Notes: The adsorption of hydrogen on a cobalt(101¯0) surface was investigated in ultrahigh vacuum (UHV) between 85 and 500 K using Video-LEED, temperature-programmed thermal desorption (TPD), work function (ΔΦ) measurements, and high-resolution electron energy loss spectroscopy (HREELS). Between 90 and 200 K, hydrogen adsorbs dissociatively with high sticking coefficient (s0≥0.8) via precursor kinetics and forms, with increasing exposure, a c(2×4), a p2mg (2×1) and a (1×2) LEED structure (hydrogen coverages aitch-thetaH=0.5, 1.0, and 1.5, respectively). While the first two structures represent true ordered hydrogen phases there is strong evidence that the (1×2) phase is reconstructed, likely in a paired-row configuration. The formation of the (1×2) phase is slightly thermally activated; its decomposition produces a sharp thermal desorption maximum (α state) appearing on the low-energy side of a β-TPD signal which reflects the hydrogen desorbing from the unreconstructed surface. The activation energies for desorption from the α and β states are 62 and 80 kJ/mol, respectively. Chemisorption in the β state [(2×1) phase up to aitch-thetaH=1.0] is associated with a ΔΦ of +207 meV, while the fully developed (1×2) reconstructed phase (α state) causes a ΔΦ of approximately −122 meV resulting in an overall work function change of +85 meV at saturation. From HREELS, we determine the H adsorption site in all superstructures to be threefold with a local CS symmetry. Our results are discussed and compared with previous findings for similar metal–hydrogen interaction systems.

Notes: Background  Biological research suggests that vascular changes may play a major role in rosacea pathogenesis. Chrysanthellum indicum is a plant-based extract containing a unique combination of phenylpropenoic acids, flavonoids and saponosids, and has a well-documented effect on vascular wall permeability and increase of the mechanical resistance of capillaries. Objective  To determine the efficacy and safety of a cream containing 1%C. indicum extract with vitamin P properties in the treatment of rosacea. Methods  This study included 246 patients diagnosed clinically as having moderate rosacea. Patients were randomly allocated to C. indicum extract-based cream (n = 125) and placebo (n = 121) groups. Patients were advised to apply the products on their face twice a day for a 12-week period. The patients were examined at the end of each 4-week period. Severity of erythema (graded by reference to six photographs), surface of erythema and rosacea overall severity scores were recorded at each visit on days 0, 28, 56 and 84. Investigators carried out a final efficacy assessment at the end of week 12. Volunteers’ final overall efficacy assessment was recorded in a self-administered questionnaire. Adverse events were identified through examination, interview and collection of comments in patients’ questionnaires. Results  Treatment with the C. indicum extract-based cream resulted in significant improvement (P 〈 0.05) in severity of erythema, overall rosacea severity compared to baseline and placebo, and investigator and patient overall efficacy assessment scores (P = 0.046 and P = 0.001, respectively) compared with placebo scores. Adverse reactions were mild, and did not differ between the C. indicum extract-based cream and the placebo groups. Conclusion  Chrysanthellum indicum extract-based cream is an effective and well-tolerated topical agent for the treatment of moderate rosacea. The mode of action of the active ingredient suggests that additional efficacy might be expected from combination with other topical treatments.

Notes: Mutants (glk) of Streptomyces coelicolor A3(2) that are resistant to the non-utilizable glucose analogue 2-deoxyglucose are deficient in glucose kinase activity, defective in glucose repression, and usually unable to utilize glucose. A 2.9 kb Bcll fragment, previously shown to restore a wild-type phenotype to a glk deletion mutant that lacks the entire segment, contains two complete open reading frames that would encode proteins of 20.1 kDa (ORF2) and 33.1 kDa (ORF3). ORF3 is transcribed from its own promoter, and also from a promoter that initiates transcription upstream of ORF2. A derivative of the temperate phage πC31 containing ORF3 alone restored a wild-type phenotype when used to lysogenize the deletion mutant. The product of ORF3 is homologous to members of a family of repressor proteins encoded by xylR In Bacillus subtilis and Lactobacilius pentosus, and by nagC in Escherichia coli. Although this might suggest that ORF3 encodes a positive activator for glucose kinase, rather than the enzyme itself, ORF3 restored the ability to metabolize glucose to an E. coli glk mutant, and activity gels of cell extracts of E. coli containing ORF3 cloned in the pT7-7 expression vector demonstrated that the ORF3 product has glucose kinase activity.

Notes: Summary After intracutaneous injection of arginine-C-14 into guinea pigs or of ornithine-C-14 into hairless mice, respectively, a radiometabolite could be found in hydrolyzed, macromolecular epidermis fractions, which by column chromatography on Dowex 50 showed the same behavior as citrulline. In the ultrafiltrable fraction of the epidermis (“the water-soluble substances”), the detected radiometabolites seemed to be citrulline and urea.

Notes: Summary Hairless mice were killed 1–6 hours after intradermal injection of tritiated histidine. The radioactivity of the ultrafiltrable portion and the non-ultrafiltrable, by sephadex G-75 excluded (macromolecular) portion of the alkali-extract of the machanically separated epidermis was determinated. By column chromatography of the ultrafiltrate on Dowex 50, cis-and trans-isomers of urocanic acid were found to contain the main radioactivity. The hydrolysate of the macromolecular portion, separated in the same manner, showed urocanic acid besides histidine and other compounds as radioactive components. However, this was found only in animals, which were killed 1 hour after the injection.

Notes: Summary The incubation of separated epidermis of hairless mice, guinea pigs and men with adenine-2-14C, adenine-8-14C or uric acid-2-14C, respectively, resulted in catabolites like urea and allantoine. Only the studies with adenine-tracers showed further break down products, which allowed to assume an atypical pathway without uric acid as metabolite.

Notes: Summary Free amino acids, ammonia and urea from the superficial stratum corneum disjunctum of uninvolved skin of patients with microbiotic eczema and of healthy controls were determined by column chromatography. After separation by paper chromatography, urocanic acid was assayed spectrometrically and by use of isotope dilution analysis. Furthermore, the values for α-amino-N referring to the fresh weight of the scraped horny material were estimated. Most of the investigated substances showed no obvious changes between the two groups. Only differences were found for histidine, glutamic acid, allo-isoleucine (P 〈 0.05) and for tryptophan (P 〈 0.01). The results are discussed.

Notes: Summary Two unknown metabolites of adenine-C14 seem to be produced bydirect action of xanthine oxidase in separated epidermis. Added hypoxanthine (in excess of the tracer) or allopurinol as inhibitors of this enzyme stopped its formation. The same effect was caused by added ATP. The reaction is apparently based upon prefered deamination of the tracer by forming nucleosids or nucleotids, respectively, by which it is excluded from the “by-pathway”.
