    Notes: With recent advances in dental technology, titanium is currently used for fabrication of crowns, fixed partial dentures, implant frameworks, and removable partial denture frameworks. The use of titanium-aluminum-vanadium (Ti-6Al-4V) alloy assumes that it imparts similar anti-corrosion characteristics to the commercially pure titanium. This clinical report describes a patient who experienced discoloration of a Ti-6Al-4V alloy removable partial denture.

    Notes: Treatment of PC12 cells with nerve growth factor induces their differentiation into sympathetic neuron-like cells and the concomitant expression of the neural cell adhesion molecule L1, a member of the Ig superfamily. To investigate the mechanism of L1-stimulated neurite outgrowth in PC12 cells, substrate-immobilized fusion proteins containing different extracellular domains of L1 were assayed for their neuritogenic activity. Surprisingly, domain Ig2 of L1, which was previously found to contain both homophilic binding and neuritogenic activities, failed to promote neurite outgrowth. In contrast, L1-Ig6 stimulated neurite outgrowth from PC12 cells. Despite this, homotypic binding of PC12 cells was significantly inhibited by antibodies against L1-Ig2, indicating that L1–L1 binding contributed to the intercellular adhesiveness of PC12 cells, but L1-stimulated neurite outgrowth depends on heterophilic interactions. Thus, PC12 cells provide a valuable model for the study of these two distinct functions of L1. Mutagenesis of L1-Ig6 highlighted the importance of the Arg-Gly-Asp motif in this domain for neuritogenesis. Inhibition studies using cyclic Arg-Gly-Asp-containing peptide and anti-integrin antibodies suggested the involvement of αvβ3 integrin. Furthermore, neurite outgrowth stimulated by L1-Ig6 was inhibited by lavendustin A and the MEK inhibitor PD98059, suggesting a signaling pathway that involves tyrosine kinase activation and the mitogen-activated protein kinase cascade.

    Notes: Lichen sclerosus (LS) affects anogenital skin alone in 80% of cases. When extragenital disease occurs, it usually affects the trunk, neck, axillae and wrist flexures. Nail involvement with LS is rare. In contrast, lichen planus (LP) commonly affects extragenital skin. Mucosal lesions occur in 50% of cases, affecting the mouth and genitalia. Nail disease in LP is common, and, if severe, can lead to destruction of the nail bed. LS and LP can coexist. We report two cases of LS with nail involvement. In the Case 1 disease was confined to the nail, and nail biopsy confirmed LS. In the Case 2, the nail changes formed part of the widespread genital and extragenital LS, confirmed histologically. We review existing literature on nail disease in LS and discuss the possible aetiology of the nail changes.

    Notes: Abstract The adaptive immune response is initiated by the interaction of T cell antigen receptors with major histocompatibility complex molecule-peptide complexes in the nanometer scale gap between a T cell and an antigen-presenting cell, referred to as an immunological synapse. In this review we focus on the concept of immunological synapse formation as it relates to membrane structure, T cell polarity, signaling pathways, and the antigen-presenting cell. Membrane domains provide an organizational principle for compartmentalization within the immunological synapse. T cell polarization by chemokines increases T cell sensitivity to antigen. The current model is that signaling and formation of the immunological synapse are tightly interwoven in mature T cells. We also extend this model to natural killer cell activation, where the inhibitory NK synapse provides a striking example in which inhibition of signaling leaves the synapse in its nascent, inverted state. The APC may also play an active role in immunological synapse formation, particularly for activation of naive T cells.

    Notes: We present an effective linear response approach to pump–probe femtosecond coherence spectroscopy in the well-separated pulse limit. The treatment presented here is based on a displaced and squeezed state representation for the nonstationary states induced by an ultrashort pump laser pulse or a chemical reaction. The subsequent response of the system to a delayed probe pulse is modeled using closed form nonstationary linear response functions, valid for a multimode vibronically coupled system at arbitrary temperature. When pump–probe signals are simulated using the linear response functions, with the mean nuclear positions and momenta obtained from a rigorous moment analysis of the pump induced (doorway) state, the signals are found to be in excellent agreement with the conventional third-order response approach. The key advantages offered by the moment analysis-based linear response approach include a clear physical interpretation of the amplitude and phase of oscillatory pump–probe signals, a dramatic improvement in computation times, a direct connection between pump–probe signals and equilibrium absorption and dispersion lineshapes, and the ability to incorporate coherence associated with rapid nonradiative surface crossing. We demonstrate these aspects using numerical simulations, and also apply the present approach to the interpretation of experimental amplitude and phase measurements on reactive and nonreactive samples of the heme protein myoglobin. The role played by inhomogeneous broadening in the observed amplitude and phase profiles is discussed in detail. We also investigate overtone signals in the context of reaction driven coherent motion. © 2001 American Institute of Physics.

