    Notes: Background: Anti-ulcer agents exert various functional effects on gastric epithelial cells. Aim: The effects of a novel gastro-cytoprotective agent (rebamipide) on epithelial restoration following bile acid damage were assessed using primary cultured rabbit gastric epithelial cells. Methods: Rebamipide was added to complete confluent cell sheets with deoxycholic acid just after creating a cell-free wound (2 mm2). The restoration was monitored and analysed by phase contrast microscopy and an image analyser for 48 h. The migration speed was measured during the initial 3 h after wounding. Cell proliferation was detected by staining for bromodeoxyuridine (BrdU) at 12-h intervals. The labelling index was calculated per unit area. The major cytoskeletal protein actin was detected by immunohistochemical staining. Results: In the controls, restoration was completed 48 h following wounding. Deoxycholic acid retarded this process. The addition of rebamipide to deoxycholic acid abolished the bile acid-induced retardation. The migration speed was 26 μm/h in the controls, 15 μm/h in the deoxycholic acid group and 27 μm/h in the deoxycholic acid plus rebamipide group. In the controls, BrdU-positive cells, which were rarely detected in the initial 24 h, were maximal at 36 h (labelling index 1.7%). In the deoxycholic acid group, proliferation was inhibited (peak labeling index; 0.5% at 48 h). Actin-containing stress fibres were detected throughout the cells and the periphery of the lamellipodia in the controls, and were disrupted in the deoxycholic acid-treated group. Rebamipide prevented these effects. Conclusions: Deoxycholic acid significantly retarded restoration by the inhibition of both cell migration and proliferation, potentially through an effect on the cytoskeleton. Rebamipide protected the mucosal cells from bile acid mediated injury.

    Notes: Abstract Electron temperature observed by the Hinotori satellite with the low inclination at the height of \sim600 km was studied in terms of local time, season, latitude, magnetic declination and solar flux intensity during a 16-month period from 1981 to 1982. The electron temperatures show steep rise in the early morning (well known as morning overshoot), decrease after that and again increase at \sim18 hours (hereafter named as evening overshoot). Generally the morning overshoot becomes more enhanced in the winter hemisphere and for higher solar fluxes. The evening overshoot becomes more pronounced in the mid-latitude in all seasons and more enhanced in the winter hemisphere in the same way as the morning overshoot. A difference is seen between 210°-285° and 285°-360° longitudes where magnetic declination is different. The longitudinal dependence of electron temperature indicates that the neutral wind also contributes to the thermal structure in the low latitude ionosphere.

    Notes: Abstract ⋅Objective This study examines the effect of the force with which a vibrating handle is gripped on the temporary threshold shift of vibratory sensation (TTSv) induced by hand-arm vibration. ⋅  Methods Six healthy subjects gripped a handle vibrating with a 1/3 octave-band vibration, with a central frequency of 200 Hz and an intensity of 39.2 m/s2. Exposure was for 1 min and 10 min, respectively. Gripping forces for the 1-min exposure were 5 N, 10 N, 40 N and 80 N, respectively, with 0 N push-pull force. Gripping forces for the 10-min exposure were the same as for the 1-min exposure, but omitting 80 N. The vibratory sensation threshold at 125 Hz was measured before and after exposure of an exposed fingertip to vibration. The differences measured determine TTS v,t at time t. TTS v,t determines TTSv,0, that is, the temporary threshold shift of vibratory sensation immediately after exposure to vibration according to the estimate made on the basis of the preceding study. The same experimental conditions were repeated 3 times on different days in a soundproof and thermoregulated room. ⋅  Results Our findings show that TTSv increases significantly with increasing gripping force. We also determined the quantitative relationships between TTSv,0 and gripping force as described by the equation where k f and c f are constants and F is gripping force. ⋅  Conclusion This study revealed the importance of ergonomic design in reducing the force with which a vibrating handle is gripped to prevent an adverse effect of local vibration. The equation devised may help in the quantitative assessment of the effect of reduced gripping force.

The vibratory sensation threshold at 125 Hz was measured before and after the vibration exposure at an exposed fingertip. According to a preceding study, we first determined the relationship between the acceleration of the vibration and the temporary threshold shift of vibratory sensation immediately after the vibratory exposure (TTSv, 0) induced by 1/3 octave-band vibration. We then measured TTSv after the exposure to a composite vibration composed of two 1/3 octave-band vibrations that might induce an equal magnitude of TTSv, 0 on the basis of the above relationship. The TTSv, 0 induced by the composite vibration was not larger than the TTSv, 0 induced by the component vibrations. This result suggests that the component of the vibration inducing the largest TTSv, 0 determines the TTSv, 0 by broad-band random vibration.

    Notes: Summary Pseudorabies virus (PRV) early protein 0 (EP0) is a transactivator containing the RING finger domain. Analysis of transactivating activity of truncated forms of the EP0 molecule consisting of 410 amino acids revealed that amino-terminal region containing the RING finger domain, amino acids 1 to 84, and the region between amino acids 114 to 242 containing acidic amino acid sequences were required for the transactivation. On the other hand, the mutant consisting of amino acids 1 to 113 exhibited a dominant-negative property.

    Notes: Summary The nucleotide sequence of the human cytomegalovirus replication origin of strain Towne (anAatII-SacI fragment corresponding to nt 90372–94637 of strain AD169) was determined and compared with AD169. Two differences were found in the nucleotide sequence level. One was the alteration of structural organization (a major difference): a 189-bp region of AD169 (nt 93337–93525) was directly repeated three times in Towne. The other was a change in the nucleotide residue level including substitution, insertion, or deletion (a minor difference). The divergent residues were predominantly localized within the nt 92591–92855 region of AD169. A replication assay revealed that replication ability remained after deletion of the 189-bp repeat but disappeared after either a 1.5-kb deletion from theAatII end or a 0.9-kb deletion from theSacI end. The 1.5- and 0.9-kb regions were relatively conserved. These results indicate that at least two regions essential for replication ability lie outside of both the relatively variable region and the 189-bp repeat and suggest that these essential regions support replication even with a spatial separation of either one (AD169) or three repeats (Towne) of the 189-bp region.
