Notizen: Based on the observation of large conductance states formed by Bacillus thuringiensis Cry toxins in synthetic planar lipid bilayers and the estimation of a pore size of 10–20 Å, it has been proposed that the pore could be formed by an oligomer containing four to six Cry toxin monomers. However, there is a lack of information regarding the insertion of Cry toxins into the membrane and oligomer formation. Here we provide direct evidence showing that the intermolecular interaction between Cry1Ab toxin monomers is a necessary step for pore formation and toxicity. Two Cry1Ab mutant proteins affected in different steps of their mode of action (F371A in receptor binding and H168F in pore formation) were affected in toxicity against Manduca sexta larvae. Binding analysis showed that F371A protein bound more efficiently to M. sexta brush border membrane vesicles when mixed with H168F in a one to one ratio. These mutant proteins also recovered pore-formation activity, measured with a fluorescent dye with isolated brush border membrane vesicles, and toxicity against M. sexta larvae when mixed, showing that monomers affected in different steps of their mode of action can form functional hetero-oligomers.

Notizen: Abstract The L5 ribosomal protein gene (rpl5) and a S14 ribosomal protein pseudogene were identified by sequence analysis in the potato mitochondrial genome. The two genes are separated by one nucleotide and are found upstream of the apocytochrome b gene (cob), an arrangement conserved also in Arabidopsis and Brassica. The rpl5 gene has an intact open reading frame while the rpsl4 locus is disrupted by a five nucleotide duplication that introduces a frameshift in the reading frame. Editing of rpl5 and pseudorps14 cotranscripts has been studied by cDNA sequence analysis. Eight C residues are edited into U in the rpl5 coding region, resulting in eight amino acid changes that increase the homology between potato and other RPL5 polypeptides. Interestingly, the rps14 pseudogene sequence is not edited at any nucleotide position.

Notizen: Abstract Over a period of 7 years the biology and phenotypic variability of Chusquea culeou were studied at 5 locations in cool temperate forests of southern Argentina. Excavated rhizomes had an average of 1.1 successful rhizome buds, and an average of 2.1 years elapsed between successive generations of rhizomes. Rhizome buds usually develop within the first four years after a rhizome forms. Height, volume and weight of a culm can be calculated from its diameter 1 m above the ground. Culm size, length of foliage leaf blades, and pattern of secondary branching differed among study sites. Dead culms were numerous and commonly remained erect for more than 7 years after dying. New culm shoots appear in spring and reach full size within a few months. Shoots can grow more than 9 cm/day. Less than half of the shoots survived a year; most were killed by moth larvae. Multiple primary branch buds emerge through the culm leaf sheaths in the second spring. The mean number of branch buds at mid-culm nodes varied between 34.8 and 81.5, and the mean number of primary branches was between 22.8 and 40.8. Number and length of branches, and number and length of foliage leaf blades at each node is related to the position of the node on a culm. Most branches grow about 3 cm and produce 1 to 3 foliage leaves annually. Foliage leaf blades generally live 2 years or more; few survive 6 years. Relative lengths of foliage leaf blades and their spacing along a branch permit recognition of annual cohorts. Both gregarious and sporadic flowering have been reported, and every year a few isolated plants flower and die. Length of the life cycle is unknown. Seedlings require up to 15 years to produce culms of mature size. Foliage branches may live more than 23 years, and culms may survive 33 years. Extensive loss of new shoots to predation suggests that gregarious flowering may be driven by a need to escape parasitism. C. culeou clumps expand slowly. Average annual rate of increase of the number of live culms in a clump was 4.6%. Methods of seed dispersal are undocumented. A dense stand of Chusquea culeou had an estimated phytomass of 179 tons/hectare (dry weight), 28% of which was underground. Net annual production was about 16 t/ha dry weight.

Notizen: Abstract Collaborative research in Spain in the field of pharmacology is studied. Co-authored papers in periodicals were one of the indicators used to quantify collaborative research results. Through Spanish publications of pharmacologists, collaborations between different institutions in the same city are mostly detected. Through foreign papers quite different networks were found, both amongst Spanish cities and with institutions in other countries. In case of Spanish pharmaceutical industry questionnaires were used to determine their links with other institutions, mostly through research projects.

Notizen: Abstract The coverage of Latin American journals by international databases influences the visibility of these countries' scientific output, and has a direct effect in their activity index per scientific discipline. Local, regional and international character of the most visible Latin American journals in SCI expanded and restricted databases is analysed, as well as its influence in the percentage share of each country in certain disciplines. Suggestions to enhance visibility of local journals are presented.

Notizen: Abstract The delimitation of a research field in bibliometric studies presents the problem of the diversity of subject classifications used in the sources of input and output data. Classification of documents according to thematic codes or keywords is the most accurate method, mainly used in specialised bibliographic or patent databases. Classification of journals in disciplines presents lower specificity, and some shortcomings as the change over time of both journals and disciplines and the increasing interdisciplinarity of research. Differences in the criteria in which input and output data classifications are based obliges to aggregate data in order to match them. Standardization of subject classifications emerges as an important point in bibliometric studies in order to allow international comparisons, although flexibility is needed to meet the needs of local studies.

Notizen: Abstract A sample of “Citation Classics” in three scientific fields was studied to uncover citing motivations. The classics were classified into basic research, methods and reviews. Number of citations received per classic, number of authors, and age of classic per category and scientific field were the parameters studied. Journals and countries accounting for the highest incidence of classics were examined. A striking parallelism was found in the parameters applied to the categories in the scientific fields studied. This parallelism suggests similar citing habits of scientists in the fields studied which should be reflected in the structures of science obtained through citation grounded bibliometric models.
