    Notes: RF Ablation and Creatine Kinase. Introduction: The primary mechanism of myocardial injury during radiofrequency (RF) catheter ablation in the heart is presumed to be thermal. Creatine Kinase has been measured in serum to assess the volume of myocardial injury after ablation. However, its thermal inactivation by RF ablation could lead to underestimation of the true volume of injury. Methods and Results: Serial RF lesions were created in 10 canine left ventricles in vivo, and serial serum Creatine Kinase activities were measured and compared to lesion volume. To assess the stability of myocardial Creatine Kinase during RF catheter ablation, 29 RF ablations were made on the epicardial surface of porcine left ventricle in vivo and a 2-mm core biopsy was rapidly removed. The cores were rapidly frozen, sectioned longitudinally in 1-mm slices, and homogenized in 0.3 M Tris buffer solution containing EDTA and dithiothreitol for subsequent analysis of Creatine Kinase activity. An additional 19 tissue cores from RF lesions were stained and used to determine mean lesion depth. Normal tissue biopsies were exposed to 60 seconds of hyperthermia (37° to 85°C, n = 190), or high-density RF current at 50X (0 to 100 mA/mm2, n = 50), and tissue Creatine Kinase activity was measured. There was no evidence of Creatine Kinase washout within the first 2 hours, and peak values were measured 5 to 7 hours postablation. Tissue Creatine Kinase activity in the first mm depth of RF lesions averaged 10% of control values and increased over the first 5 mm of lesion depth. The mean Creatine Kinase activity within the hemisphere of ablated myocardium was calculated to be 31% of control. Creatine Kinase activity declined significantly at temperatures above 65°C, but no difference in tissue Creatine Kinase activity was observed among differing levels of RF current exposure in the absence of significant heating. Conclusions: Creatine Kinase activity in myocardial tissue is significantly diminished within the RF lesion. Creatine Kinase activity is not stable at temperatures above 65°C, which are routinely achieved within the central zone of RF ablation, and is unaffected by RF current in the absence of hyperthermia. Measurements of serum Creatine Kinase activity after RF catheter ablation may significantly underestimate the volume of myocardial injury.

    Notes: Early and accurate diagnosis of Burkholderia cepacia infection is important, particularly if segregation is to prevent patient-to-patient transmission. We have examined the serum response to a B. cepacia-specific 80-kDa outer membrane protein. 21 patients colonised with B. cepacia and Pseudomonas aeruginosa for 2–51 months (mean 11 months) were age- and sex-matched with 21 patients colonised with P. aeruginosa but not B. cepacia. The 80-kDa protein was recovered by electroelution from outer membrane proteins, separated by SDS-PAGE, coated onto ELISA plates, reacted with patient sera diluted 1:200, protein A-peroxidase and chromogenic substrate. We found that 19/24 patients colonised with B. cepacia and P. aeruginosa had high values, 2/24 patients had intermediate values, and 2/24 patients had a low value. 20/21 patients colonised with P. aeruginosa alone had low values and 1/21 had an intermediate value. We found that in the longitudinal serum samples studied from four patients only one patient developed high values after the first isolation of B. cepacia suggesting that seroconversion does not occur immediately after the first sputum culture of B. cepacia. We conclude that an ELISA test using B. cepacia-specific 80-kDa outer membrane protein can distinguish B. cepacia colonised and non-colonised patients and may be useful in the early diagnosis of B. cepacia infection.

    Notes: Abstract Burkholderia cepacia (Pseudomonas cepacia) is now recognised as an important pathogen in cystic fibrosis patients, and several reports have suggested that sputum-culture-proven colonisation occurs despite the presence of specific antibody. In an attempt to establish the use of antibody studies as diagnostic and prognostic indicators of B. cepacia infection, we have examined the IgG response to B. cepacia outer membrane proteins and lipopolysaccharide in patients also colonised with P. aeruginosa. The B. cepacia strains were grown in a modified iron-depleted chemically defined medium and outer membrane components examined by SDS-PAGE and immunoblotting. IgG antibodies were detected against B. cepacia outer membrane antigens, which were not diminished by extensive preadsorption with P. aeruginosa. The response to B. cepacia O-antigen could be readily removed by adsorption of serum either with B. cepacia whole cells or purified LPS, whereas we were unable to adsorb anti-outer membrane protein antibodies using B. cepacia whole cells. The inability to adsorb anti-outer membrane protein antibodies using B. cepacia whole cells maybe due to non-exposed surface epitopes. Several B. cepacia sputum-culture negative patients colonised with P. aeruginosa had antibodies directed against B. cepacia outer membrane protein. This study suggests that there is a specific anti-B. cepacia LPS IgG response, which is not due to antibodies cross-reactive with P. aeruginosa. Our studies indicate that much of the B. cepacia anti-outer membrane protein response is specific and not attributable to reactivity against co-migrating LPS.

    Notes: Ellipsoid shaped akaganeite (β-FeOOH) particles, 70 nm long and 10 nm in diameter, were prepared by forced hydrolysis of ferric chloride. The β-FeOOH particles were suspended in a lamellar liquid crystal phase at 0 °C and reduced with dilute sodium borohydride solution to iron particles. These conditions allowed us to slow the reduction reaction and isolate partially reduced iron particles. Transmission electron microscopy images showed that the particles were being reduced from the outside and there were nanometer-sized spherical iron clusters on the surface. Fully reduced iron particles were chains of cubes, with the cubes about 20 to 25 nm in size. The cubes were connected either corner to corner, edge to edge, or face to face. The face to face chains of cubes had the desired acicular shape, with aspect ratios exceeding five. This face to face chain of cubes microstructure was also observed in commercial iron particles. © 1999 American Institute of Physics.

    Notes: Multimode simulations of the evolution of the laser-driven, ablative Rayleigh–Taylor instability on planar, plastic targets are performed in three dimensions, with FAST3D–CM. The initial mass density target perturbations are random, with a power law dependence of k−2, a RMS surface finish of 0.1 μm, and perturbation wave numbers ranging from 2π/dmax to (square root of)2×(12π/dmax), for dmax=128 μm. At early nonlinear times, the perturbations grow to tile the target with approximately hexagonal bubbles that are of the shortest, initially seeded wavelengths not stabilized by density gradients. This tiling occurs on a time scale that is comparable to the eddy turnover time of the dominant bubble wavelength. When the target thickness is large compared to the dominant, short wavelengths, the bubbles continue to burn into the target and to evolve to progressively longer spatial scales. Predictions from second-order mode coupling and saturation models are found to be consistent with the simulation results.

    Notes: A simple instrument to determine field emission tip work functions and shape functions from simultaneous current–voltage (I–V) characteristics and field emission energy distributions of field emitter tips and tip arrays is described. This instrument uses a cylindrical energy analyzer with a few correcting elements to simulate a hemispherical analyzer and provides a low cost and more compact alternative to a commercial hemispherical spectrometer.
