    Notizen: Abstract: Protein tyrosine kinases that are known to have major roles in the control of cell growth and transformation are abundant and have numerous phosphoprotein substrates in the adult CNS. Although less well characterized than serine/threonine kinases, tyrosine kinases are also concentrated in the synapse. The effect of genistein, a selective inhibitor of tyrosine kinase activity, was examined on the in vitro release of endogenous dopamine (DA) from superfused mouse striatal slices. Fractional release of DA was significantly increased over basal release levels by genistein (100 and 200 µM). The effect was concentration dependent and rapidly reversible on washout of the kinase inhibitor. No significant change from basal release levels was observed with two structural analogues of genistein that do not inhibit tyrosine kinase activity at the same concentration. We have previously described alterations in basal and evoked DA release from the striatum of the weaver (wv/wv) mutant mouse, and genotypic differences in fractional release were also observed with genistein stimulation. The total evoked release was 25–50% greater from the wv/wv striatum. These results suggest a modulatory role for tyrosine kinase activity in neurotransmitter release and perhaps an alteration of kinase-regulated mechanisms in the DA-deficient wv/wv striatum.

    Notizen: Abstract: The weaver mutant mouse (wv/wv) has an ∼70% loss of nigrostriatal dopamine (DA) neurons, but the fractional DA release evoked by amphetamine (but not a high potassium level) has been shown to be greater from striatal slices of the weaver compared with +/+ mice. In the present work we tested the hypothesis that fractional DA release from weaver striatum would be greater when release was mediated by the DA transporter. Serotonin (5-HT)-stimulated fractional DA release was greater from weaver than from +/+ striatum. The release evoked by 5-HT in the presence of 10 µM nomifensine (an antagonist of the DA transporter) was less than in its absence, but the difference between weaver and +/+ striatum remained. In the presence of nomifensine, 1-(m-chlorophenyl)biguanide, classified as a 5-HT3 agonist, also induced a greater fractional release from weaver compared with +/+ striatum. When veratridine was used at a low concentration (1 µM), the fractional evoked release of DA was higher from the weaver in the presence and absence of nomifensine. These findings suggest that the reason for the difference in the responsiveness of the two genotypes to these release-inducing agents is not related to DA transporter function.

    Notizen: Abstract Cesium atoms, enclosed in a glass vessel, are excited by a weak electrical discharge and irradiated by monochromatic light with wavelengths corresponding to the sharp, diffuse, and fundamental series. By light absorption, highly excited atoms are produced, which are ionized by electron collisions. These ions diffuse into the space charge region of a diode path, compensate partially the space charge, and increase the thermionic diode current. By this method, ionic currents of about 10 ions per sec can be detected. If the wavelength of the incident light is varied continuously, the complete absorption spectrum of excited cesium atoms can be found in the recorded diode current.

    Notizen: Abstract A case of aggressive plasma cell leukemia with unusual morphological and cytogenetic features is reported. A 65-year-old man was admitted to hospital due to anemia, thrombocytopenia, and renal insufficiency. Bone marrow examination and peripheral blood smear revealed a large number of pleomorphic cells with convoluted and multilobulated nuclei. Immunohistochemistry of the bone marrow biopsy was negative for anti-keratin antibodies CAM.5.2 and AE1/ AE3, but positive for EMA. The immunophenotypic features of these cells were suggestive of plasma cell orgin with positivity for CD38, CD56, CD9, and CD44 and a weak positivity for CD71 and CD45 (40% of the cells), while all other markers of hematopoeitic origin were negative. Furthermore, a serum protein electrophoresis showed a monoclonal component type IgG-kappa of 70 g/l. The cytogenetic analysis demonstrated a hypotetraploid clone with multiple numerical and structural abnormalities. Although some of the aberrations found are associated with plasma cell malignancies — e.g., structural rearrangement of chromosome 1, del(6q), and monosmy 13—the karyotypic complexity in the present case is unusual. The course of the disease was very aggressive, and the patient died 3 days after admission.

    Notizen: Abstract We present exact diagonalisation data for the ground state of finite two-dimensional spin 1/2 Heisenberg antiferromagnets with structural disorder. The combination of antiferromagnetic coupling and structural disorder creates frustation and a local singlet formation which may strongly influence the magnetic ordering. In detail we study the magnetic radial distribution function, the magnetic order parameter M2 as well as a parameter measuring the spin misfit and present these quantities in dependence on the strength of the structural disorder. We estimate the critical disorder where the antiferromagnetic long-range order breaks down.

    Notizen: Abstract Field studies were conducted to assess the turnover and the leaching of nitrogen in arable soils of Lower Saxony (NW Germany). The mean surplus N (difference between N inputs by fertilization and N export by the yield; 146 field plots) from 1985–1988 amounted to 38 kg ha-1 yr-1 in fine textured (clay, loam, silt) and to 98 kg ha-1 yr-1 in coarse (sandy) soils. Leaching of nitrate calculated by a simple functional model for simulation of the N regime over the winter period (i.e. mineralization and leaching) was 16 kg ha-1 in the fine and 63 kg N ha-1 in coarse soils (mean values of the winter periods 1985–1988 from 256 plots). Before the 1960s, the depth of the Ap horizons rarely exceeded 25 cm in arable soils of the former FRG. During the last three decades, ploughing depth has increased to at least 35 cm. The mass balance calculations for total N after ploughing to 35 cm in loess soils of southern Lower Saxony (105 farm plots) yielded a mean increase in total N by about 900 kg ha-1 in 20 years. With respect to soil organic matter equilibria, N accumulation will continue for at least another 10 years on 67% of the examined farm plots. This study suggests that long term N immobilization is one of the most important sinks for nitrogen in arable soils of Germany. For simulation of the N dynamics over the growing season and for long time periods total nitrogen dynamics need to be considered.

    Notizen: Abstract Tyrosine hydroxylase activity was assayed in microdissected substantia nigra and striata from seven strains of mice (BALB, CBA, YBR, WB, IS, MOLG, and CAST). In the substantia nigra where tyrosine hydroxylase activity is thought to be proportional to dopaminergic neuron number, only CBA had a different (lower) enzyme activity compared with BALB. However in the striatum, tyrosine hydroxylase activity was larger for IS, MOLG and CAST compared with BALB. Further investigation of the CAST striatum showed that dopamine content and dopamine uptake activity were also higher in comparison with BALB. All three dopaminergic parameters were larger because of lower protein levels in the CAST striatum. A lower absolute amount of glutamic acid decarboxylase activity in CAST versus BALB striatum was consistent with the possibility of a smaller CAST striatum. In contrast to dopamine, the serotonin content in CAST striatum was reduced in proportion to the decrease in protein content. We suggest that the CAST striatum is smaller than BALB striatum and is innervated by proportionally fewer serotoninergic terminals, but the amount of dopaminergic innervation of the CAST striatum is not altered by the size of the target.
