    Notes: Electron microscope autoradiography was used to study cartilage from regenerating limbs of adult newts, Triturus, after intraperitoneal injections of proline-3H. The labeling in the endoplasmic reticulum, small vesicles, Golgi vacuoles, ground cytoplasm and extracellular matrix was compared during the secretion of radioactive products. The data appear to indicate that a large part of the radioactive secretion probably leaves the cell after having been in only one cellular compartment. Although this compartment may be the endoplasmic reticulum, a considerable amount of radioactivity fluxes through the ground cytoplasm and the possibility cannot be excluded that some secretory components leave the cell directly from the ground cytoplasm. The data appear incompatible with the hypothesis that all the radioactivity seen in the extracellular matrix arrived there via a single pathway involving first the endoplasmic reticulum and then the Golgi vacuoles. It is not, however, incompatible with a hypothesis that a fraction of the radioactive product uses this pathway.

    Notes: The possibility of sympathetic innervation to melanophores in fish was investigated using a catecholamine histochemical fluorescence method. Catecholamine-containing fibers were observed to be in close anatomic proximity to the conjunctival and dermal melanophores of the cunner (Tautogolabrus adspersus). It is suggested that the catecholamines released from these adrenergic fibers are capable of causing aggregation of pigment with blanching of skin color.

    Notes: Using microradioautography, the origin of the gastric parietal (and zymogen) cells was deduced by observing the time of appearance of labelled parietal cells in relation to the other labelled cells in the gastric mucosa. In order to see whether the parietal cell divides in the adult mouse, time grain count curves of the labelled parietal cells were made from animals which were killed at 1-291 days after thymidine 3H injection DPT). Parietal cell survival was followed by observing the disappearance of the labelled parietal cell population. Parietal cells appear to be entirely derived from other cells since a significant number of labelled parietal cells does not appear unless the animal is allowed to survive for several days. Parietal cells do not appear unless the animal is allowed to survive for several days. Parietal cells do not seem to undergo mitosis in the adult mouse because the time grain count curves did not shift with time. Although a few labelled parietal cells persisted in the 291 DPT animals, a large majority of the labelled parietal cells disappeared by 90 DPT and had a half life of roughly 23 days. This suggests an element of renewal for the parietal cell population.

    Notes: Using horse radish peroxidase as a tracer for fluid flow, rapid diffusion was demonstrated between sinusoidal lumina and the interhepatocytic spaces of the murine liver. The marker passed rapidly between adjacent sinusoidal cells and filled the space of Disse. Appreciable amounts of peroxidase were also seen in pinocytic and phagocytic vacuoles within sinusoidal cells. Many junctions in the wall of bile canaliculi impeded the passage of the marker, but in some sites this hindrance did not occur. This provides a path for the direct exchange of material between vascular and biliary contents. Such a mechanism may be physiologically controlled. Junctions in bile ductules prevent diffusion of horse radish peroxidase.

    Notes: Foreign plasma injection induces a profound and somewhat complex change in the size and location of the colony forming unit (CFU) cell compartment. Injection of foreign plasma before irradiation induces an increase in CFU cells as judged by endogenous colonies as well as by a modification of the endogenous method which excludes spleen colony formation from in situ spleen cells. However, the enlargement does not take place in the most populous CFU cell areas, the spleen and marrow. The concentration and/or total number of CFU cells in spleen and marrow was not increased by plasma injection whether judged by the number of transplantable cells or by the number of migrating endogenous cells.These studies emphasize the complexity of this cellular system and suggest that the use of but one type of stem cell assay may yield results which do not reflect changes within the total compartment. Evidence for cell damage in vitro as a factor influencing results in studies involving transplantation was searched for but was not forthcoming.

    Notes: Because monocytes and their precursors cannot be recognized with certainty in tissues, an approach to the study of monocyte kinetics was made through examination of the peripheral blood. Injection of a single pulse of tritiated thymidine into rats resulted in the appearance of labeled monocytes identified as circulating peroxidase-positive mononuclear cells. The increase in the percent of labeled cells and in the mean grain count per cell followed a course described by a mathematical model with a generation time of 21 hours and a DNA synthesis time of 12.5 hours. The generation and synthesis times appear to be very uniform for the monocyte so that the phasing of cells represented by the uptake of label could be followed for more than two generations, a property not shared by neutrophils or lymphocytes. Monocytes appear in the circulation within eight hours of DNA synthesis.

