    Notes: Abstract The amiE gene of Brevibacterium sp. R312 encoding wide spectrum amidase was isolated by complementation of a Brevibacterium sp. mutant using a plasmid gene bank of chromosomal DNA. The amiE structural gene and its promoter were localized on a 1.8-kb fragment by subsequent subcloning and complementation studies. In Brevibacterium sp., the investigation of amidase activities related to one copy of the gene suggested that the regulation of the amiE gene expression was under negative control. High expression levels have been obtained in Brevibacterium sp. and, after substitution of the amiE promoter by the tac promoter, in Escherichia coli.

    Notes: Abstract A mutant strain of Candida molischiana was selected. Analysis of the exocellular activity of Candida molischiana 35M5N grown on different carbon sources revealed that the biosynthesis of β-glucosidase is derepressed in this yeast strain. The strain is not a hyper-producer mutant. There were no observed differences in the endocellular and parietal activities of the wild and mutant strains. However, the mutant strain produced 35-fold more enzyme than the wild-type in the culture medium with glucose as carbon source. When glucose was used as carbon source, the mutant strain produced 90% more exocellular enzyme than when cellobiose was used as the carbon source.

    Notes: Abstract: A rapid and effective method is described for electroporation of Brevibacterium sp. R312, a coryneform strain producing nitrile hydratase and amidase. The transformation efficiency of the method is 108 transformants per μg of plasmid under optimal conditions. Parameters optimised included field strength (11.8 kV cm−1), pulse length (2.4 ms), plasmid DNA concentration (0.25 μg ml−1 and cell density (1010 cells ml−1). Surprisingly, the transformation efficiency did not vary with the growth stage, in contrast to results in the literature. A shuttle vector was constructed containing several unique cloning sites down-stream of the SP6 RNA polymerase promoter.

    Notes: Summary Brevibacterium sp. strain R312 has an endocellular β-glucosidase, a nitrile hydratase and an amidase that can break down some cyanoglucosides. Nonsterile cassava pulp suspensions were fermented using this strain and 70%–80% reduction of nitrile compounds, in particular cyanoglucosides and α-hydroxynitriles, was observed. This type of nitrile-hydratase-active microorganism could be a solution for the detoxification of cassava. Experiments conducted with the yeast Candida molischiana and C. wickkerhamii showed no improvement in detoxification.

    Description / Table of Contents: Zusammenfassung Beträchtliche metabolische Veränderungen wurden bei Hefe, während der fünfzehn ersten Stunden der Sporenbildung unter Zugabe von Essigsäure, beobachtet. Der Kohlenhydratenvorrat, die Trockensubstanz und der Proteingehalt nehmen zu. Die Aktivität der Fumarase und Isocitritase wird kaum verändert. Dadurch wird die Annahme bestätigt, daß eine Abweichung des Metabolismus der Essigsäure während der Sporenbildung stattfindet.
    Notes: Abstract We have noticed important metabolic modifications during the first 15 hours of the sporulation process of yeast in contact with acetic acid. Carbohydrate reserves, dry weight and proteins increase. On the other hand, the activity of fumarase and isocitritase in cell homogenates are hardly altered. The results prove true the hypothesis according to which the metabolism of acetic acid is altered during sporulation.

    Notes: Summary Theophylline levels in plasma, erythrocytes and cantharides-induced blister fluid (BF) have been correlated with peak expiratory flow (PEF) in asthmatic patients. Nine asthmatic men received an IV infusion of 240 mg theophylline at a constant rate for 30 min. The theophylline concentrations in erythrocytes and blister fluid were 37 % and 78 % of those in plasma. There was not a strong correlation between erythrocyte or blister fluid penetration and the clinical effect of theophylline.

    Description / Table of Contents: Résumé L'accès des lésions du rectum moyen peut être difficile. Ce travail résume notre expérience d'un abord postérieur du rectum chez 22 hommes et 13 femmes d'une âge de 21 à 96 ans. Les indications chirurgicales comprennent des tumeurs villeuses, des prolapsus du rectum, des sténoses rectales et des fistules du rectum. Aucune complication post-opératoire ne fut observée chez 20 malades, toutefois une fistule se developpa chez 7 patients dont 3 durent être traités par colostomie proximale et cure chirurgicale. 4 fistules guérirent spontanément. 2 malades ont developpés une éventration périnéale. L'examen anatomopathologique des tumeurs villeuses a montré une infiltration néoplasique dans trois cas nécessitant une résection rectale avec une anastomose coloanale termino-terminale. Chez 2 malades une incontinence légère est apparue traitée par Biofeedback. Une douleur périnéale résiduelle est rapportée par 2 malades. L'abord postérieur du rectum est particulièrement utile pour les lésions bénignes trop hautes pour une exérèse trans anale et trop basse pour une résection abdominale.
    Notes: Abstract Access to lesions in the mid-rectum can be difficult. This report summarizes our experience with a posterior approach to the rectum in 22 men and 13 women, age range 21 to 96 years. Surgical indications included villous tumours, rectal prolapse, rectal strictures or rectal fistulae. No postoperative complications were observed in 20 patients, but fistulae developed in seven patients, of whom three required proximal colostomy and surgical treatment. Four healed spontaneously. Two patients developed sacrococcygeal hernia. Pathologic examination of villous tumour showed extensive malignant change in three cases requiring rectal resection with end-to-end colo-anal anastomosis. In two patients mild incontinence developed, treated by biofeedback. Residual peri-anal pain was reported by two patients. The posterior approach to the rectum is particularly useful for benign lesions too high for a transanal resection and too low for a transabdominal resection.
