embryo. Among these cell types is one whose morphology is fibroblast-like. Using indirect immunofluorescence and Western blot analysis with antibodies directed against various isoforms of actin, many of these fibroblast-like cells were found to express smooth muscle actin isoforms. Northern blot analysis of RNA indicated the presence of a smooth muscle-specific isoform of myosin heavy-chain mRNA in immortal lines of these fibroblast-like cells. These results suggest that these fibroblast-like cells resemble fetal myofibroblastic or myoepithelial cells, which have a wide distribution during embryonic development.

    Notes: A method is described whereby serial sections are photographed through a microscope and printed on a clear acetate film. The pictures are mounted on Plexiglas rectangles and placed back to back. A solid stack of plates with the reconstructed structure visible inside, or a slide-box arrangement of every other section may be produced. Variations of the technique are presented so that one can print high-contrast or low-contrast pictures, reconstruct simple or complex structures, and work more rapidly with less equipment. This is a simple, accurate, and rapid method.
