    Notes: Histopathological studies in a patient with cutaneous vasculitis revealed sub-epidermal micro-abscess formation as seen in dermatitis herpetiformis (DH). Direct immunonfluorescence of perilesional skin revealed granular IgA fluorescence at the dermo-epidermal junction and deposition of IgA and C3 within vessel walls, whereas immunofluorescence studies of uninvolved skin revealed IgA and C4 deposition within upper dermal vessels only. IgA containing complexes were detected in the serum, and evidence of classical pathway activation was provided by demonstrating CIq and C4 within vessel walls with absence of properdin. Prior to referral there had been a favourable therapeutic response to dapsone. This case illustrates that some forms of leucocytoclastic vasculitis may closely resemble DH and that direct immunofiuorescence of uninvolved skin is required to differentiate the two conditions.

    Notes: The two cases reported in this paper allow the differentiation pagetoid reticulosis from solitary lesions of mycosis on the basis of clinical, histological ultrustructural immunohistochemical and enzyme histochemical studies. The first patient presented with a slowly growing asymptomatic scaly plaque of 30 years duration Histology of the lesion was typical of pagetoid reticulosis. Immunohistochemical studies showed that the abnormal cells present in the cutaneous infiltrate were not T lymphocytes and ultrastructural studies suggested these were of histocytic origin. The second patient presented with a poikilodermatous lesion present for 14 years with the histology of plaque stage mycosis fungoides. Enzyme and immunohistochemistry showed the predominant T cell nature of the cutaneous infiltrate and electron microscopy showed numerous mycosis fungoides cells. In contrast to generalized forms of mycosis fungoides, no peripheral blood monocyte chemotactic defect was found in either case. Both lesions responded to radiotherapy.

    Notes: Abstract. Aspects of the response of Kallar grass (Diplachne fusca) to root salinity have been studied. Field observation on the high level of tolerance of this species have been mirrored in pot trials and a 50% yield (dry matter) was obtained at ECe 22.3. While salt stress led to Na and Cl uptake, most of these ions appeared to be secreted selectively from the leaves. The shoot K+ content on a tissue water basis remained unaffected by salt stress and the shoot tissue had a high K+ selectivity. Osmotic adaptation was mainly brought about by tissue dehydration and not net salt accumulation although the compatible solute glycinebetaine was accumulated in fairly high concentrations.The organic and inorganic chemical composition of the shoots, including the trace elements Zn, Fe, Cu and Mn, is such as to make this species a highly promising plant for the exploitation of saline soils which might otherwise not be amenable to agriculture use.

    Notes: Abstract: The dopamine (DA)-containing nerve terminals in the caudate nucleus arise from cell bodies located in the substantia nigra (pars compacta), and it is possible that p-tyramine- and m-tyramine-containing neurons may also exist in this nucleus. We have studied the effects of unilateral electrolytic lesions of the pars compacta in rat on levels of DA, p-tyramine, m-tyramine, and homovanillic acid in the caudate nucleus after various survival times. At 12 and 24 h following lesioning the ipsilateral level of p-tyramine was significantly reduced compared with the contralateral side, whereas the concentrations of m-tyramine, DA, and homovanillic acid were significantly increased. Thus, in the short term, the lesion results in an increase in DA turnover, which is accompanied by an increase in m-tyramine levels and a decrease in p-tyramine levels. Similar changes occur following pharmacological treatments (chlorpromazine, d-amphetamine, l-DOPA) that increase DA turnover. At survival times of 2, 11, and 25 days, the ipsilateral concentrations of m-tyramine, DA, and homovanillic acid were reduced along with p-tyramine. These longer-term alterations in amine levels are most likely a consequence of degeneration of nigro-striatal axons. Placement of a lesion 1 mm dorsal to the usual position centering on the pars compacta produced different biochemical changes from those seen after the pars compacta lesion. One day following this lesion the concentration of p-tyramine was slightly reduced; DA was unaffected, but the concentration of m-tyramine was profoundly increased, even more so than after the pars compacta lesion. This could indicate the existence of specific m-tyramine-containing cell bodies located dorsal to the substantia nigra. The results suggest that p- and m-tyramine in the caudate nucleus originate from neurons in or close to the substantia nigra. The results in the short term following the lesion support the observation that there is an inverse relationship between p-tyramine concentration and DA turnover in the caudate nucleus.

    Notes: Since the mid 1950s, there have been many developments in serological techniques for testing for plant viruses. Certain of these can provide great savings in time, labour and cost in routine testing situations and are well suited for use by advisory services, certification schemes and healthy stock programmes, statutory and quarantine authorities, and plant breeders. The recent developments are described under the headings flocculation in liquid media, gel diffusion, labelled antibodies and electron microscope serology, Flocculation tests depend on observation of aggregates formed in a liquid medium. The microprecipitin and chloropiast agglutination tests are simple forms which are still widely employed routinely. However, sensitivity is greatly improved by using flocculation tests in which antibodies are adsorbed to the surface of larger inert carrier particles, such as polystyrene latex spheres, tanned red blood cells or bentonite. Latex particles are the most widely used of these because they are easy to sensitise with antibodies, little antiserum is required and the sensitised latex can be stored for years without loss of activity. The latex test is also very simple to do, is well suited for routine checks on either small or large numbers of samples which can be grouped and can be used with many virus–crop combinations.Gel diffusion tests depend on observation of precipitin lines formed in an agar gel medium. They are of two main types, single (= simple) diffusion and double diffusion. Both can be used effectively with viruses which have isometric or ‘near isometric’ particles but not with those which have elongated particles because these do not diffuse readily through agar. Recent developments which involve breaking elongated particles into sub-units or fragments which can diffuse through agar now permit gel diffusion to be applied with almost all viruses. Various treatments are useful in breaking particles, the most effective of which employ detergents or other disruptive chemicals such as pyridine, pyrrolidine and ethanolamine, However, although gel diffusion tests which incorporate one or other of these treatments have been employed routinely for testing for viruses in several different crops, such tests are relatively insensitive, require much antiserum and conditions must be carefully controlled to avoid formation of nonspecific precipitin lines.Antibodies can be labelled to make virus—antibody aggregates readily observable, or to obtain increased sensitivity in testing, or both. The only form of labelled antibody test widely employed in routine screening for plant viruses is enzyme-linked immunosorbent assay (ELISA), in which antibody is labelled with an enzyme and positive results are observed as an enzyme-mediated colour reaction. Since its introduction to plant virology in 1976, ELISA has been very widely applied in situations where large numbers of either individual or grouped
