Summary The influence of several drugs on the binding of promazine and chlorpromazine to bovine serum albumin was studied by means of Sephadex gel filtration. The following results were obtained: 1. Promazine was displaced from its protein binding sites by acidic as well as by basic drugs. 2. Highly bound drugs were not effective or were comparatively less effective as promazine-displacing agents. 3. Suramine, sulfamethoxydiazine, tetracycline, oxytetracycline and chlortetracycline caused an increase of promazine and chlorpromazine binding to albumin. 4. The viscosity of drug-containing albumin solutions suggested an alteration in the structure of the albumin molecule by the drugs. Different factors were discussed, which may either increase or decrease the binding ability of an albumin solution.
