    Notes: Abstract: In recent reports, ciliary neurotrophic factor (CNTF) has been implicated as an injury factor involved in regulating astrogliosis in the CNS. In this study, we used a rat oligodendroglial progenitor cell line that is highly responsive to CNTF to examine CNTF-induced alterations that may play a role in activation of the glial fibrillary acidic protein (GFAP) gene. We determined that CNTF induces the transient translocation of Stat1α/p91 to the nucleus. This nuclear translocation was followed by GFAP promoter activation and an up-regulation of GFAP mRNA and protein. Levels of CNTF-α receptor mRNA, however, were unaffected by addition of the ligand. Transfection studies using an upstream 5′-flanking, 1.9-kb rat GFAP promoter linked to a luciferase reporter gene revealed CNTF-induced transcriptional activation within 1 h of ligand exposure. Moreover, serial-deleted constructs identified a distal (−1,857 to −1,546 bp) and a proximal (−384 to −106 bp) region as being important for CNTF-induced GFAP promoter activation. These two regions showed a strong degree of overlap for CNTF- and serum-induced activation of the GFAP gene. Analysis of the two regions revealed several cis-elements that are thought to be involved in GFAP regulation and/or the regulation of other genes by members of the interleukin-6 family of cytokines. Moreover, we are the first to report the presence of several putative CNTF-responsive elements within our identified distal and proximal regions in the GFAP gene promoter.

    Notes: Positive temperature coefficient resistors are evaluated in the self-heating mode as pulse generators and as opening switches in inductive storage pulse power supplies. Results from both carbon-filled polymer devices, as well as from barium-titanate-based devices, are shown. Theoretical switched powers of close to a 1/4 MW are calculated for honeycomb-shaped BaTiO3 positive temperature coefficient resistors. Switching times in the millisecond range are achieved. Pulse duration varies with ambient temperature, but this can be counteracted by using capacitative triggering. Surface flashover was one of the critical failure mechanisms. The results point towards further improved device designs.

    Notes: Fully strained layers of GaInP on InP significantly thicker than the theoretical critical thickness were grown by metalorganic molecular beam epitaxy. The excess strain energy of these layers is most probably accommodated by long line defects observed by atomic force microscopy. The thickness at which the long line defects appear is about four times the Matthews and Blakeslee critical thickness [J. W. Matthews and A. E. Blakeslee, J. Cryst. Growth 27, 118 (1974)]. Partial relaxation is measured by x-ray at about 14 times the theoretical critical thickness. © 2001 American Institute of Physics.

    Notes: Flow cytometric analysis (FCA) and silver colloidal nucleolar organizer region-associated protein staining (AgNOR) have been used individually in assessing the histopathologic nature of various human tumors. However, few researchers have investigated the relationship between the two techniques in a single series. In a retrospective study, we examined 36 premalignant lesions of the oral cavity by FCA and AgNOR on formalin-fixed, paraffin-embedded tissue submitted to the University of Tennessee, Memphis, oral pathology laboratory. Three categories of epithelial dysplasia were represented (9 mild. 9 moderate, 6 severe), as well as four epithelial hyperplasias without dysplasia, three squamous cell carcinomas, and five fibrous nodules as controls. Parameters recorded for each case included age. race, gender, site, light microscopic diagnosis (LMD). DNA index (DI), total proliferative index (TPI), S-phase (S), range of nucleolar organizer regions (RNOR), and mean number of nucleolar organizer regions (MNOR). The average maximum nucleolar organizer region count (AMXNOR) for each LMD category was also calculated. The objective of the study was to determine if FCA or AgNOR aided in the subjective LMD of oral premalignant lesions and if the parameters recorded for the specimens exhibited any positive correlation. The FCA results indicated an abnormal DI in 6 of the 24 dysplastic lesions. A positive partial correlation was seen between DI and MNOR (r = 0.434: P〈0.012) and TPI and S (r = 0.774; P 〈 0.0001), holding gender and race constant. Additionally, the AMXNOR exhibited a slight tendency to increase for each increasing grade of dysplasia but this could not be confirmed statistically. The results indicate that there is a positive correlation between FCA and AgNOR but that neither FCA nor AgNOR were a diagnostic aid in grading oral mucosal dysplasia.

    Notes: Nodular fasciitis is a benign reactive fibroblastic proliferation which usually arises from the superficial or deep fascia of the extremities and trunk. In 1981 a distinct variant, intravascular fasciitis (IVF), was reported which possessed the same clinical and histological features of more ordinary forms of nodular fasciitis but, in addition, had a striking involvement of associated arteries and/or veins. A case of intraoral IVF is presented and its clinical and histologic features are compared with those of IVF found elsewhere in the body. A brief discussion of etiology and differential diagnostic considerations is also included.

    Notes: Summary To investigate the effects of long-term all-trans-retinoic acid (RA) treatment on epidermal differentiation in vivo, rhino mice were treated topically with 0.005% RA, and their epidermis was analysed histologically and biochemically after 5, 13 and 26 weeks of treatment. Effects of RA were observed first in the living layers of the epidermis, and then in the non-viable stratum corneum. Five weeks of topical RA led to thickening of the spinous and granular compartments, induction of keratins K6, K16 and K17, and suppression of filaggrin expression. After 13 and 26 weeks of RA treatment, the number of anucleate cornified cell layers remained similar to controls, but additional changes in histology and protein expression were observed. The results showed that prolonged administration of topical RA induced epidermal hyperproliferation, but did not suppress differentiation, in contrast to results observed in keratinocyte cultures. However, the distinct histological and biochemical changes observed in the spinous, granular and cornified layers of RA-treated skin after 26 weeks of treatment, suggested that the progeny of RA-treated basal cells undergo a modified programme of terminal differentiation. Considering the present data together with results of previous in vivo studies, we propose that long-term topical RA treatment retards, or specifically modulates, the later stages in epidermal differentiation, or programmed cell death.
