    Notes: Abstract: Male squirrel monkeys (Saimiri sciureus) were surgically prepared with cranial guide cannulae for acute microdialysis sampling of the putamen nucleus, a dopamine (DA)-rich brain region. On the day of an experiment, an animal was placed in a Plexiglas restraining chair and a microdialysis probe was inserted through the guide into the putamen. Perfusates of artificial cerebrospinal fluid were collected every 20 min over several hours and analyzed via HPLC with electrochemical detection. DA D2/D3 agonist drugs were administered either orally (p.o.) or subcutaneously (s.c.), and changes in levels of DA in the dialysates were measured. All of the drugs tested, i.e., quinpirole (0.5 mg/kg p.o.), talipexole (0.75 mg/kg p.o. or s.c.), and PD 135222 (7 mg/kg p.o.), decreased spontaneous DA overflow by ∼40–50% during the first 2 h following dosing. In animals that routinely underwent the microdialysis procedure up to 23 times over a 2-year period, there was neither an appreciable change in basal DA overflow nor a significant change in the magnitude of drug response. These data suggest that DA D2/D3 agonists attenuate DA neuronal overflow in the primate brain, similar to effects seen in rodents. Furthermore, these results also demonstrate the utility of repeated intracerebral microdialysis as a tool to monitor dynamic changes in neurochemical activity in monkeys over a prolonged period of time.

    Notes: In this article we argue that the state-vector phase-space representation recently proposed by Torres-Vega and co-workers [introduced in J. Chem. Phys. 98, 3103 (1993)] coincides with the totality of coherent-state representations for the Heisenberg-Weyl group. This fact leads to ambiguities when one wants to solve the stationary Schrödinger equation in phase space and we devise two schemes for the removal of these ambiguities. The physical interpretation of the phase-space wave functions is discussed and a procedure for computing expectation values as integrals over phase space is presented. Our formal points are illustrated by two examples. © 1997 American Institute of Physics.

    Notes: Since transdermal nicotine is of value in the treatment of active ulcerative colitis but is often associated with side-effects, an alternative in the form of topical therapy with nicotine enemas has been developed.〈section xml:id="abs1-2"〉〈title type="main"〉MethodsIn an open study, 22 patients with active colitis, all non-smokers, were asked to take a 100 mL enema containing 6 mg of nicotine every night for 4 weeks. Pre-trial treatment using mesalazine (n=16), oral prednisolone (8), cyclosporin (1) and azathioprine (1) was kept constant for the month prior to assessment and during the study period. Symptoms, with stool frequency, were recorded on a diary card and an endoscopy was performed with rectal biopsy at the beginning of the study and after 4 weeks.〈section xml:id="abs1-3"〉〈title type="main"〉ResultsSeventeen of the 22 patients completed 1 month of treatment. Mean duration of relapse was 29 weeks, range 3–94. Sixteen of 17 improved their St Mark's score. Urgency and stool frequency improved in 12 patients, sigmoidoscopic and histological scores in 10. Three patients had a full remission of symptoms with normal sigmoidoscopy. Six of 10 with a partial response continued with the enemas for a second month and five showed further improvement with full remission in two. The enema appeared effective when added to conventional treatment and produced few side-effects.〈section xml:id="abs1-4"〉〈title type="main"〉ConclusionTopical nicotine therapy for ulcerative colitis may have a place in future management, but controlled studies are needed.

    Notes: Abstract Although specific bacteria, dental plaque, and age are associated with periodontal disease, there are currently no reliable predictors of periodontitis severity. Studies in twins have suggested a genetic contribution to the pathogenesis of periodontitis, but previous attempts to identify genetic markers have been unsuccessful. The pro-inflammatory cytokines interleukin-1 (IL-1) and tumor necrosis factor alpha (TNFα) are key regulators of the host responses to microbial infection. IL-1 is also a major modulator of extracellular matrix catabolism and bone resorption. We report a specific genotype of the polymorphic IL-1 gene cluster that was associated with severity of periodontitis in non-smokers, and distinguished individuals with severe periodontitis from those with mild disease (odds ratio 18.9 for ages 40–60 years). Functionally, the specific periodontitis-associated IL-1 genotype comprises a variant in the IL-1B gene that is associated with high levels of IL-1 production. In smokers severe disease was not correlated with genotype. In this study, 86.0% of the severe periodontitis patients were accounted for by either smoking or the IL-1 genotype. This study demonstrates that specific genetic markers, that have been associated with increased IL-1 production, are a strong indicator of susceptibility to severe periodontitis in adults.

    Notes: Abstract Background: Transforming growth factor alpha (TGF-α) and its receptor (EGF-R) may regulate normal and malignant epithelial cell growth by an autocrine mechanism. We investigated the role of TGF-α in regulating head and neck SCC tumor growth. Methods: TGF-α and EGF-R levels were measured in 7 SCC cell lines and 14 SCC biopsies by RIA, Scatchard, and Western analysis. TGF-α autocrine stimulation of DNA synthesis in SCC cell lines was assessed by incubation with TGF-α neutralizing antibodies and tyrphostin AG 1478, a selective and potent inhibitor of EGF-R kinase. Results: All SCC cell lines synthesized TGF-α and expressed elevated EGF-R levels compared to normal keratinocytes. Twelve of the 14 SCC biopsies contained TGF-α protein and 8 had specific EGF-R. Exogenous TGF-α or EGF significantly increased DNA synthesis in 4 of 5 SCC cell lines. TGF-α neutralizing antibodies or tyrphostin AG 1478 reduced DNA synthesis in the two SCC cell lines (FaDu and SCC9) tested. Conclusions: These results indicate that SCC cell lines and tumors usually synthesize TGF-α, have elevated levels of EGF-R, and are mitogenically stimulated by a TGF-α autocrine system. Selective inhibition of the TGF-α system by EGF-R kinase inhibitors or TGF-α neutralizing antibodies may be useful strategies for treating SCC that overexpress TGF-α and its receptor.
