    Notizen: The various treatments of sedimentation equilibrium are compared on a theoretical and an experimental basis. Particular attention is paid to the polyelectrolyte nature of the problem and the choice of a neutral component. The effective density gradients of several cesium salts for DNA are measured. Two previous theories for the effective density gradient are shown to be equivalent, and the experimental values are interpreted with respect to these theories. It is clear t hat sedimentation equilibrium in a density gradient may be used for the determination of unambiguous molecular weights.

    Notizen: Crystal structures and electrical properties of radical-cation salts of the chiral organic donor TMET (S,S,S,S,-bis-(dimethylethylenedithio)tetrathiafulvalene) are described. Two structural types, 2:1 with octahedral anions Pf-6, AsF-6, SbF-6, I-3 (incommensurate), and 3:2 with tetrahedral anions BF-4, CIO-4, ReO-4 are observed. Resistivity measurements between 2 and 298 K indicate that the 3:2 types are organic metals, while the other compounds are semiconductors. (TMET)3(CIO4)2 is metallic down to about 120 K at ambient pressure and remains metallic down to 2 K at 8 kbar.

The influence of temperature on the linewidth and chemical shift indicates an equilibrium of Cu(CH3CN)+4 and Cu(C5H5N)+4 with another complex of lower symmetry. The preferential solvation of Cu (I) by pyridin in a mixture acetonitrile/pyridine is clearly shown.

    Notizen: The preparation of the new organic metallic compound (TSeT)2-I is reported. The crystal structure of the free donor, TSeT, is discussed and compared with the crystal structure of (TSeT)2-I. The latter has the orthorhombic space group P21212 and hence is not isomorphous with the analogous compounds (TSeT)2-Cl and (TSeT)2-Br, described by Shchegolev et al. [4].In contrast to those complexes (TSeT)2-I consists of only one set of crystallographically equivalent chains and therefore its transition to the low-temperature conducting state (Fig. 6) is also of different type.The d. c. resistivity at 300 K ranges from 10-3 to 1.5 ·. 10-4 Ω cm. The temperature dependence of the d.c. resistivity of the title compound is given: in the temperature range from 300 to 80 K the normalized d.c. resistivity is described by ρ(T)/ρ(300) ∝ T1.7. Between 80 and 70 K a slight deviation from the above law has been detected which is taken as an indication of a phase transition; its nature is, however, not yet definable. The low-temperature conducting state reached at about 70 K shows an almost constant resistivity in the interval 60 K to 45 K. Its normalized d.c. resistivity in the range 4.5 K-50 K is described by ρ(T)/ρ(300) ∝ (T-Tm)3, with Tm = 50 K. The value of the normalized resistivity extrapolated to O K is 0.75.

    Notizen: A previously developed immobilization technique involving latex coatings on solid particulate supports was investigated further for penicillin G production by Penicillium chrysogenum. Several modifications were found to decrease the germination lag time, including a higher spore concentration, a thinner latex layer, an increased latex porosity, and a decreased drying time. This approach enabled the development of immobilized mycelial pellets within 2-3 days from the onset of biocatalyst preparation and incubation.A continuous immobilized-cell airlift bioreactor produced penicillin G in a series of runs in which the production phase lasted up to 30 days. The productivity of this system was 3-6 times greater than the productivity of the corresponding free-cell shake flask fermentation.

    Notizen: (R,S)-N-Acetyl-leucine-2-naphthylamide can be resolved into its enantiomers on a (S)-DNBLeucine chiral stationary phase. The mechanism of this separation was investigated using as model complexes the 1:1 S,S- and the 1:1 R,S-cocrystals of the above naphthylamide and N-3,5-dinitrobenzoylleucinemethylamide, a soluble analogue of the stationary phase. The observed enantiomeric elution order can be rationalized from their X-ray data.

    Notizen: The product of the c-fms proto-oncogene is related to, and possibly identical with, the receptor for the macrophage colony-stimulating factor, M-CSF (CSF-1). Unlike the product of the v-erbB oncogene, which is a truncated version of the EGF receptor, the glycoprotein encoded by the v-fms oncogene retains an intact extracellular ligand-binding domain so that cells transformed by v-fms express CSF-1 receptors at their surface. Although fibroblasts susceptible to transformation by v-fms generally produce CSF-1, v-fms-mediated transformation does not depend on an exogenous source of the growth factor, and neutralizing antibodies to CSF-1 do not affect the transformed phenotype. An alteration of the v-fms gene product at its extreme carboxyl-terminus represents the major structural difference between it and the c-fms-coded glycoprotein and may affect the tyrosine kinase activity of the v-fms-coded receptor. Consistent with this interpretation, tyrosine phosphorylation of the v-fms products in membranes was observed in the absence of CSF-1 and was not enhanced by addition of the murine growth factor. Cells transformed by v-fms have a constitutively elevated specific activity of a guanir.c nucleotide-dependent, phosphatidylinositol-4,5-diphosphate-specific phospholipase C. We speculate that the tyrosine kinase activity of the v-fms/c-fms gene products may be coupled to this phospholipase C, possibly through a G regulatory protein, thereby increasing phosphatidylinositol turnover and generating the intracellular second messengers diacylglycerol and inositol triphosphatc.

    Notizen: The macrophage colony-stimulating factor, CSF-1 (M-CSF), is a homodimeric glycoprotein required for the lineage-specific growth of cells of the mononuclear phagocyte series. Apart from its role in stimulating the proliferation of bone marrow-derived precursors of monocytes and macrophages, CSF-1 acts as a survival factor and primes mature macrophages to carry out differentiated functions. Each of the actions of CSF-1 are mediated through its binding to a single class of high-affinity receptors expressed on monocytes, macrophages, and their committed progenitors. The CSF-1 receptor (CSF-1R) is encoded by the c-fms proto-oncogene, and is one of a family of growth factor receptors that exhibits an intrinsic tyrosine-specific protein kinase activity. Transduction of c-fms sequences as a viral oncogene (v-fms) in the McDonough (SM) and HZ-5 strains of feline sarcoma virus has resulted in alterations in receptor coding sequences that affect its activity as a tyrosine kinase and provide persistent signals for cell growth in the absence of its ligand. The genetic alterations in the c-fms gene that unmask its latent transforming potential abrogate its lineage-specific activity and enable v-fms to transform a variety of cells that do not normally express CSF-1 receptors.

    Notizen: The problem of determining the molecular weight of DNA samples by sedimentation equilibrium in a buoyant-density gradient is considered for the case of DNA samples with density heterogeneity. By determining apparent molecular weights in two or more buoyant mediums, quantitative measure of the amount of density heterogeneity can be determined. This method may be employed to determine both the true molecular weight and the extent of base composition heterogeneity.
