    Notes: Spores of a thymine-requiring strain of Bacillus subtilis 168, which is also temperature sensitive for the initiation of chromosome replication, were germinated and allowed to grow out at the permissive temperature in a minimal medium containing no added thymine. Under these conditions, there was no or very limited progression into the elongation phase of the first round of replication. In a significant proportion of the outgrown cells, a Z ring formed precisely at mid-cell and over the centrally positioned nucleoid, leading eventually to the formation of a mature division septum. When initiation of the first round of replication was blocked through a temperature shift and with thymine present, the Z ring was positioned acentrally. The central Z ring that formed in the absence of thymine was blocked by the presence of a DNA polymerase III inhibitor. It is concluded that the very early stages of a round of replication (initiation plus possibly limited progression into the elongation phase) play a key role in the precise positioning of the Z ring at mid-cell and between replicating daughter chromosomes.

    Notes: Summary Periodate oxidation of purified type 5 Adenovirus (Ad5) led to a mean loss of infectivity of 6.84 logs. There were no significant differences in adsorption and penetration between oxidized and mock-oxidized virus. However, after infection with oxidized virus, no synthesis of viral structural proteins could be detected and a 78.5% inhibition of viral DNA synthesis was observed. Labelling experiments performed by treating oxidized and mock-oxidized virus with tritiated sodium borohydride revealed that the fiber glycoprotein was one of the proteins labelled in oxidized virus whereas no labelled proteins were detected in non oxidized virus. In addition, it was found that one mol of formaldehyde generated during oxidation of sugar residues was bound per 500 base pairs in oxidized virus. One consequence of this in situ generation of formaldehyde is the formation of DNA-protein crosslinks. The DNA so crosslinked showed different patterns of restriction fragments with endonucleases such as Hpa I, Hind III and Kpn I but not with Xho I.

    Notes: Abstract The sediments of Loe Pool, a eutrophic coastal lake in south west England, consist largely of laminated clays and clay-gyttjas. Studies of the diatom microstratigraphy of frozen sediment cores from the Pool indicate that the laminations are annual, and that they contain pairs of light and dark bands formed by seasonal variations in the supply of sediment to the Pool from its catchment. Analysis of the magnetic properties of individual laminations demonstrates the presence of physical and mineralogical microstratigraphic variations, which may also be related to seasonality. A varve chronology, which is confirmed by 137Cs analysis and historical records, has been used to provide a timescale for the interpretation of data from other paleolimnological studies. A close agreement between variations in the abundance of sedimentary Sn, and the history of mining in the catchment, has been found. Similarly, analysis of total organic matter, total phosphorus, sedimentary chlorophyll a, sterols, diatoms and Cladocera in the uppermost sediments all indicate eutrophication of the Pool in the period AD 1940 to the present.

    Notes: Abstract There is currently much interest in mapping critical loads for nitrogen deposition as part of a strategy for controlling nitrogen emissions. While nitrogen deposition may cause acidification and excess nutrient effects, the former were considered previously in studies of sulphur deposition. In the UK, work on developing nutrient nitrogen critical loads maps has used several methods and databases. Two approaches are described here, one a steady state calculation using a nitrogen saturation limit for soil systems, the other an empirical estimate of critical loads set to prevent changes to vegetation communities. The empirical method uses national species records and land cover data derived from satellite imagery. Maps drawn from the available data are dependent upon a number of factors which reflect the approach used. To apply the nutrient critical loads to a strategy for future abatement measures, the nutrient nitrogen values for soils have been incorporated within a “critical loads function” which takes into account both acidity and nutrient effects as related to deposition loads for sulphur and nitrogen. This function may be used with deposition data to identify the need for sulphur and nitrogen emission reductions.
