    Notes: Human monocyles release cytostatic protein factors (CF) upon activation with lymphokines und lipopolysacchairide. CF has been purified from monocyte supernatants by ion-exchange chromatography. chromatofocusing and gel filtration, and this resulted in a 10,000-fold reduction in the amount of protein in the purified CF preparation compared to the amount in the monocyte supernatant. About 5% of the eytostatic activity was recovered after purification. CF constitutes a population of proteins heterogeneous with respect to ionic charge and differing in their isoelectric points, since CF eluted as a broad peak both upon ion-exchange chromatography and chromatofocusing. The isoelectric points of the CF proteins were determined by chromatofocusing to be between 6.0 and 5.0. The molecular weight of CF as determined by gel filtration was in the range of 45,000 to 35,000 daltons. Only one monocyte-secreted protein with a molecular weight of 40,000 was detected upon sodium dodecyl sulphate-polyacrylamide gel electrophoresis of proteins in the purified CF preparations obtained after the final gel filtration step, und this protein may consequently be CF. If the 40,000-dalton protein is CF. one may estimate that about 106 monocytes produced roughly 0.1 μg CF upon activation and that significant cytostasis may be detected with less than nanogram quantities of CF.

    Notes: Struvite or magnesium ammonium phosphate MgNH4PO4 has been proposed asactive component in setting surgical cements. The usual formulation is one in which the magnesium component in the powder is either magnesium hydrogen phosphate trihydrate or trimagnesium phosphate or a mixture of these two compounds. As the cement liquid a concentrated solution of diammonium phosphate is taken. To make the cement attractive as a bone substitute material a calcium phosphate filler is generally incorporated. Thus such materials are a type of pseudo calcium phosphate cements. This study was intended to find out which calcium phosphate and which magnesium compound are the most suitable. In the first series of experiments a mixture of 12 g Mg3(PO4)2 and 4 g MgHPO4.3H2O was used as the magnesium component in the powder. To that powder 30 g of either precipitated hydroxyapatite PHA or CaHPO4 or CaHPO4.H2O or b-TCP or a-TCP was added. The cement liquid was a 3.5 M solution of (NH4)2HPO4. At specific liquid/powder ratios L/P suitable setting times were obtained for the different formulations. However, the compressivestrengths after immersion of the cements in 0.9% saline solution at 37°C varied over a large range. The best formulation was that with a-TCP which reached a compressive strength of 57 MPa after 18 h of immersion.In the second series of experiments 20 g of Mg3(PO4)2 was used as the magnesium component in the powder. Again 30 g of either of the above mentioned calcium phosphates was used as filler and again a 3.5 M solution of (NH4)2HPO4 was used as the cement liquid. At the appropriate L/P ratios the respective setting times were longer than in the first series of experiments but all five formulations appeared to result in good compressive strengths varying from 41 MPa for the formulation with b-TCP to 67 MPa for the formulation with PHA.In the third series of experiments 30 g a-TCP was taken as the calcium phosphate inthe powder. As magnesium components mixtures of Mg3(PO4)2.8H2O and MgHPO4.3H2O and Mg3(PO4)2 were used. Again the cement liquid consisted of a 3.5 M solution of (NH4)2HPO4. The formulations with Mg3(PO4)2.8H2O had the shortest setting times and the lowest compressive strengths, whereas those with Mg3(PO4)2 had the longest setting times and the highest compressive strengths. Therefore, it is advantageous to use Mg3(PO4)2 as themagnesium component

    Notes: Summary 4′-6-diamidino-phenylindole (DAPI) which is used for the detection of mycoplasmic infections of cell cultures or cytofluorometric investigations proved to be a sensitive agent for the fluorometric assay of alkali extracted DNA in cultured cells and human epidermis. Various experiments to optimize assay conditions led to the following procedure: 1 part of freeze dried tissue (1 mg/ml) and 1 part of 0.2 mol/l NaOH (w/v) were heated at 95° for 40 min. After cooling for 1 h to room temperature, 0.1 ml alkaline extract was mixed with 2.9 ml DAPI-solution (0.2 μg/ml in McIlvaine buffer containing 1 mmol/l MgCl2). After 10 min in the dark the emitted fluorescence was estimated at 453 nm with an excitation at 360 nm. This assay procedure allowed a detection limit of 25 ng DNA.

    Notes: Summary Ro 15-1788 is a specific benzodiazepine antagonist, which has also been shown to have some agonist properties. Since benzodiazepine receptors are involved in the physiological mechanisms of vision, a possible intrinsic effect of Ro 15-1788 was sought in 6 healthy volunteers by study of psychophysical flicker thresholds, including critical fusion frequency and low frequency modulation threshold, and pattern reversal visual evoked response, using double blind cross-over methodology. In each session 2 tablets of Ro 15-1788 30 mg were administered. Using a two factorial univariate analysis of variancé, no change was detected in any of the parameters studied.

    Notes: Summary The effects of gonadal steroids upon the discharge frequencies of the weakly electric fishApteronotus rostratus were examined. Androgens elicited frequency decreases, while estrogens had no effect. The androgen-induced decreases occurred with very short latencies and lasted only for several hours. The medullary nucleus that controls the discharge frequencies of these animals was subsequently isolated in an in vitro preparation. The effects of steroids were tested on the isolated pacemaker, and were found to be similar to those seen in intact animals. Thus, steroid effects upon discharge frequencies appear to be a consequence of direct effects upon the pacemaker. The short time course of the response suggests non-genomic effects of steroids upon neuronal activity in the pacemaker.

    Notes: Summary Previous studies indicated that gonadal steroids can induce changes in both motor and sensory aspects of the electrosensory system of weakly electric fish: androgens decrease the electric organ discharge frequencies and electroreceptor best frequencies of the South American gymnotoidSternopygus. The relationship between these two effects, however, was not known. In the present study, electric organ discharges (EODs) ofSternopygus dariensis were eliminated by means of spinal cord transections. This was done in order to allow an independent assessment of the influences of gonadal steroids upon electroreceptor tuning and those structures in the CNS responsible for establishing the discharge frequency. Transection alone affected neither the rhythmic discharges of the pacemaker nucleus that normally controls the discharge frequency, nor the best frequencies of electroreceptors. Similarly, administration of the androgen 5α-dihydrotestosterone (DHT) to transected animals also had no significant effect upon electroreceptor tuning. DHT did, however, cause significant decreases in the discharge rates of the pacemaker nucleus. Thus, the effects of gonadal steroids upon discharge frequencies in intact animals are a direct consequence of CNS influences, while effects upon electroreceptor tuning likely arise as a secondary consequence of the changed discharges of hormone-treated animals.
