    Notes: The regional distributions of thyrotrophin-releasing hormone (TRH) and substance P in postmortem human spinal cord were determined by radioimmunoassay in fresh tissue taken from 22 patients who died without known neurological disease. Dorsal, ventral, and intermediolateral spinal cord regions were obtained from different segmental levels (lumbar L1, 2, 3, and 4; thoracic groups T1–3, T4–6, T7–9, and T10–12) together with selective regions of grey matter of lumbar spinal cord. The effects on peptide levels of (a) the age of the patient, (b) the postmortem time interval, and (c) freezing the tissue samples prior to assay were assessed. Levels of 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) were determined in regional lumbar and thoracic tissue using HPLC with electrochemical detection. Substance P was found in the highest concentration in the dorsal spinal cord, with no significant segmental differences. In contrast, TRH was present in higher levels in the ventral rather than the dorsal spinal cord, with segmental differences. There was a significant difference in the 5-HT/5-HIAA ratio between dorsal and ventral spinal cord, with the highest ratio in the ventral spinal cord. There were no significant differences in substance P, TRH, or 5-HT levels in spinal cords between 5 and 20 h postmortem or from patients aged between 65 and 90 years. Freezing the tissue (– 80°C for 24 h) prior to assay significantly reduced TRH and substance P levels compared to samples assayed immediately without prior freezing. The levels and distribution of peptides and indoleamines in human spinal cord are compared with those previously reported in the rat and other species and the possible roles of these substances in the control of motor function and autonomic control are discussed.

    Notes: The presence of fluctuating dipoles in a polar liquid has been suggested to cause an intrinsic tail of localized states extending below the conduction band edge at −V0. If such preexisting states are the final states for photoinjection into such a liquid, then they cannot be described by the conventional treatment which assumes free-particle states. In this elementary model, the effect of both free and localized final states on the Fowler theory for photoemission is determined. Well above threshold Fowler's 1/2-power law is found. The near-threshold behavior is governed by the exponential tail in the final density of states. The results compare well with recent results on photoinjection into H2O and NH3. We find V0 to lie near −0.80 eV in NH3.

    Notes: 3H-fucose was injected intravenously or intravitreously into albino rats. After time intervals of 10, 40, and 50 min, 1, 1.5, and 4 hr, 1, 3, and 7 days, and 1, 2, and 4 weeks after injection, the animals were sacrificed by intracardiac perfusion with gluteraldehyde. Samples of the ciliary body were prepared for light and electron microscope radioautography. Light microscope autoradiographs showed that the cells of both the inner and outer layers of ciliary epithelium actively incorporated 3H-fucose label in a reaction that peaked in intensity at 4 hr after injection, and then progressively declined. Electron microscope radioautographs revealed that, at early time intervals, most of the label was localized to the Golgi apparatus. With time, the plasma membrane of both cell types became increasingly labeled, and accounted for 60-70% of the total silver grains at 4 hr after injection. Adjacent to the basal cell surface of the inner layer cells, the fibers of the zonula became increasingly labeled from 1.5 hr onwards, providing strong evidence that these cells secrete glycoproteins to the zonula. When vinblastine was administered 30 min before 3H-fucose injection, followed by sacrifice 1.5 hr later, a much larger proportion of label remained localized to the Golgi apparatus than in controls, and the plasma membrane and zonula were much less labeled. These results suggest that, as documented in other cell types, microtubules may play a role in the intracellular transport of membrane and secretory glycoproteins in these cells.
