    Notes: Human blood lymphocyte subpopulaiions, revealed by a panel of commercially available monoclonal antibodies by means of a resetting technique, were submitted to direct cyto-chemicat analysis and were shown to have distinguishing characteristics. T cells reactive with OKT3 antibody (T3+) displayed higher beta-glucuronidase and dot-like alpha-napnthyl acetate acid esterase (ANAE) activity than T3− cells. Helper/inducer cells (T4+) were characterized by a high level of dot-like ANAE activity, whereas cytotoxic/suppressor cells (T8+) displayed selective naphthol-ASD-chloroacetate esterase activity. These results provide evidence for association of some cytoplasmic enzyme activities with the expression of membrane differentiation markers defined by monoclonal antibodies.

    Notes: Client acceptability of treatment procedures has recently become a significant concern in the assessment of psychotherapeutic effectiveness (Kazdin, French & Sherick, 1981; Wolf, 1978). Formally, acceptability refers to the fairness, appropriateness and intrusiveness of the treatment procedure as judged by clients, lay persons and nonprofessionals (Kazdin, 1980a). Unfortunately, marital and family therapists have yet to utilize this criterion in an evaluation of their psychotherapeutic interventions. Consequently, the purpose of the present investigation was to examine the acceptability of four differing therapeutic models (psychoanalytic, behavioral, systems and eclectic) used in the treatment of marital discord.

    Notes: Summary The cerebellar output function was studied using cerebellopetal proprioceptive stimulation hich produces simple and complex excitatory discharges as well as inhibitory activity in the Purkinje cells. The activity of the intracerebellar nuclei (and of the entire brain stem) was measured by the energy consumption as revealed with the [14C]-2-deoxyglucose method. The stimulations consisted of repetitive (1–20 c/s) electrical excitation of the nerve leading to the inferior oblique, to the masseteric and to the gastrocnemius soleus muscle. Compared to a group of non-stimulated controls, heavy bilateral labeling was obtained in the posterior pole of the fastigial nucleus. This was not observed with stimulation of the vibrissal pad which, however, produced a clear increase of [14C]-2-deoxy glucose uptake in the secondary trigeminal complex. Labeling of the posterior part of the fastigial nuclei was suppressed by ablation or pharmacologic inactivation of the overlying cerebellar cortex which suppresses the inhibitory activity of the Purkinje cells into the nuclear cells. Labeling of the posterior fastigial nuclei was also decreased in animals not stimulated but with ablation or pharmacologic inactivation of the overlying cerebellar ortex. The hypothesis proposed is that the marking results are the consequence of an increased activity in the Purkinje cell terminals. The activity of the Purkinje cells was also recorded extracellulary both before and during repetitive stimulation of a muscle nerve. The discharge activity of those in the cerebellar vermis and giving axons to the posterior fastigial nucleus was increased by the stimulation, whereas the activity of those of the hemispheral parts remained unchanged. Units in the fastigial nucleus were also recorded. Their activity was found to be deeply depressed so that only a few units were encountered and no further decrease of their discharge could be detected with the stimulation of a muscle nerve. Nevertheless, using the present data and those previously obtained, the conclusion is advanced that the cerebellar output function is actually decreased during afferent cerebellar stimulation.

    Notes: Summary Plasminogen activators are membrane-associated, arginine-specific serine proteases which convert the inactive plasma zymogen plasminogen to plasmin, an active, broad-spectrum serine protease. Plasmin, the major fibrinolytic enzyme in blood, also participates in a number of physiologic functions involving protein processing and tissue remodelling, and may play an important role in tumor invasion and metastasis. In HTC rat hepatoma cells in. tissue culture, glucocorticoids rapidly decrease plasminogen activator (PA) activity. We have shown that this decrease is mediated by induction of a soluble inhibitor of PA activity rather than modulation of the amount of PA. The hormonally-induced inhibitor is a cellular product which specifically inhibits PA but not plasmin. We have isolated variant lines of HTC cells which are selectively resistant to the glucocorticoid inhibition of PA but retain other glucocorticoid responses. These variants lack the hormonally-induced inhibitor; PA from these variants is fully sensitive to inhibition by inhibitor from steroid-treated wild-type cells. Cyclic nucleotides dramatically stimulate PA activity in HTC cells in a time- and concentration-dependent manner. Paradoxically, glucocorticoids further enhance this stimulation. Thus glucocorticoids exert two separate and opposite effects on PA activity. The availability of glucocorticoid-resistant variant cell lines, together with the unique regulatory interactions of steroids and cyclic nucleotides, make HTC cells a useful experimental system in which to study the multihormonal regulation of plasminogen activator.

    Notes: Summary The cereal nerve-giant interneurone synapse of the cockroach,Periplaneta americana, has been investigated using the ‘single axon, oil-gap’ method, in order to identify the sensory receptors whose activity resulted in a postsynaptic inhibitory response in the giant interneurones. Submaximal electrical stimulation of the cereal nerve X evokes a biphasic postsynaptic potential in the giant interneurones GI 1, GI 2 and GI 3, including an ipsp superimposed on a depolarizing psp. The sensory origin of the cereal nerve X has been studied using the cobalt back-filling method. Distally, nerve X is divided into two branches. The inner branch contains about 500 neurones which innervate small sensilla trichodea located on the ipsilateral half of the supra-anal plate and a few campaniform sensilla on the lateral extended portion of the same plate. The electrical stimulation of this branch causes a monosynaptic depolarizing psp. The outer branch contains about 70 neurones and is exclusively devoted to a chordotonal organ according to electron microscopical evidence. This is laterally located near the basal articulation of each cercus. The electrical stimulation of this branch induces an ipsp. This inhibitory chordotonal organ can be activated by the lateral movement of the whole cercus. These new data require a re-examination of the organization of the giant interneurone receptive fields for a better understanding of their participation in cockroach behavior.
