Notes: We consider that the microvascular bed may play a role in the initiation and maintenance of growth from primordial to primary follicles. Therefore, using immunochemistry, we examined microvessels in calf and cow ovaries to identify the presence of factor VIII-related antigen endothelial cells. A vessel-poor zone was observed in the cortex of immature and mature cow ovaries. Primordial and primary follicles were assembled in this zone. It is concluded that follicular dormancy is likely to be maintained by the scarcity of microvessels and thus by the consequent poverty of the blood supply.

Notes: Summary Biologic and non-biologic materials clinically used as hemostyptica, as vascular prostheses, or as temporary skin substitutes were implanted to the chick chorioallantoic membrane (CAM) during days 9–14 of incubation. The histological study revealed an intact CAM after application of cellulose gauze (Tabotamp). The fibrin tissue adhesive (Tissucol), the collagen sponge (Tachotop), or the gelatin sponge (Gelfoam) induced different amounts of connective tissue. Some fibroblasts were about to grow into the fibrin adhesive. Fibroblasts and capillaries filled the interstices of the collagen sponge poorly, and in the case of the gelatin sponge markedly. Neutrophils and giant cells of the foreign body type occurred more often in the gelatin sponge than in the collagen sponge. With regard to the non-biologic materials, expanded polytetrafluorethylene induced squamous metaplasia of the chorionic epithelium. Dacron caused ulceration with the onset of connective tissue ingrowth, based on a reaction of giant cells of the foreign body type. Polyurethane foam (SYSpur-derm) showed bleeding into the implant due to spikes of the material. Eosinophilic granulocytes were absent in all materials studied. The results are related to different clinical findings. The correlation allows consideration of the CAM as an in vivo model in screening materials for their biocompatibility and their connective tissue reaction.

Notes: Summary A new polyester resin of low viscosity was used to study the three-dimensional structure of capillary sprouts of mature follicles and of corpora lutea. Vascular microcorrosion casts were obtained from ovaries of immature golden hamsters between days 4 and 7 after stimulation with pregnant mares' serum gonadotrophin (PMSG). On days 4 and 5 after PMSG, a nodular appearance of the organ was due to mature follicles and corpora lutea. Capilary sprouts were seen as dead-ending structures originating from the thecal capillary network. The sprouts were either concentrated around (follicle type 1) or towards (follicle type 2) the center of the antrum. Increased vascular permeability resulted in resin leakage only in type 2. Intact corpora lutea were characterized by radially grown capillary sprouts. Regressing corpora lutea showed a marked reduction of the vascular bed as displayed by filling defects in casts on day 6 and 7 after PMSG. It is assumed that: 1. Dead-ending capillary structures, resin leakage, and filling defects are not always artifacts of the microcorrosion cast technique, but can express functional microcirculatory changes. 2. The concentric growth of thecal capillary sprouts may be related to the presence of an intact follicular basal membrane, and the radial growth of sprouts be taken as a sign of a digested basal membrane.

Notes: Abstract Five cell types recently isolated from the bovine corpus luteum differed in their epithelioid morphology and their cytoskeleton, but shared common criteria of microvascular endothelial cells1,2. To give strong evidence for the separate entity, the growth rate of the 5 phenotypically different cells was studied. They were seeded at low density on day 0. Most of these cells were treated with 200 to 1000 U recombinant bovine interferon-γ (IFN-γ) for 3 days. The untreated remainder served as controls. Cell counts were made for all cultures on days 4, 7, 10 and 13. morphology: 13 d after treatment with IFN-γ senescent cells as well as intact cells occurred in cultures of cell types 1 to 4. Cultures of cell type 5 were apparently unchanged and resembled their untreated counterparts. Desminpositive cells in cultures of cell type 2 developed cell processes. Growth rate: In the absence of IFN-γ, the growth rate was high for cell types 3 and 4, moderate for cell type 1, and low for cell types 2 and 5. The presence of IFN-γ caused anti-proliferative effects. These were higher for cell types 3 and 4 than for cell types 1 and 2. IFN-γ could be cytotoxic on cell type 3. In contrast, the cytokine tended to support the cell growth of cell type 5. These findings substantiate the postulate that endothelial cells exhibiting separate morphology in culture also function differently.

Notes: Abstract We have recently detected a subgroup of cytokeratin (CK)-positive luteal cells in the bovine corpus luteum of the early and mid-luteal phase, but not in that of pregnancy. Since, according to the literature, neurophysin (NP)-positive luteal cells behave comparably, simple immunohistochemistry and double labeling were used to identify in serial sections whether the presence of NP coincided with that of CK. The numbers of CK-positive cells and NP-positive luteal cells were comparable throughout the estrous cycle, decreasing from early to late luteal phase. While few CK-positive cells were found in the former thecal layer during the early luteal phase, many CK-positive cells appeared in the former granulosal layer. NP-positive cells were only detected in the former granulosal layer. During the mid-luteal phase, the CK-positive cells consisted of small and large luteal cells, but only large NP-positive cells were found. Roughly 80% of the large CK-positive cells contained NP, whereas CK was lacking in more than 50% of the NP-positive cells. The corpora lutea of pregnancy contained neither CK-positive nor NP-positive cells. The significance of the simultaneous occurrence of CK and NP remains to be elucidated.

Notes: Summary The purpose of this study was to investigate whether albumin (Alb) can be detected in ovarian rat granulosa cells. Using immunocytochemistry and morphometrics, the percentages of Alb-positive follicles (follicle-index), of Alb-positive granulosa cells (granulosa-index), and of strongly reacting follicles (intensity-index) were evaluated in intact and regressing follicles of different diameter groups during different stages of the estrous cycle. In intact follicles, the follicle- and the granulosa-index increased from small-sized to large-sized follicles. Although the follicle-index did not change in any group during the stages of the estrous cycle, the granulosa-index was higher during proestrus than during the other stages. Intact follicles showed a stronger immunoreactivity than regressing follicles throughout the stages of the estrous cycle. Thus, Alb may be a requirement for the control of follicle growth in fertile rats. This Alb function may be attributable to Alb binding to specific cell-membrane components followed by the intracellular uptake of Alb-bound substances.
