    Notizen: Abstract: Nicotinic acetylcholine receptors play a major role in excitatory neurotransmission in insect CNSs and constitute an important target for insecticides. Here, we report the isolation and functional characterisation of two cDNAs encoding nicotinic acetylcholine receptor α subunits from a major insect pest, the peach-potato aphid Myzus persicae. These two subunits, termed Mpα1 and Mpα2, are respective structural homologues of the Drosophila Dα2/Schistocerca gregariaαL1 α-subunit pair and the Drosophila ALS α subunit. Xenopus oocyte expression confirmed that each Myzus subunit can form functional acetylcholine- or nicotine-gated channels. However, some electrophysiological and pharmacological properties of the Myzus subunits were distinct from those encoded by the corresponding Drosophila subunits. Coexpression of the Myzus subunits with the chick β2 subunit revealed other differences from the Drosophila system, as only very limited potentiation of agonist-induced currents was observed with Mpα2 and none with Mpα1. Available data therefore indicate that structurally homologous insect nicotinic acetylcholine receptor α subunits from different species can exhibit distinctive physiological and pharmacological characteristics.

    Notizen: The gelada baboon is a graminivorous primate whose ecology is unusually sensitive to ambient temperature. A systems model of the socio-ecology of the gelada is used to predict the impact of global warming on the species’ altitudinal distribution. The species’ lower altitudinal limit will rise by ≈ 500 m for every 2 °C increase in global mean temperature. A 7 °C rise in temperature would be sufficient to result in the species being confined to a small number of isolated mountain peaks, where its chances of survival will be greatly reduced. Changes in local climate are also likely to have significant effects on agricultural practice on the Ethiopian highlands, and this in turn is likely to have repercussions for the distribution patterns of the gelada by further constraining the habitat available to them.

    Notizen: The enzyme methylglyoxal synthase (MGS) was partially purified from Escherichia coli extracts, and the amino-terminal sequence of candidate proteins was determined, based on the native protein being a tetramer of about 69 kDa. Database analysis identified an open reading frame in the E. coli genome, YccG, corresponding to a protein of 16.9 kDa. When amplified and expressed from a controlled promoter, it yielded extracts that contained high levels of MGS activity. MGS expressed from the trc promoter accumulated to approximately 20% of total cell protein, representing approximately 900-fold enhanced expression. This caused no detriment during growth on glucose, and the level of methylglyoxal (MG) in the medium rose to only 0.08 mM. High-level expression of MGS severely compromised growth on xylose, arabinose and glycerol. A mutant lacking MGS was constructed, and it grew normally on a range of carbon sources and on low-phosphate medium. However, the mutant failed to produce MG during growth on xylose in the presence of cAMP, and growth was inhibited.

    Notizen: SUMMARY: Transforming growth factor β (TGFβ) is a multifunctional cytokine for which three isoforms, TGFβ1, -2 and -3 have been characterized in mammals. Several reports suggest an involvement of TGFβ in the cellular and molecular basis of renal fibrosis. Most of these studies have focused exclusively on TGFβ1. Here we demonstrate using reverse transcriptase polymerase chain reaction (RT-PCR) that transcripts from all three isoforms of TGFβ were detected in 31 renal biopsies, control tissue and cultured human mesangial cells. A novel alternative transcript of TGFβ2 was identified in renal tissue. the variation in transcript levels detected using this technique did not exhibit a striking correlation with any specific disease type. In addition, complement deposition and localization (i.e. mesangial or capillary wall) does not appear to have any direct correlation with the occurrence of a particular TGFβ isoform transcript. It is likely that dynamic changes in all TGFβ transcript levels occur in normal and diseased kidneys as a response to physiological and pathological demands. Most investigations have focused on one TGFβ isoform neglecting the contribution of the others. Studies which analyse all known isoforms and their alternative transcripts may establish the interplay between these during the progression of renal disease.

    Notizen: The putative outer membrane location of the OMP90 (formerly POMP) family from the ovine abortion strain of Chlamydia psittaci was investigated by immunoelectron microscopy. Using a non-embedding technique, antigens were shown to be localised on the outer membrane surface of both elementary and reticulate bodies, the infectious and non-infectious forms of Chlamydiae respectively. Antibodies affinity-purified against the expressed amino- and carboxy-terminal halves of one of the family members, OMP90A, demonstrated that the amino half is surface-exposed while the carboxyl half is most probably localised internally. Surface localisation on elementary bodies indicates the importance of these proteins as protective antigen candidates.
