    Notes: Abstract  A computer program has been designed to manage marker information in recombinant inbred-line populations. The objective is to select pairs of inbred lines (either recombinant-inbred or doubled-haploid) to be intercrossed, in order to accumulate all or most favourable alleles, either with additive effects or with interactive effects. The population size required to have a 95% chance of obtaining the best line from a given cross is computed, taking into account the number of QTLs and the probability that no recombination event occurs in any of the QTL confidence intervals. It is shown that the accuracy of QTL location greatly affects selection efficiency and that a recurrent selection scheme is highly preferable for pyramiding many QTLs. An application to the bread-making quality improvement of wheat is presented.

    Notes: Abstract Dictyostelium RLC null cells have defects in cytokinesis and development that can be rescued by expression of either the wild type Dictyostelium RLC or an RLC mutant that cannot be phosphorylated by MLCK (S13A) (Ostrow et al., 1994). The wild type and S13A mutant LCs rescued the cells equally well, despite the fact that RLC phosphorylation increases purified Dictyostelium myosin's activity 5-fold. In this report, we assess the ability of foreign RLCs to rescue the RLC null phenotype. The RLC from smooth muscle myosin, whose activity is tightly controlled by phosphorylation, rescued the null cell phenotype. The purified hybrid myosin had an activity and motility comparable to phosphorylated Dictyostelium myosin. In contrast, cells expressing skeletal muscle RLC were deficient in cytokinesis and development, despite having an activity and motility similar to that of myosin with the unphosphorylatable S13A mutant RLC. Neither foreign LC was phosphorylated when expressed in Dictyostelium. These results suggest that the level of actin-activated ATPase activity and motility is not the sole determinant of proper myosin function in vivo. Other heavy chain/light chain interactions, which occur only with the native RLC and smooth muscle RLC, appear to be necessary for optimal function.

    Notes: Abstract Emissions of NOx and SO2 were monitored in the presence of ammonia liquor in a 0.09 m2 and 2 m high stainless-steel fluidized-bed combustor. Experiments were carried out at 2 m/s fluidizing velocity, 40% excess air, and 870 °C bed temperature. Ammonia liquor with 7% ammonia by weight was injected into the freeboard of the combustor 52 cm above the distributor through a water-cooled injector. A 65 : 34 primary/secondary air ratio was maintained throughout the investigation. Approximately 70% of NO and 20% of SO2 was reduced at an NH3/NO molar ratio of 2 : 1, respectively. However, a higher reduction in SO2 emissions (62%) was achieved at a very high NH3/NO molar ratio of 7 : 1. These experiments showed that ammonia addition did have a significant effect in SO2 reduction if injected in an excess amount. The injection of ammonia liquor combined with staged combustion was found to be very effective in reducing NOx emissions. A reduction of about 50% was achieved at an NH3/NO molar ratio of 0.6 : 1 which is also coupled with a very low level of ammonia in the flue.

    Notes: Abstract In Drosophila, genetic loss of the tumour suppressor protein Dlg (in dlg mutants) or p127 (in lgl mutants) leads to loss of epithelial structure and excess proliferation in the imaginal discs and brain of the developing larva. These phenotypes show most of the characteristic features of human neoplasia, so study of the gene products may contribute to our understanding of cancer. Both proteins occur in high molecular-mass complexes in the membrane-associated cytoskeleton, and they both appear to play dual roles as structural proteins and active partners in signal transduction. Dlg is a membrane-associated guanylate kinase homolog (MAGUK) found at septate junctions between epithelial cells, as well as at neuromuscular junctions. Specific domains of the protein are required for membrane targeting and for localisation in junctions, and for epithelial cell proliferation control; all of these functions are probably mediated through binding to other proteins. Loss of Dlg results in the absence of septate junctions, delocalisation of several proteins including Fasciclin III, Coracle, actin and tubulin, and loss of cell polarity. p127, although mostly associated with the plasma membrane, is in most cell types also present in the cytoplasm. It shows a dynamic subcellular distribution, and its cytosolic and membrane-associated forms play distinctive roles by interacting with different binding partners, in particular the non-muscle myosin II heavy chain. Defects associated with the lgl temperature-sensitive allele include loss of the columnar organisation of epithelial cells, indicating that p127 contributes to cell structure, presumably by stabilising the plasma membrane. In addition to their organising functions, both Dlg and p127 appear to be involved in signal transduction pathways. Study of these genes shows that some proteins play both structural and functional roles, and that cancer can involve changes in the organisation of signalling pathways in addition to changes in individual pathway components.

    Notes: Abstract A new device was developed to assess fatigue life under biaxial tensile loading at elevated temperatures. It makes use of an annular disk specimen and can be easily mounted onto a standard push-pull machine so that the axial force is converted into radial forces extending across the disk specimen. Therefore, a positive ratio of the tangential to the radial stress can be imposed at the reduced section of the disk specimen; this ratio depends on the specimen configuration and may be fixed to a value ranging from 0.5 to 0.9 by varying the inner diameter of the disk. The proposed device has performed successfully and was used to study the cyclic behavior of Type-304 stainless steel subjected to various biaxial tensile stress states at room temperature and at 200°C. The data obtained from this experimental procedure have been analyzed to evaluate the effectiveness of some correlations already available for treating the biaxial cyclic stress-strain response in terms of the uniaxial behavior. This analysis shows that a successful correlation should account for all the stress components. The authors discuss the concept used in the modeling of the material cyclic behavior and the formulation of a biaxial fatigue damage parameter necessary for an effective analytical life prediction methodology.
