    Notes: Abstract: A number of 5-hydroxytryptamine (5-HT) uptake inhibitors have been shown to displace the binding of [3H)imipramine to rat cortical membranes in a complex manner with Hill slopes less than unity. Norzimeldine displaced the binding of [3H]imipramine in a biphasic manner with IC50 values for the two components of about 30 nM and 30 μM. This latter site alone was found in tissues that had been treated with a protease. Binding to both of these sites was displaced by 10μM desipramine. The protease-sensitive [3H]imipramine binding sites were found to be saturable, high-affinity binding sites with a KD of 8 nM. The number of these sites varied between brain regions and was positively correlated with the regional distribution of [14C]5-HT but not [3H]noradrenaline uptake. This was not the case however for the protease resistant but desipramine-displaceable binding sites. Since most previous [3H]imipramine binding studies have been performed with high concentrations of desipramine (10 μ. M) to define “specific binding,” these data would suggest that either protease-sensitivity or displaceability by 1 μM norzimeldine would give more reliable estimates of the specific binding.

    Notes: Abstract: Human caudate nucleus, putamen, substantia nigra, and nucleus accumbens were analyzed for the effects of age on dopaminergic binding sites. Decreases in the number of dopaminergic binding sites were detected with age in caudate nucleus (44 specimens from three sample groups) and substantia nigra (n = 12). In caudate nucleus, the decline in [3H]2-amino-6,7-dehydroxy-1,2,3,4-tetrahydronaphthalene sites was three times greater than for [3H]spiperone, but age changes were significant in only two of the three sampling groups. No age changes in binding were detected in the putamen (n = 44) or nucleus accumbens. Age, sex, and tissue source all significantly contributed to variance. However, cause of death, time from death to tissue freezing, and length of storage did not influence dopaminergic binding in the caudate nucleus or putamen. Relative to the life-span, the age-correlated decrease in dopaminergic binding sites of human brain approximates that in aging rodent striatum. Comparisons of altered dopaminergic binding with other age-correlated changes suggest that neuronal loss may not be involved in the loss of binding sites before midlife.

    Notes: Glycogen synthase kinase-3β (GSK-3β) is implicated in regulating apoptosis and tau protein hyperphosphorylation in Alzheimer's disease (AD). We investigated the effects of two key AD molecules, namely apoE (E3 and E4 isoforms) and β-amyloid (Aβ) 1–42 on GSK-3β and its major upstream regulators, intracellular calcium and protein kinases C and B (PKC and PKB) in human SH-SY5Y neuroblastoma cells. ApoE3 induced a mild, transient, Ca2+-independent and early activation of GSK-3β. ApoE4 effects were biphasic, with an early strong GSK-3β activation that was partially dependent on extracellular Ca2+, followed by a GSK-3β inactivation. ApoE4 also activated PKC-α and PKB possibly giving the subsequent GSK-3β inhibition. Aβ(1–42) effects were also biphasic with a strong activation dependent partially on extracellular Ca2+ followed by an inactivation. Aβ(1–42) induced an early and potent activation of PKC-α and a late decrease of PKB activity. ApoE4 and Aβ(1–42) were more toxic than apoE3 as shown by MTT reduction assays and generation of activated caspase-3. ApoE4 and Aβ(1–42)-induced early activation of GSK-3β could lead to apoptosis and tau hyperphosphorylation. A late inhibition of GSK-3β through activation of upstream kinases likely compensates the effects of apoE4 and Aβ(1–42) on GSK-3β, the unbalanced regulation of which may contribute to AD pathology.

    Notes: —Monoamine oxidase (MAO) activity in the brains of 15 suicides, of whom 8 were alcoholics, was compared to a control material of 20 individuals without known mental disorder. At autopsy 13 different parts of the brain were macroscopicaily dissected out and the MAO activity in the samples estimated with β-phenylethylamine and tryptamine as substrates.The MAO activity in all parts of the brain investigated was found to be significantly lower in the alcoholic suicides as compared to the controls, while there was no significant difference between the non-alcoholic suicides and controls. Different variables which might have influenced the MAO activity were investigated. There was no significant correlation between age and tryptamine-oxidizing activity, but a positive correlation between age and β-phenylethylamine-oxidizing activity was found. There was also a significant difference between the series in the time lapse between death and autopsy and in the time during which the dead body was kept at room temperature. However, neither of these variables could explain the differences between the series. The results thus demonstrate a connection between low MAO activity in the brain and suicidal behaviour among alcoholics.

    Notes: Data from the total urban population 75 years and older in Kungsholmen, Stockholm were used to calculate the distribution of home help services. A structured questionnaire was used to collect information about education, occupation, living conditions, Katz Activities of Daily Living Scale, contact with the family, medical treatment and also questions about help needed and functional ability. Home help was considered to be provided when the subjects reported receiving home help service from the municipality. The number of hours of home help and the services received were registered. The results show that 86% of the elderly population in the Municipality of Kungsholmen live in their own homes, even when they are very old. Twenty-seven percent of those over 74 years and 33% of those over 80 years received home help services. The people receiving help were mostly women, single living, older and with disability detected by the Katz Activities of Daily Living Scale. The predicting factors of hours of home help are older age, condition of single living, disability detected by the Katz Activities of Daily Living Scale and less years of education. Our results also show that, among those who receive home help service, those with disability in the activities of daily living get more hours of help. However, a considerable group of elderly people are disabled but do not receive any home help. Our data stress the need for more attention by health care planners of domiciliary and intensive care and residential facilities for the high risk population (very old, living alone and with disability).

    Notes: Objectives:To study differences in nutritional, dental status and oral function between institutionalised patients with Alzheimer's disease and cognitively healthy elderly people living in the community. Design:Comparison was made between two groups, Alzheimer's disease sufferers and healthy controls, using established criteria for anthropometric, mental and dental state. Setting:An institution and residential area in Stockholm, Sweden. Subjects:Forty patients with Alzheimer's disease living in a nursing home and 40 age-and gender-matched control subjects living independently. Intervention:Dental status and anthropometric variables. Results:Overnutrition was less frequent among the demented than the controls and more demented were undernourished. Dental status was similar in the two groups with few edentulous subjects but only 2 of 7 edentulous subjects with Alzheimer's disease wore dentures. Having natural teeth and many functional oral zones is important for food consistency choice, but not for nutritional status. In the Alzheimer group, the stage of dementia has a strong association to the ability to eat unaided and an association with dental status. Conclusion:There are differences in nutritional status between Alzheimer's patients in institutions and cognitively healthy elderly living at home. The choice of food consistency is correlated to dental status but nutritional status is not shown to be influenced by dental status. However, the ability to eat unaided is strongly correlated to cognitive status.
