    Notes: In a large, double-blind, multicentre study, 269 patients with confirmed endometriosis were randomly allocated to receive either danazol (200 mg twice daily; n= 137) or gestrinone (2.5 mg twice weekly; n= 132) for 6 months. The two groups were comparable in terms of the staging of endometriosis by the American Fertility Society (1979) score. After the sixth month of treatment, repeat laparoscopy was performed. Clinical assessment, haematological and biochemical investigations were carried out during the 6 months of treatment and for a further 12 months' follow-up and are compared between the two groups. A total of 15 patients from the gestrinone group, including four patients with hirsutism, and 17 patients from the danazol group, including six patients with headache, withdrew because of adverse symptoms. An additional 22 patients, including 10 from the gestrinone group and 12 from the danazol group withdrew because of lack of efficacy, pregnancy, elevated hepatic function tests or for reasons unrelated to the trial. Total American Fertility Society scoring showed an improvement of 73.3% in 101 patients receiving gestrinone and 72.7% in 99 patients receiving danazol. The results showed a significant reduction in the severity of dysmenorrhoea by the third month in the danazol group and at 6 months in both groups. There was a significant (P 〈 0.001) increase in weight observed in both groups during treatment. Overall, the tolerability of danazol and gestrinone was good; however, significantly more patients with gestrinone complained of hirsutism while significantly more with danazol complained of leg cramps. During the 12 months of follow-up, mild, moderate or severe degrees of lower abdominal pain, dysmenorrhoea and deep dyspareunia all fluctuated, with no statistically significant increase in frequency in either group.

Early diversification by gene-conversion and somatic hypermutation in rabbit appendix and chicken bursa of Fabricius are similar processes; the chicken bursa and the rabbit appendix have homologous functions. However, diversification in bursa starts during embryonic development whereas it starts in rabbit appendix about 2 weeks after birth in the presence of antigens and superantigens that ma) contribute to positive and negative selection, affect B-cell expansion and mold the repertoire. The biochemical steps leading to diversification by gene conversion are unknown. However elevated levels of RAD51 mRNA in both chicken bursa and young rabbit appendix suggest that repair of double stand breaks may be involved. The base changes found in expressed rabbit VH sequences derived from rearrangement of known germline VH genes followed by one or more gene conversions occur with frequencies similar to those found in analyses of somatic hypermutation. The Ser codons in CDR1 and CDR2 of rabbit VHl I genes are all AGY rather than TCN, suggesting that they may represent intrinsic hotspots for hypermutation comparable to those described in human and mouse VHr. Somatic hypermutation may further refine antibody affinities in rabbit germinal centers.

    Notes: Nerve growth factor (NGF) has been widely used in animal models to ameliorate age-related neurodegeneration, but it cannot cross the blood–brain barrier (BBB). NGF conjugated to an antibody against the transferrin receptor (OX-26) crosses the BBB and affects the biochemistry and morphology of NGF-deprived basal forebrain neurons. The rapid actions of NGF, including electrophysiological effects on these neurons, are not well understood. In the present study, two model systems in which basal forebrain neurons either respond dysfunctionally to NGF (aged rats) or do not have access to target-derived NGF (intraocular transplants of forebrain neurons) were tested. One group of transplanted and one group of aged animals received unconjugated OX-26 and NGF comixture as a control, while other groups received replacement NGF in the form of OX-26–NGF conjugate during the 3 months preceding the electrophysiological recording session. Neurons from animals in both the transplanted and aged control groups showed a significant increase in firing rate in response to acute NGF application, while none of the conjugate-treated groups or young intact rats showed any response. After the recordings, forebrain transplants and aged brains were immunocytochemically stained for the low-affinity NGF receptor. All conjugate treatment groups showed significantly greater staining intensity compared to controls. These data from both transplants and aged rats in situ indicate that NGF-deprived basal forebrain neurons respond to acute NGF with an increased firing rate. This novel finding may have importance even for long-term biological effects of this trophic factor in the basal forebrain.

    Notes: Biopsies of human gingiva were studied with special reference to the ultrastructural changes of the basal lamina and anchoring fibrils occurring after periods of in vitro cultivation. Characteristic changes were found in the basal lamina and anchoring fibrils. In the early stages in vitro, the basal lamina showed detachment from the epithelial cells, interruption, irregularity in thickness, and reduplication; the anchoring fibrils increased in number. In the later stages in vitro, the basal lamina showed a more severe detachment and fragmentation, proliferation into the connective tissue, and face–to–face coalescence, while the anchoring fibrils became stable in number and free of association with the connective tissue components.

    Notes: Pacific sand lance Ammodytes hexapterus in Kachemak Bay, Alaska, showed no sexual dimorphism in length-to-weight (gonad-free) ratio or length-at-age relationship. Most matured in their second year, males earlier in the season than females, but females (31%) attained a higher gonadosomatic index than males (21%). Sand lance spawned intertidally once each year in late September and October on fine gravel or sandy beaches soon after the seasonal peak in water temperatures. Sand lance in Cook Inlet and Prince William Sound displayed similar maturation schedules. Schools were dominated 2: 1 by males as they approached the intertidal zone at a site where spawning has taken place for decades. Sand lance spawned vigorously in dense formations, leaving scoured pits in beach sediments. Fecundity of females (93–199 mm) was proportional to length, ranging from 1468 to 16 081 ova per female. About half of the overall spawning school fecundity was derived from age group 1 females (55% of the school by number). Spawned eggs were 1·02 mm in diameter, demersal, slightly adhesive, and deposited in the intertidal just below the waterline. Sand lance embryos developed over 67 days through periods of intertidal exposure and sub-freezing air temperatures.

    Notes: A novel microsporidian parasite is described, which infects the crustacean host Gammarus duebeni. The parasite was transovarially transmitted and feminised host offspring. The life cycle was monomorphic with three stages. Meronts were found in host embryos, juveniles, and in the gonadal tissue of adults. Sporoblasts and spores were restricted to the gonad. Sporogony was disporoblastic giving rise to paired sporoblasts, which then differentiated to form spores. Spores were not found in regular groupings and there was no interfacial envelope. Spores were approximately 3.78 × 1.22 μm and had a thin exospore wall, a short polar filament, and an unusual granular polaroplast. All life cycle stages were diplokaryotic. A region from the parasite small subunit ribosomal RNA gene was amplified and sequenced. Phylogenetic analysis based on these data places the parasite within the genus Nosema. We have named the species Nosema granulosis based on the structure of the polaroplast.

    Notes: Summary With a self-neutralizing, rocket-borne parachute aspiration probe concentration and mobility measurements of positively and negatively charged carriers were made between 72 and 40 km over Sardinia. Between 72 km and 60 km a ‘heavy’ and a ‘light’ group of positively charged particles were found. The ‘heavy’ group was more abundant than the ‘hight’ group in the greater heights. For the negatively charged particles only one mobility group was found which roughly corresponded to the mobility of the ‘light’ group of the positively charged carriers. The probe was designed so that no electrons could enter the analyzer section of the aspiration system. By this the electron concentration was given by the difference in concentration between the positively and negatively charged particles.