    Notes: An electron microscope examination of the minute bristle organules in the Drosophila eye revealed an organisation characteristic of insect hair sensilla. They were derived from four concentrically arranged cells which were active at the mid-pupal stage in producing the bristle, socket and receptor structures. Two of the cells degenerated towards the end of pupation, the mature organule consisting of a bipolar sense cell and an accessory cell.Growth in length of the bristle was accompanied by a proliferation of longitudinally oriented microtubules which gradually disappeared after maximum growth had been achieved. The breakdown of microfibrillar aggregates, which were also present as transient structures in the developing bristle and showed some correspondence with the longitudinal ridges formed at the surface, may similarly be related to the establishment of cell shape by the deposition of the cuticle which occurred at the same time.

    Notes: In 85 frogs and 29 rats, the entire gastrocnemius muscle was removed. After removal of as much connective tissue as possible, about two-thirds of the remaining muscle was finely minced with a scissors. These minced fragments were orthotopically re-implanted, and the overlying skin was sutured. As the implanted muscle fragments degenerate, new muscle fibers appears in the regenerate. The proportion of connective tissue to muscle is usually greater than normal especially in the frog. Grossly, normal relationships are established with the cut ends of the Achilles and proximal tendons, as well as with the blood vessels and nerves. The total diameter of the regenerated muscle is almost always less than half of that seen in normal muscles. Regeneration of muscle is much more extensive in the rat than in the frog, and it occurs almost twice as rapidly. The histology of the regenerative process is described for both the frog and the rat.

    Notes: Electron microscope observations on the differentiating Drosophila eye show an extensive proliferation of parallel arrays of microtubules at periods preceding, or coinciding with, alterations in cellular morphology. In the retinular cells they are aligned in the direction of elongation and close to the developing rhabdomeres, forming a cylinder around the central ommatidial axis. At a later stage, in the cone cells, they are aligned in the direction of cellular contraction. Thus as in other developing systems microtubules appear to be directly involved in the morphogenesis of the Drosophila eye. In the retinular cells they gradually disappear during elongation, whereas they persist in the cone cells. The pigment cells contain few of these structures. The distribution of two types of specialised cell attachments, adhering zones and septate desmosomes is discussed in relation to intercellular morphogenesis and communication. The rhabdomeres originate from infoldings of the plasma membrane which later grow out into typical microvilli. Unusul cytoplasmic granules are described in the pigment cells of early pupae.

    Notes: Rats were given single doses of an orally-active estrogen by mouth on days 1-5 of pregnancy. Examination of uteri on day 8 for implantations revealed that the median effective doses for preventing 50% of the eggs shed from implanting were 0.047, 0.050, 0.035, 0.145 and 0.380 mg/kg for days 1, 2, 3, 4 and 5 respectively. It was found that failure of pregnancy after treatment with the minimal 100% effective dose (ED100) on days 1, 2 or 3 was due to expulsion of eggs from the reproductive tract by day 5. Treatment of rats with the ED100 on day 4 caused expulsion of some eggs from the tract by day 5, but also had effects on the uterus: the position of the blastocysts within the lumen was abnormal and their orientation relative to the mesometrial-anti-mesometrial axis disturbed. No decidual reaction was observed on day 6 in such rats or on day 9 in pseudopregnant rats treated similarly and subjected to uterine traumatization on day 5. Treatment with ED100 on day 5 had no effect on egg transport or on the positioning of blastocysts within the lumen on day 6. A decidual reaction was seen around the blastocyst on day 6, but in pseudopregnant rats treated similarly and subjected to uterine traumatization on day 5 no decidual reaction was found on day 9. The pontamine blue reaction was negative in nearly all rats dosed on day 4 and examined on days 6-8. In rats dosed on day 5, however, the blue reaction was negative on day 6, but positive on day 7, i.e., delayed by 24 hours compared to controls and appearing later than the decidual reaction. The blastocysts were unattached in these rats on day 6, but may have attached briefly on day 7 (suggested by the positive blue reaction) and subsequently failed to develop further owing to disappearance of the decidual tissue.
