Conclusion Intracellular determination of sulphatide by the two methods gave very good correlation. The detection limit for the colorimetric methods was in the low nanomolar range per mg protein, providing enough specificity and sensitivity for the determination of sulphatide in fibroblasts after a loading test. While the determination of radiolabelled 3-OH-sulphatide is quite time-consuming and requires special techniques, the colorimetric assay is relatively simple and can be performed in any laboratory equipped for biochemical analyses. The colorimetric test can be used to develop more sensitive sulphatide loading tests which will be better able to discriminate between the various forms of typical and atypical MLD.
