    Notes: The omega function, defined for strong and monovalent weak electrolytes, is a useful regulating principle for the description of electrophoretic processes. The omega function profile, which is established by the initial electrolyte distribution in an electrophoretic column, will not be changed by current flow. The function is based solely upon migration and is only applicable to those regions where diffusional dispersion can be neglected. Computer simulations are used to follow the evolution of the concentration gradients of various electrophoretic systems and to verify the proposed constancy of the omega function profile. The limitations of the theory are investigated and the value of these predictions to the experimentalist is discussed.

    Notes: A commercially available electrooptical device, coupled to a microcomputer, is capable of analyzing densitometric values with a resolution of 280 × 128 points (35 840 individual points). A complex autoradiogram of a two-dimensional electrophoretic gel is completely sorted into X and Y coordinates and exposure values differeing by as little as 0.01 O.D. (optical density) units or over an O.D. range of 0 to 4.00 in less than 2 min. For other purposes such as comparison of two gels, immunoelectrophoretic patterns, enzyme linked immunosorbent assay or microtitration plates, dot-blot assays, or tissue samples, a comparison may be made and all differences stored in the computer in less than one second and further decisions or computer-driven actions decided on that basis.

    Notes: The general equations which describe the electrophoretic transport of components in solution are restated using Newman's general concept of mobilities. A concise derivation of the moving boundary equation and the regulating function ω from the continuity equation is presented. Various other regulating principles across moving and stationary boundaries are also discussed, which permits a review of the features and interrelationshpis of the electrophoretic models based on electromigration only. The effect of considering an interactive (dissociating) solvent on the mathematical treatment is discussed.

    Notes: The electric field variation across moving boundaries in three component systems is analyzed with a model based on electromigration only (moving boundary equation and regulating function ω). A decrease in the electric field strength across a boundary in the direction of migration is the most general requirement for the establishment of steady state migrating boundaries. The formation of that electric field gradient is shown to be based on two fundamental constraints. These are (i) the higher net mobility of the leading component compared to that of the terminating constituent and (ii) the higher concentration ratio of the leading component to the terminating component ahead of the moving boundary compared to that in the terminating zone. The relations are discussed with respect to strong and weak electrolytes where all three components are present on either side of the boundary. The predictions are verified experimentally. Limitations of the model based on migration only are discussed.

    Notes: A digital simulation of the behavior of amphoteric sample components in moving steady state boundaries is presented. Complete computer simulation data, including profiles of concentration, conductivity and pH as functions of time, are given for both cationic and anionic electrolyte configurations which incorporate one amphoteric sample constituent. The condensation of ampholytes in steady state moving boundaries is shown to proceed via an isotachophoretic mechanism and not by isoelectric focusing. Mobility (velocity) relationships necessary for sample components to form steady state zones are discussed.

    Notes: Computer simulation is used to analyze a system of two electrophoretic columns coupled by mixing the anolyte of one with the catholyte of the other. A mathematical model is presented which is used to predict the pH gradients formed by monovalent buffers in this system, when the currents in the columns are unequal. In the column with the higher current a pH gradient is created which increases from anode to cathode and is potentially useful for isoelectric focusing. The breadth of this gradient is dependent upon the ratio of the currents. The function of the second column is the compensation of buffer migration which occurs in the first column, thereby maintaining constant electrolyte composition. The effects of buffer pKs and mobilities are evaluated.

    Notes: Isoelectric focusing in the continuous flow mode can be more quickly and economically performed by admitting a stepwise pH gradient composed of simple buffers instead of uniform mixtures of synthetic carrier ampholytes. The time-consuming formation of the pH gradient by the electric field is thereby omitted. The stability of a three-step system with arginine - morpholinoethanesulfonic acid/glycylglycine - aspartic acid is analyzed theoretically by one-dimensional computer simulation as well as experimentally at various flow rates in a continuous flow apparatus. Excellent agreement between experimental and theoretical data was obtained. This metastable configuration was found to be suitable for focusing of proteins under continuous flow conditions. The influence of various combinations of electrolytes and membranes between electrophoresis chamber and electrode compartments is also discussed.

    Notes: The impact of the type of membrane used to separate the electrolyte compartments from the separation cell for continuous flow field step electrophoresis is examined using both experimental and computer simulation data. It is shown that the distinct transport characteristics of ion exchange membranes and dialysis membranes may have an effect on the stability of the pH and conductivity gradients created by the buffer system. These effects are interpreted using computer simulation data. The limitations of the model used to portray the transport characteristics of ion exchange membranes are discussed as are the experimental conditions which provide the greatest stability for field step electrophoresis.
