Notes: 3-Deoxy-D-manno-octulosonate 8-phosphate (KDO8P) synthase catalyzes the biosynthesis of an essential component of the lipopolysaccharide of all Gram-negative bacteria. The structure and mechanism of KDO8P synthase are being actively studied as this enzyme represents an important target for antibiotic therapy. The structure of the Escherichia coli KDO8P synthase in cubic crystals (space group I23) has recently been determined and the enzyme shown to be a tetramer of identical subunits. However, this information is challenged by biochemical studies, which suggest that the enzyme behaves in solution as a homotrimer. Here, the preparation and preliminary X-ray analysis of monoclinic crystals of KDO8P synthase are reported. The crystals belong to space group P21, with unit-cell parameters a ∼ 50, b ∼ 140, c ∼ 74 Å, β ∼ 105°. The structure of KDO8P synthase in the monoclinic crystal form was determined by molecular replacement, using as a search model one of the subunits of the enzyme in the cubic crystals. A tetramer of KDO8P synthase with 222 local symmetry is also present in the asymmetric unit of the P21 crystals, with a solvent content of 43%. The observation that the same quaternary structure of KDO8P synthase is observed in two different crystal forms belonging to distinct crystal systems (monoclinic and cubic) suggests that a tetramer is the native form of the enzyme.

Description / Table of Contents: Polymere mit Urethaneinheiten wurden durch Lösungspolymerisation der Bisphenole A, C und F mit Diisocyanaten wie Toluol-2,4-diisocyanat, Hexamethylendiisocyanat, Isophorondiisocyanat bzw. Diphenylmethandiisocyanat hergestellt.
Notes: Polymers containing urethane units were synthesized by solution polymerization of bisphenols A, C, and F with diisocyanates viz. toluene-2,4-diisocyanate, hexamethylene diisocyanate, isophorone diisocyanate, and diphenylmethane diisocyanate, respectively. Thus obtained polymers were pale yellow amorphous powders. These polymers were characterized by infrared spectral studies and nitrogen content. The number-average molecular weights (M̄n) were estimated from conductometric titration and limiting viscosity number [η] in dimethyl formamide. From thermogravimetric analyses, the thermal behaviour was studied. Kinetic parameters for their degradation were computed.

Notes: Liquid prepolyesters (PPE) were obtained from castor oil and dibasic acids, viz. oxalic, malonic, succinic, glutaric, adipic, suberic and sebacic acids. All these prepolyesters were subsequently copolymerized with acrylamide by radical polymerization initiated with benzoyl peroxide. The resulting prepolyester (PPE)/polyacrylamide (PAm) interpenetrating polymer networks (IPN's) were characterized by elemental analyses, solubility behaviour, IR spectral study, differential scanning calorimetry and thermogravimetry. The transition behaviour was analyses for one IPN by the Ozawa and the Kissinger methods for calculating the various kinetic parameters.

Notes: Castor oil is reacted with hexamethylene diisocyanate under different experimental conditions varying the NCO/OH ratio to yield liquid prepolyurethanes (PPU's). All these polyurethanes were interpenetrated with methyl methacrylate (MMA) and a crosslinker EGDM by radical polymerization initiated by benzoyl peroxide. The novel PPU/MMA interpenetrating polymer networks (IPN's) were obtained as tough films by transfer molding. The characterization of these IPN's includes resistance to chemical reagents, thermal behavior (DSC, TGA), and the mechanical properties, namely, tensile strength, modulus of elasticity, elongation at break (%), and hardness. The morphological behavior (SEM) and dielectrical properties such as electrical conductivity, dielectric constant (ε′), dielectric loss (ε″), and loss tangent (tan δ) were estimated.

Notes: Mechanical and dielectric properties of rubber mixtures of epoxy/polyurethane based on castor oil loaded with glass fiber is of great interest from a technological point of view. Epoxy resin was prepared from bisphenol A and epichlorohydrin in alkaline medium. Polyurethane was prepared from the hydroxyl group of castor oil reacted with 2,4-toluene diisocyanate. Elemental analysis and infrared (IR) spectra are presented as evidence for the formation of these materials. A homogeneous solution of epoxy resin and polyurethane was prepared in acetone containing a curing agent 1,3-propane diamine. The glass-reinforced laminates were prepared by interacting polymer blends on the glass fiber through compression moulding. Chemical resistance to solvents, flexural strength, izod impact strength, and Rockwell hardness studies on a series of specimens were investigated. It was observed that they behave as tough materials. An attempt was made to study the effect of electrical conductivity by changing the compositions of rubber mixtures during reinforcement with glass fibers. The dielectric constant (E′) dielectric loss (E″), and loss tangent (tan δ) of the specimens and their dependence of temperature was studied.

Notes: Liquid prepolyurethanes were synthesized from castor oil and toluene-2, 4-diisocyanate (TDI) under different experimental conditions and varying NCO/OH ratios. All these prepolyurethanes were subsequently reacted with ethyl acrylate/ethylene glycol dimethacrylate mixtures by radical polymerization using benzoyl peroxide as initiator to obtain interpenetrating polymer networks (IPNs) by transfer molding. The novel polyurethane/poly(ethyl acrylate) IPNs are found to be tough films. These IPNs are characterized in terms of their resistance to chemical reagents, thermal behavior (DSC, TGA), mechanical behavior including tensile strength, Young's modulus, and elongation. The dielectric properties, namely electrical conductivity (σ), dielectric constant (ε′), dielectric loss (ε″), and loss tangent (tan δ) were computed. The mechanothermal behavior was analyzed by dynamic mechanical spectroscopy. The morphological behavior was studied by scanning electron microscopy.
