Notes: Abstract It has been demonstrated that the administration of CTLA4Ig protein can induce the suppression of allograft and xenograft rejection. The purpose of this study is to determine the effect of adenovirus-mediated gene transfer with CTLA4Ig gene on the transgene expression and suppression of alloimmune response in allogeneic cardiac transplantation of rats. Adenoviral vectors with β galactosidase or human CTLA4Ig cDNA (Adex/LacZ, Adex/hCTLA4Ig) were constructed. These vectors were transduced to the liver of Lewis (LEW) rats by intravenous injection. Then LEW rats received heterotopic cardiac transplantation from Dark Agouti rats. Experiments were performed using both CTLA4Ig gene transduction and/or immunosuppressant FK506. The transgene expression after adenovirus-mediated transfer with CTLA4Ig cDNA lasted for several months, compared with several weeks after that in controls, which was proportional to the blood concentration of CTLA4Ig protein. By the administration of 1 × 109 PFU of Adex/hCTLA4Ig, the survival of cardiac grafts was significantly prolonged, compared with controls or the use of 1 × 108 PFU of Adex/hCTLA4Ig. In the rats with beating grafts over 100 days, the blood concentrations of CTLA4Ig were undetectable. The combination therapy using a low titer Adex/hCTLA4Ig and low-dose of FK506 was synergistically effective on this cardiac transplantation model. In conclusion, adenovirus-mediated gene transfer with CTLA4Ig gene was efficient for the prolongation of both transgene expression and allograft survival.

Notes: Abstract The precursor of the non-Aβ component of Alzheimer’s disease amyloid (NACP), also called α-synuclein, is a major component of Lewy bodies in Parkinson’s disease (PD) as well as of neuronal and oligodendroglial cytoplasmic inclusions in multiple system atrophy. We previously reported argyrophilic, tau-negative glial inclusions in the midbrains of patients with PD and have now conducted immunocytochemical and ultrastructural examinations. The PD glial inclusions also are immunoreactive for NACP/α-synuclein, but not for β-synuclein, and ultrastructurally are composed of filamentous structures about 25–40 nm in diameter. Double immunolabeling showed that the inclusions were present in both astrocytic and oligodendroglial cells. They were located within the substantia nigra in 13 of 30 patients with PD and outside the nigra in 24. The number of inclusions was correlated with the severity of nigral neuronal loss. These findings indicate that abnormal accumulation of NACP/α-synuclein in glial cells is a pathological feature of PD related to its progression.

Notes: Abstract We studied a cystic ganglioglioma (GG) located in the right frontal lobe of the brain. Interestingly, the fibrillary spindle glial cells were often arranged in a fascicular pattern, and the generally uniform, round-to-oval delicate nuclei appeared to resemble those of ependymoma; and the neoplastic neurons often contained neurofibrillary tangles (NFTs). The glial component was positive for glial fibrillary acidic protein and occasionally contained granular or microvesicular structures positive for epithelial membrane antigen. Ultrastructural investigation revealed that the glial cells were ependymal in nature; intracytoplasmic lumina and intercellular microrosettes lined with cilia and microvilli, as well as long zonulae adherentes, were evident. In addition, chromogranin A-positive granular staining, neurosecretory-granule-like structures, and parallel arrays of microtubules were sometimes associated with the blood vessels. We considered the present case to be an unusual example of GG with an ependymoma, more precisely a tanycytic ependymoma, as the glial component; to our knowledge, the existence of ependymoma as the main glial component of this particular tumor has not been described before. The occurrence of NFTs, which has been reported in several cases of GG, was an additional, unusual feature.

Notes: Abstract The directional solidification and the interface structure of BaTiO3-CoFe2O4 eutectic grown by the floating zone melting method were investigated. The microstructure of the eutectic were found to consist grains of lamellar or fibrous morphology. Two types of crystal structures of BaTiO3 were observed for different growth conditions. The orientation relationships between BaTiO3 and CoFe2O4 (the spinel-type crystal structure) are a) hcp BaTiO3: (111) CoFe2O4 // (001) BaTiO3 and (110) CoFe2O4 // (110) BaTiO3, and b) tetra/cubic BaTiO3: (001) CoFe2O4 // (001) BaTiO3 and (100) CoFe2O4 // (100) BaTiO3. The interface has misfit dislocations due to the accommodation of lattice mismatch.

Notes: Abstract The influence of female mice (Mus musculus) on intermale aggression and the female choice between dominant and subordinate males were tested. Normal male mice were paired with a castrated male or a female. Two pairs of the same kind were housed together after acclimation for 3 days in half (home cage) of the apparatus, which was composed of eight cages connected by tunnels. One of the two normal males became dominant. The dominant males became more aggressive in the presence of females than in the presence of castrated males. The two normal males dwelt in their own home cages. The two females dwelt together and preferred to dwell in the home cages of the dominant male. The two castrated males dwelt together and seemed to decide their dwelling cage more freely than the subordinate normal male.
