    Notes: Abstract  The Ser-170 residue of a trypsin-like enzyme from Streptomyces fradiae (SFT), which is considered to be the active-site serine, was replaced with alanine by site-directed mutagenesis to improve the affinity chromatography step for a Kazal-type trypsin inhibitor, pancreatic secretory trypsin inhibitor (PSTI). The resulting mutant SFT, designated as [S170A]SFT, was expressed in Streptomyces lividans and purified to homogeneity. [S170A]SFT was catalytically inactive, but still had the ability to bind tightly to PSTI and to soybean trypsin inhibitor with dissociation constants of 3.1×10-7 M and 1.9×10-8 M respectively. We further demonstrated that recombinant human PSTI secreted into Saccharomyces cerevisiae culture broth could be purified to homogeneity with a one-step [S170A]SFT-affinity column. The purified PSTI contained no molecules intramolecularly cleaved by active trypsin, which are found when trypsin-affinity chromatography is used for the purification. This eliminated the need for further separation of intact PSTI from intramolecularly cleaved PSTI by high-performance liquid chromatography, thus simplifying and improving its purification process.

    Notes: Abstract A formulation that finds the optimal design of a controlled structure is proposed. To achieve this goal, a composite objective composed of structural and control objectives is introduced to be optimized, and the effect of the control weighting is examined. A feedback control law is defined before the structural optimization and then the composite objective will only become a function of structural design variables. As a result, optimal structural design and control forces in steady state are obtained.

    Notes: Abstract An integrated design procedure which is composed of structural design, control design, and actuator locations design is proposed in this paper. First, a composite objective function, formed by a structural and a control objective, is optimized in steady state through the homogenization design method. Then an independent modal space control algorithm (IMSC) is performed on this optimal structure to reduce the dynamic response. Finally, to minimize the control force while still obtaining the same modal response for the controlled modes, the optimal choice for actuator locations is discussed.

    Notes: The topological optimization of components to maximize crash energy absorption for a given volume is considered. The crash analysis is performed using a DYNA3D finite element analysis. The original solid elements are replaced by ones with holes, the hole size being characterized by a so-called density (measure of the reduced volume). A homogenization method is used to find elastic moduli as a function of this density. Simpler approximations were developed to find plastic moduli and yield stress as functions of density.Optimality criteria were derived from an optimization statement using densities as the design variables. A resizing algorithm was constructed so that the optimality criteria are approximately satisfied. A novel feature is the introduction of an objective function based on strain energies weighted at specified times. Each different choice of weighting factors leads to a different structure, allowing a range of design possibilities to be explored.The method was applied to an automotive body rear rail. The original design and a new design of equal volume with holes were compared for energy absorption.
