    Notizen: Cell wall synthesis was studied by determining the incorporation of [14C]-glucose into epidermal and cortical cell walls of etiolated Pisum sativum L. cv. Alaska stem segments. Walls were fractionated into the matrix and cellulose components, and incorporation into these components assessed in terms of the total uptake of label into that tissue. When segments were allowed to elongate, the stimulation of total glucose uptake by indole-3-acetic acid (IAA) and fusicoccin (FC) was greater than their stimulation of incorporation. IAA and FC thus did not stimulate precursor incorporation in elongating segments. When elongation was inhibited by calcium, however, IAA and FC significantly promoted wall synthesis in the cortex and vasular tissue (which shows almost no growth or acidification response to auxin). In these tissues incorporation into matrix and cellulose was promoted approximately equally. In the epidermis (thought to be the tissue responsive to auxin in the control of growth), FC promoted a significant increase in wall synthesis, although less than that in the cortex, while there was some evidence of a similar promotion by IAA. Both IAA and FC had a greater effect on incorporation into the matrix component of the wall than into cellulose. The results that FC caused a substantial promotion of cell wall synthesis which was not due solely to elongation, and that the inner non-growth responsive cortical tissues can respond to IAA. Moreover, a comparison of the effects of IAA and FC on the different components of the wall suggests that the response in the epidermis differs from that in the other tissues.

    Notizen: Abstract We describe a new and highly effective optical frequency discriminator and laser stabilization system based on signals reflected from a stable Fabry-Perot reference interferometer. High sensitivity for detection of resonance information is achieved by optical heterodyne detection with sidebands produced by rf phase modulation. Physical, optical, and electronic aspects of this discriminator/laser frequency stabilization system are considered in detail. We show that a high-speed domain exists in which the system responds to the phase (rather than frequency) change of the laser; thus with suitable design the servo loop bandwidth is not limited by the cavity response time. We report diagnostic experiments in which a dye laser and gas laser were independently locked to one stable cavity. Because of the precautions employed, the observed sub-100 Hz beat line width shows that the lasers were this stable. Applications of this system of laser stabilization include precision laser spectroscopy and interferometric gravity-wave detectors.

    Notizen: Abstract The effect of light on the size of intact protoplasts isolated from the primary leaves of etiolated Triticum aestivum was studied. A 2-min red-light irradiation in the presence of 1 mM KCl was sufficient to cause a swelling of protoplasts compared with those maintained in darkness. The effect was photoreversible by far-red light over two light cycles, indicating the involvement of phytochrome. At 4°C, escape from reversibility occurred between 2 and 5 min after the exposure to red light. In exposure-response experiments, 20 s red light at 27 μmol m-2s-1 was sufficient to saturate the response. Exogenous gibberellic acid added in darkness in the presence of KCl also induced protoplast swelling. Gibberellins may act as an intermediate in the phytochrome-induced swelling of protoplasts.

    Notizen: The routine use of the INEPT pulse sequence is shown to be the method of choice for the assignment of multiplicity from 13C NMR spectra where no prior information about the 1J(CH) coupling constants is available, or where these are expected to vary over a large range. This technique is illustrated using spectra obtained from polyhalogenated monoterpenes isolated from the red alga Plocamium angustum, which have 1J(CH) values varying from 127 to 203 Hz.
