    Notes: Summary: Somatic hypermutation and selection of immunoglobulin (Ig) variable (V)-region genes, working in concert, appear to be essential for memory B-cell development in mammals. There has been substantial progress on the nature of the cis-acting DNA elements that regulate hypermutation. The data obtained suggest that the mechanisms of Ig gene hypermutation and transcription are intimately intertwined. While it has long been appreciated that stringent phenotypic selection forces are imposed on the somatically mutated Ig V regions generated during a T-cell dependent B-cell response, the mechanisms involved in this selection have remained enigmatic. Our studies have questioned the role of foreign antigen deposited on follicular dendritic cells in affinity-based positive selection of V regions, and have shown that this selection takes place in a “clone-autonomous”’ fashion. In addition, our data strongly suggest that affinity for antigen alone is not the driving force for selection of B-cell clones into the memory compartment. In contrast, we suggest that a combination of positive selection for increased foreign antigen binditig, and negative selection of antibody V regions that are autoreactive at the onset of the response, or have acquired autoreactivity via hypermutation, results in the “specificity maturation” of the memory B-cell response

    Notes: The subsolidus and primary crystallization phase field of the “2212” (Bi:Sr:Ca:Cu) phase in the BSCCO system have been investigated. At 830°C, this phase was observed to be in equilibrium with ten phases. Sixteen four-phase equilibrium regions that surround the 2212 phase have been identified at this temperature. The melting events as-sociated with these four-phase regions were studied with differential thermal analysis, quenching experiments, and wicking experiments, in which samples of the liquid for analysis were absorbed by capillary action. The melt com-positions were obtained by using quantitative energy-dis-persive X-ray spectrometry. The 2212 phase melts incon-gruently, beginning at ∼825°C. Its primary crystallization field encompasses a compositional range of 24–42 mol% BiO1.5, 7–33 mol% SrO, 2–27 mol% CaO, and 19–43 mol% CuO. The compositions of melts in equilibrium with the 2212 phase were mostly on the calcium-deficient side of the 2212 solid solution. An approximate polygonal model of the 2212 primary crystallization field has been presented. The addition of silver depresses the melting temperatures in the vicinity of the 2212 phase, from 4°C to 22°C. Silver entered the melt at a saturation level of 2–8 mol%. In the more copper-rich liquids, silver replaced some of the copper, which resulted in a shift of the 2212 crystallization field away from the copper oxide corner.

    Notes: Abstract A molecular marker map, including Mlo mildew resistance, of the spring barley cross Derkado (Mlo-resistant) × B83-12/21/5 (Mlo-susceptible) was scanned for yield QTLs to determine whether the association of Mlo resistance with reduced yield was due to linkage or pleiotropy. Over the mapped portion of the genome of the cross, the QTL with the greatest effect upon yield was located within a 22 cM region between mlo and the simple sequence repeat HVM67 on chromosome 4(4H). The association of Mlo resistance with lower yield was therefore due to a repulsion linkage. Analysis of yield component characters revealed QTL alleles for reduced grain number and earlier heading date in the same region, also associated with Mlo resistance. Genotyping of a range of cultivars and sources of Mlo resistance with the HVM67 simple sequence repeat showed that the Derkado HVM67 allele was rare as it was found only in one other cultivar and four land-races or sources of disease resistance. Grannenlose Zweizeilige, the source, and Salome, the carrier of Mlo resistance in Derkado, have the same HVM67 genotype, although Salome was a mixture of two genotypes. The entire mlo-HVM67 chromosomal segment from Grannenlose Zweizeilige is therefore thought to have been transmitted to Derkado, possibly through joint selection for Mlo resistance and early heading. L92, synonym EP79, was another source of Mlo resistance with the same HVM67 allele as Derkado but recombination must have occurred during the breeding of Atem as it possesses a different HVM67 allele which is present in all the other Mlo sources and cultivars surveyed.
