Notes: We describe a method for the generation of knowledge-based potentials and apply it to the observed torsional angles of known protein structures. The potential is derived using Bayesian reasoning, and is useful as a prior for further such reasoning in the presence of additional data. The potential takes the form of a probability density function, which is described by a small number of coefficients with the number of necessary coefficients determined by tests based on statistical significance and entropy. We demonstrate the methods in deriving one such potential corresponding to two dimensions, the Ramachandran plot. In contrast to traditional histogram-based methods, the function is continuous and differentiable. These properties allow us to use the function as a force term in the energy minimization of appropriately described structures. The method can easily be extended to other observable angles and higher dimensions, or to include sequence dependence and should find applications in structure determination and validation.

Notes: [Auszug] For the quantitative determination of some amino-acids we have used the method of descending electro-phoresis according to Mikes5 and Bode's evaluation method6. The methods of cultivation of the strain of Penicillium chrysogenum NG on a rotary shaker and the analytical methods used have been ...

Notes: Summary An indirect volumetric determination of beryllium is based on the liberation of acid by reaction with salicylate and its alkalimetric titration. Free acid in Be solutions can also be titrated alkalimetrically after masking of Be with fluoride. For the determination of free acid and beryllium in presence of each other an aliquot is used for the determination of free acid after addition of fluoride and another one for the determination of the sum of Be and free acid after addition of salicylate. Relative errors are ± 2% at the most. Al, Ti, Fe, U and other ions forming strong complexes with salicylate interfere with the beryllium determination. La and Zn can be masked by oxalate. Mg can be present in a 10-fold excess.
