    Notes: Abstract: Specific antisera were raised against the A and B chains of glycoprotein III. lmmunoblotting revealed that in adrenal medulla both chains migrate very closely together in two-dimensional electrophoresis. Both chains with slightly differing molecular sizes are found in several endocrine tissues and in brain, kidney, liver, and serum. The mRNA has an analogous widespread distribution. In primary cultures of chromaffin cells the level of message becomes significantly increased by treatment with hista-mine or 12-O-tetradecanoylphorbol 13-acetate/forskoIin. However, the increase is small when compared with that of secretogranin II. The subcellular localization of glycoprotein III in endocrine organs and in the posterior pituitary was investigated by subcellular fractionation and immuno-electron microscopy. Glycoprotein III was found to be confined to the large densecore vesicles of these organs. For a discussion of the function of glycoprotein III, its localization in these organelles has to be taken into account.

    Notes: The Conditional Source-term Estimation (CSE) method was recently proposed to close the chemical source terms occurring in the spatially filtered transport equations of species and enthalpy for Large Eddy Simulation (LES) of nonpremixed reacting flows [W. K. Bushe and H. Steiner, Phys. Fluids 11, 1896 (1999)]. The model is based on the Conditional Moment Closure hypothesis, which provides fairly accurate predictions for the conditional averages of the chemical reaction rates as functions of the conditionally averaged composition vector and temperature with the mixture fraction being an appropriate conditioning variable. In CSE the conditionally averaged composition vector and temperature are obtained by mapping the corresponding spatially filtered scalar fields resolved by the LES into the conditioning (i.e., mixture fraction) space. After the conditional averages of the chemical reaction rates are approximated in mixture fraction space, these are mapped into the physical space to close the source terms in the LES transport equations for the reactive scalars. The present simulation of a turbulent reacting jet is the first test of this new closure in a self-sustained predictive LES. A two-step reduced chemical kinetic mechanism for methane–air flames was used. The results of the simulation, which are in reasonable agreement with available experimental data, prove the model's predictive capabilities as well as its robustness and feasibility for LES. © 2001 American Institute of Physics.

    Notes: Real gas and heat transfer effects are included in the solution of the blast wave resulting from a point or line explosion. Due to the extremely high temperatures, particularly at the beginning of shock propagation, the assumption of perfect gas is not valid. The equation of state takes into account the high temperature effects of vibration, dissociation, electronic excitation, and ionization, as well as the intermolecular forces at high pressures. The computation of the flow problem is carried out using the method of characteristics which has been modified to the effects of radiative and conductive heat transfer. The results show considerable deviation in the initial stages of the explosion from the perfect gas solution, in particular the temperature profiles and the thermodynamic state immediately behind the front.

    Notes: Summary Sequential biopsies taken from a patient with a solitary giant xanthogranuloma, an exaggerated macronodular (〉5 cm in diameter) variant of juvenile xanthogranuloma, and from a patient with benign cephalic histiocytosis, revealed a characteristic time sequence of histopathological findings. Early stages of the diseases showed a monomorphous infiltrate of mononuclear vacuolated histiocytes positive for KiM1p. HAM56 and factor XIIIa and were characterized by clusters of comma-shaped bodies. This was followed by a polymorphous mixture of various mononuclear and mullinucleate histiocytes additionally labelling with KP1 (CD68) and, in occasional cells, for the adherence of peanut agglutinin. A variety of ultrastructural changes were found, including dense and regularly laminated bodies or lipid droplets. Our findings indicate that both entities are variants of a xanthogranulomatous reaction.

    Notes: This series presents six cases of a rare variant of dermatofibroma, characterized by marked clear cell change. All lesions occurred on the lower extremities of middle-aged adults (four women, two men), mostly with the clinical diagnosis of fibrohistiocytic lesion. Histological examination revealed well circumscribed, faintly stained dermal to subcutaneous lesions which were due to the overwhelming presence of clear cells (〈90%), some with prominent PAS-positive cytoplasmic granulation. Overlying epidermal hyperplasia as well as storiform arrangement of spindle cells, sclerotic collagen and some interspersed lympho-histiocytic infiltrate at the periphery of the lesion indicated the fibrohistiocytic origin. Individual histopathological peculiarities included : bizarre giant cells in two cases, prominent perivascular lympho-histiocytic response, perifollicular arrangement and haemangiopericytoma-like features with iron deposition in one case each. Immunohistochemically three of four lesions showed moderate reactivity for factor XIIIa and two of four with an anti-metallothionen marker E9, but were otherwise negative with a broad panel of markers. Electronmicroscopy in two cases revealed large pools of glycogen beside focal, prominent endoplasmic reticulum and lysosomes in some granular cells, but only optically translucent cells in cases of clear cells. Recognition of clear cell dermatofibroma is important as the differential diagnosis includes some entities with more serious outcome/considerations such as metastases of renal cell carcinoma, xanthogranulomatous reactions, balloon cell naevus/melanoma and clear cell sarcoma.

    Notes: We report on eleven cases of a distinctive dermal spindle cell lipoma characterized by a mixture of mature adipocytes and spindle-shaped cells in a fibromucinous background. Six lesions showed a moderately well circumscribed plexiform pattern, five a well demarcated nodular pattern with compression of surrounding connective tissue and prominent stromal fibrosis. Clinically, the plexiform type mostly occurred in the thigh-groin-buttock area and the nodular type in the head-neck or acral location. While plexiform lesions were predominantly seen in middle-aged females, nodular types occurred in young adults of either sex. No recurrence was seen in five patients with follow-up. The tumour cells were vimentin positive and a thin cytomplasmic rim of S-100 protein positivity was seen in mature adipocytes. Ultrastructural studies revealed lipoblastic differentiation of spindle-shaped cells with lipid droplets and basal lamina formation. Dermal spindle cell lipomas seem to be the dermal counterpart of the most subcutaneously located spindle cell lipoma.

    Notes: In situ hybridization was used to study the mRNA levels for secretogranin II and VGF in comparison with those of oxytocin and vasopressin in the hypothalamus of rats. VGF is a widespread constituent of large dense core vesicles which is selectively induced in PC12 cells by nerve growth factor. After adrenalectomy the mRNA levels of secretogranin II, VGF and vasopressin were increased 4- to 5-fold in the parvocellular neurons of the paraventricular nuclei. In lactating rats the message for oxytocin and secretogranin II were significantly elevated in the magnocellular neurons of the paraventricular and supraoptic nuclei, whereas for VGF only a smaller non-significant increase was observed. As shown by immunoelectron microscopy secretoneurin (a peptide derived from secretogranin II) and oxytocin are co-stored in the large dense core vesicles of the hypothalamo-neurohypophysial neurons.These results demonstrate that stimulation of both parvo- and magnocellular neurons of the hypothalamus induces a concomitant increase of the messages for secretogranin II and VGF together with those of vasopressin and oxytocin.
