    Notes: Summary Potassium cyanide (KCN) resistant peroxidase is generally accepted by hematologists as a selective stain for the eosinophilic cell line. However, it has been demonstrated biochemically that not only the peroxidases of neutrophils but also those of eosinophils can be inhibited by KCN. Therefore, bone marrow smears of hematologically normal patients were subjected to the peroxidase reaction in the presence of varying concentrations of KCN. It was found that with increasing concentrations of KCN not only neutrophils but also eosinophils were inhibited. Moreover, there were always neutrophilic promyelocytes that were still positive when a considerable number of the eosinophils was already inhibited. Therefore, it can be concluded from our results as well as from biochemical data that there is no concentration of KCN which demonstrates the total of the eosinophilic cell line selectively. The implications of these findings are discussed.

    Notes: Summary Thirteen lymph node biopsy specimens with Hodgkin's disease (six cases of the nodular sclerosis type; six cases of the mixed cellularity type; one case of the lympocytic depletion type) were studied with scanning cytophotometric measurements. The DNA content of small lymphocytes, large lymphocytes, atypical mononuclear cells and Hodgkin cells was measured. The sizes of the nuclei and mitotic indices were also measured. There were no lymphocytes with aneuploid DNA stemlines. Large lymphocytes showed proliferative activity which is possibly reactive, although its neoplastic character cannot be excluded. Hodgkin cells exhibited aneuploid values in seven cases and atypical mononuclear cells in only four cases. The nuclear sizes and the DNA content of the atypical mononuclear cells ranged between the values of lymphocytes and Hodgkin cells. The mixed cellularity type showed higher DNA values, more frequently occurrence of aneuploid stemlines and a higher frequency of mitotic figures than did the nodular sclerosis type. The results confirm the different grades of malignancy of the various histopathological categories of Hodgkin's disease.

    Notes: Summary Morphological and cytochemical findings in a case of acute myeloid leukemia (AML) with 3 relapses are described. The disease began as a subtype M4 with atypical eosinophils according to the French-American-British (FAB)-classification. At the time of the first relapse, the phenotype had converted into an M2-subtype, while the second relapse presented the picture of an M1-type of acute myeloid leukemia with non-granulated peroxidase-positive blasts. When the third relapse occurred, the diagnosis was again an M4-type of acute myeloid leukemia. The observed changes of the phenotype can be interpreted as an expression of tumor cell heterogeneity, even though the observed emergence of new leukemic clones would be attributed entirely or in part to effects of the applied cytostatic drugs, because tumor cell heterogeneity concerns also the response to therapy, which may result in a growth advantage for certain leukemic subpopulations. In so far, AML behaves like all other human malignant neoplasias. In addition, morphological changes in leukemias such as are reported here, have been observed long before the introduction of cytostatic drugs. Also, the validity of the FAB-classification is discussed in the light of the presented observation. It is concluded that the FAB-classification is not a classification in a strict sense, but merely a crude and simplistic list of subtypes of acute myeloproliferative diseases that is difficult to reproduce and that disregards major aspects of this group of diseases, for instance tumor cell heterogeneity.

    Notes: Summary The presence of alkaline phosphatase was investigated in slide preparations and smears of lymph nodes with the azo dye method. In the case of normal or hyperplastic lymph nodes, capillaries and arterioles are regularly and most strongly positive; the phosphatase activity in the endothelium of the venules, the larger blood vessels and the lymph vessels is irregular. One occasionally finds a few positive fibroblasts in the capsule and trabeculae. In addition, the pulpa sometimes shows a reaction in the region of the sinus, especially of the marginal sinus, and in the region of the germinal centers which appears to be associated with fibers. The activity in the region of the germinal centers is most fully developed with severe follicular lymphatic hyperplasia. A considerable degree of alkaline phosphatase activity can be noted at the edge of tubercles and tubercular caseous foci and in the scarified areas of the lympho-granulomatosis. In lymph node smears, elements interpreted as large reticulum cells were found to be about 16% phosphatase active; in part these cells are for certain vascular endothelium and fibroblasts. In the smears, we also found small lymphoid reticulum cells which gave a clear cut phosphatase reaction. No alkaline phosphatase activity could be demonstrated in the tissue mast cells, at least, that is, in their granula. Phosphatase activity could be clearly demonstrated in slide preparations as in smears in a part of the cells from 2 lymphosarcomas.

Die einkernigen Entzündungszellen der Hautfensterpräparate enthielten in 40,2% einen, in 43,0% zwei, in 14,4% drei, in 2,2% vier und in 0,2% fünf Nucleolen. 3. Diese Verteilung wurde zu jedem Zeitpunkt des Entzündungsablaufes (bis 36 Std) unabhängig von der Häufigkeit des Deckglaswechsels gefunden. 4. Die Anordnung der Nucleolarsubstanz in den einkernigen Entzündungszellen zeigte zu keinem Zeitpunkt Übereinstimmung oder Ähnlichkeit mit den Nucleolen von Blutlymphocyten. 5. Für eine auf den Deckglaspräparaten zu verfolgende Entwicklung von Makrophagen aus Lymphocyten ergab die Untersuchung keinen Anhalt. 6. Die Befunde sprechen dafür, daß die einkernigen Zellen der akuten Entzündung genetisch einheitlich sind.
