Zusammenfassung Die Mikrodiskelektrophorese mit Polyacrylamidgelen wird als neue Methode zum Nachweis von Desoxyribonucleasen im Schweiß beschrieben. Unterschiede der Nuclease-Aktivität im Schweiß und Urin gesunder und an Mucoviscidose erkrankter Kinder ergaben sich nicht. Die höchste Aktivität im Schweiß findet sich nach saurer Inkubation (pH 5,0) unter Zusatz von EDTA. Nach Inkubation in schwach alkalischem Milieu (pH 7,4) und Ionenzusatz kann nur höchstens 1 enzymaktive Bande nachgewiesen werden. Im Urin lassen sich durch saure Inkubation (pH 5,0) und Magnesium- und Calciumionenzusatz sowohl bei Patienten als auch bei Kontrollpersonen 4 Nuclease-Banden deutlich darstellen. Die Ergebnisse werden im Zusammenhang mit früheren Untersuchungen der DNase-Aktivität in Organen Mucoviscidosekranker und histochemischen Befunden der gesunden Haut diskutiert.

Summary Micro disc-electrophoresis (polyacrylamide gela) is introduced as a new method for evaluation of DNases in the sweat. Activity of sweat and urine DNases was not different in healthy children and in patients suffering from mucoviscidosis. DNase activity in the sweat reaches its peak after acid incubation (pH 5,0; EDTA supplement). Incubation in a slighty alkaline medium (pH 7,4), electrolytes added, produces not more than one band of enzymatic activity. In urinary material 4 DNase-bands are demonstrated by acid incubation (pH 5,0; with magnesium and calcium ion supply) in patients as well as in normal persons. The results are discussed referring to earlier findings concerning DNase activity in the organs of patients with mucoviscidosis, and to histochemical investigations of the normal skin.
