    Notes: Using antibodies directed against the basement membrane (BM) components laminin and type IV collagen, basement membrane patterns were studied in borderline epithelial tumors of the ovary. To determine the potential use of BM immunohistochemistry in the histopathologic diagnosis of tumors of borderline category, BM patterns were compared with those in cystadenomas and cystadenocarcinomas. In cystadenomas, regular and intact BM were found at the interface between the epithelial cells of the cysts and the adjacent stroma. Cystadenocarcinomas displayed an irregular pattern, with areas of intact BM between tumor cells and stroma, as well as areas with irregular discontinuities. Tumors of borderline malignancy shared a continuous BM pattern with cystadenomas, However, 30% of the borderline malignant tumors contained small areas with BM interruptions resembling those of invasive carcinoma. From the results of this study we conclude that BM patterns in ovarian tumors of borderline malignancy are mostly similar to those of benign epithelial tumors. Focal defects possibly indicating early invasive growth, however, occur in 30% of the cases.

    Notes: The kinetics of rise and decay of ground-state atomic oxygen was investigated in a low-pressure O2 radio-frequency discharge. The ground state of O was monitored using laser-induced-fluorescence spectroscopy. The discharge was operated in two modes: a continuous mode and a pulsed mode. Typical discharge settings are rf power of 100 W, a pressure of 10 Pa, and a gas flow of 10 sccm. Using a pulsed mode the behavior of the O density was determined in a pure O2 discharge with no reaction products, and in a discharge during etching of organic polymers. Production and loss mechanisms for O in a pure O2 discharge are discussed. For an O2 discharge during etching of resist it is found that under the given experimental conditions atomic oxygen does not play a significant role in the chemical etch mechanisms involved. © 1995 American Institute of Physics.

    Notes: Summary Secondary somatic embryogenesis is the phenomenon whereby new somatic embryos are initiated from somatic embryos. Such cultures have been described in at least 80 Gymnosperm and Angiosperm species. In the initial step (primary somatic embryogenesis) such cultures have to be started from plant explants. In general, primary somatic embryogenesis from vegetative plant explants is, indirect and mostly driven by auxin (AUX) or auxin and cytokinin (AUX/CYT) supplemented media, whereas, from zygotic embryos it is direct and driven, to a larger extent, by CYT or growth regulator free media. Primary somatic embryogenesis from floral plant explants is between these two extremes. Indirect and direct somatic embryogenesis should be seen as two extremes of one continuum: in indirect somatic embryogenesis the embryos develop up to the (pre)-globular stage and in direct somatic embryogenesis to mature stages before they are subjected to secondary embryogenesis. In general, secondary embryogenesis requires no growth regulators in species with CYT driven primary embryogenesis. Whereas, continuous exposure to growth regulators is needed in species with CYT/AUX or AUX driven primary embryogenesis. In most species somatic embryos can be converted into shoots, although the frequencies are mostly low. In general, somatic embryos induced by growth regulator free or CYT supplemented media meet more difficulties in shoot development than embryos induced by AUX supplemented media. Applications of secondary somatic embryogenesis for plant breeding are discussed.