    Notes: We analyze the nonstationary vibrational states prepared by ultrashort laser pulses interacting with a two electronic level molecular system. Fully quantum mechanical expressions are derived for all the moments of the coordinate and momentum operators for the vibrational density matrices associated with the ground and excited electronic states. The analysis presented here provides key information concerning the temperature and carrier frequency dependence of the moments, and relates the moments to equilibrium absorption and dispersion line shapes in a manner analogous to the "transform methods" previously used to describe resonance Raman scattering. Particular attention is focused on the first two moments, for which simple analytical expressions are obtained that are computationally easy to implement. The behavior of the first two moments with respect to various parameters such as the pulse carrier (center) frequency, pulse width, mode frequency, electron-nuclear coupling strength, and temperature is investigated in detail. Using rigorous analytical formulas, we also discuss the laser pulse induced squeezing of the nuclear distributions as well as the pulse induced vibrational heating/cooling in the ground and excited states. The moment analysis of the pump induced state presented here offers a convenient starting point for the analysis of signals measured in pump–probe spectroscopy. The moment analysis can also be used, in general, to better understand the material response following ultrashort laser pulse excitation. © 2001 American Institute of Physics.

    Notes: We demonstrate novel lock-in detection techniques, using wavelength selective modulation of ultrafast pump and probe laser pulses, to discriminate between vibrational coherence and electronic population decay signals. The technique is particularly useful in extracting low frequency oscillations from the monotonically decaying background, which often dominates the signal in resonant samples. The central idea behind the technique involves modulating the red and/or blue wings of the laser light spectrum at different frequencies, ΩR and ΩB, followed by a lock-in detection at the sum or difference frequency, ΩR±ΩB. The wavelength selective modulation and detection discriminates against contributions to the pump–probe signal that arise from degenerate electric field interventions (i.e., only field interactions involving different optical frequencies are detected). This technique can be applied to either the pump or the probe pulse to enhance the off-diagonal terms of the pump induced density matrix, or to select the coherent components of the two-frequency polarizability. We apply this technique to a variety of heme-protein samples to reveal the presence of very low-frequency modes (∼20 cm−1). Such low-frequency modes are not observed in standard pump–probe experiments due to the dominant signals from electronic population decay associated with resonant conditions. Studies of the diatomic dissociation reaction of myoglobin (MbNO→Mb+NO), using wavelength selective modulation of the pump pulse, reveal the presence of an oscillatory signal corresponding to the 220 cm−1 Fe–His mode. This observation suggests that the spin selection rules involving the ferrous iron atom of the heme group may be relaxed in the NO complex. Mixed iron spin states associated with adiabatic coupling in the MbNO sample could explain the fast time scales and large amplitude that characterize the NO geminate recombination. © 2001 American Institute of Physics.

    Notes: 1. We studied whole-ecosystem metabolism in eight streams from several biomes in North America to identify controls on the rate of stream metabolism over a large geographic range. The streams studied had climates ranging from tropical to cool-temperate and from humid to arid and were all relatively uninfluenced by human disturbances.2. Rates of gross primary production (GPP), ecosystem respiration (R) and net ecosystem production (NEP) were determined using the open-system, two-station diurnal oxygen change method.3. Three general patterns in metabolism were evident among streams: (1) relatively high GPP with positive NEP (i.e. net oxygen production) in early afternoon, (2) moderate primary production with a distinct peak in GPP during daylight but negative NEP at all times and (3) little or no evidence of GPP during daylight and a relatively constant and negative NEP over the entire day.4. Gross primary production was most strongly correlated with photosynthetically active radiation (PAR). A multiple regression model that included log PAR and stream water soluble reactive phosphorus (SRP) concentration explained 90% of the variation in log GPP.5. Ecosystem respiration was significantly correlated with SRP concentration and size of the transient storage zone and, together, these factors explained 73% of the variation in R. The rate of R was poorly correlated with the rate of GPP.6. Net ecosystem production was significantly correlated only with PAR, with 53% of the variation in log NEP explained by log PAR. Only Sycamore Creek, a desert stream in Arizona, had positive NEP (GPP: R 〉 1), supporting the idea that streams are generally net sinks rather than net sources of organic matter.7. Our results suggest that light, phosphorus concentration and channel hydraulics are important controls on the rate of ecosystem metabolism in streams over very extensive geographic areas.

    Notes: Enterococcus faecium strain 9631355 was isolated from animal sources on the basis of its resistance to the growth promotant avilamycin. The strain also exhibited high-level resistance to evernimicin, a drug undergoing evaluation as a therapeutic agent in humans. Ribosomes from strain 9631355 exhibited a dramatic reduction in evernimicin binding, shown by both cell-free translation assays and direct-binding assays. The resistance determinant was cloned from strain 9631355; sequence alignments suggested it was a methyltransferase and therefore it was designated emtA for evernimicin methyltransferase. Evernimicin resistance was transmissible and emtA was localized to a plasmid-borne insertion element. Purified EmtA methylated 50S subunits from an evernimicin-sensitive strain 30-fold more efficiently than those from a resistant strain. Reverse transcription identified a pause site that was unique to the 23S rRNA extracted from resistant ribosomes. The pause corresponded to methylation of residue G2470 (Escherichia coli numbering). RNA footprinting revealed that G2470 is located within the evernimicin-binding site on the ribosome, thus providing an explanation for the reduced binding of the drug to methylated ribosomes.
