Plantarum 69: 517–522.A synthetic auxin 2-(2-methyl-4-chloro)phenoxypropionic acid (MCPP), analogue of 2,4-D, alters maize (Zea mays L. H-30) germination while inducing callus formation. The effect of this auxin on protein synthesis and phosphorylation of the embryonic tissues was explored. Total cytoplasmic proteins were analysed for 14C or 32P incorporation into trichloroacetic acid precipitable material. MCPP significantly stimulated protein synthesis as well as protein phosphorylation. The protein synthesis pattern was highly altered in the presence of MCPP as analysed by two-dimensional gel electrophoresis. Analyses by Sephadex G-100 chromatography and by two-dimensional gel electrophoresis of phosphorylated proteins indicate that the effect of MCPP on protein phosphorylation was only quantitative.

    Notes: Summary 1. The medicinal leech crawls along a solid substrate by repeated alternating extensions and shortenings of the body. Extension occurs with the posterior sucker attached and the head sucker free. The head sucker then attaches, followed by shortening and release of the tail sucker. The tail sucker is then pulled toward the head, where it reattaches to the substrate. The head sucker then releases, and another crawling cycle begins (Figs. 1, 5). 2. There are two crawling variants: inchworm crawling, in which the head and tail suckers are closely apposed at the end of a cycle and the body forms a loop above the substrate, and vermiform crawling, in which the suckers are placed farther apart and the body remains fairly close to the substrate (Fig. 1). 3. The cycle period and the distance traveled during a cycle are greater in inchworm than in vermiform crawling; however, the velocity of travel is the same for both (Fig. 2). 4. For both variants, the interval between head sucker attachment and tail sucker release is similar at all cycle periods and has a value consistent with direct interneuronal conduction of a signal from head sucker sensory neurons to tail sucker motor neurons. The interval between tail sucker attachment and head sucker release, however, is longer and varies with the cycle period, suggesting a more complex interneuronal circuit in the pathway from tail sucker sensory neurons to head sucker motor neurons (Fig. 4). 5. The onsets of the components of the crawling cycle (extension, post-extension pause, shortening, and post-shortening pause) show an anteroposterior lag (Figs. 5, 7). For both variants, the travel time between segments varies directly with the period (Fig. 8). 6. For both crawl types, the durations of the cycle components vary directly with the period, with several exceptions (Figs. 9, 10). 7. A model is presented that summarizes the coordination of the various motor events in a cycle of leech crawling (Figs. 11 and 12).

    Notes: Abstract Three commercial methods (enzyme immunoassay, fluoroimmunoassay and latex agglutination) and a complement fixation test were compared for detection of cytomegalovirus antibodies using a panel of 490 serum samples from blood and organ donors, from immunocompromised patients and paired sera from five patients with recent cytomegalovirus infection. An indirect immunofluorescence test for antibodies to cytomegalovirus was used for classifying samples giving discrepant results by any of the four methods. All methods showed high sensitivity and specificity, but the enzyme immunoassay and the latex agglutination tests had the highest sensitivity. Latex agglutination is recommended for large-scale screening of cytomegalovirus antibodies in blood and organ donors. Negative results obtained by latex agglutination should be confirmed by sensitive enzyme immunoassays.

    Notes: Abstract The percentage of parasitisation and index of adherence ofTrypanosoma cruzi has been studied when host HeLa cells or metacyclic forms were pretreated with neuraminidase or with poly-l-lysine. The percentage of parasitisation was significatively reduced (P≦0.001) when cells were pretreated with poly-l-lysine while pretreatment with neuraminidase caused no apparent effects. On the other hand, the adherence of the metacyclic forms pretreated with poly-l-lysine or neuraminidase was significantly higher than that of the control group.

    Notes: Abstract A method is described for the isolation and purification of the intracellular amastigotes ofTrypanosoma cruzi from cultured Vero cells. Host cells were infected with metacyclic forms obtained in Grace's medium. Six days after infection, the cells were subjected to treatment with trypsin to obtain the intracellular forms. The parasites were collected and purified by Percoll discontinuous gradient centrifugation.

    Notes: Many studies of calcium phosphate precipitation have been made using relaxation techniques in which the concentrations of the lattice ions are allowed to decrease as equilibrium is approached. Since the nature of the phases that form depend markedly on the solution composition, this decrease can lead to concomitant phase transformations during the crystallization experiments. The results of the present constant composition (CC) studies show that defect apatites may be formed under conditions of sustained supersaturation with a non-stoichiometric coefficient dependent on the pH of the growth medium. An important factor in analyzing these experiments is the initial surface modification and ion-exchange processes involving H+ and Ca2+ ions after inoculation of the supersaturated solutions. Thereafter, active growth sites may be eliminated as the crystals undergo lattice perfection. Transformation of dicalcium phosphate dihydrate to octacalcium phosphate, involving dissolution and subsequent nucleation and growth of the new phase, is also influenced by surface roughening of the initial phase. Typical inhibitors that reduce the rate of growth of seed crystals in supersaturated solutions may actually induce the nucleation of calcium phosphate phases when immobilized on inert surfaces. This may be a factor in the modulation of crystal growth in many biological systems.
