    Notes: Introduction: The aim of this study was to investigate whether the characteristics of endocardial ventricular repolarization are associated with differential risk for sudden death. Prolonged surface QT interval is associated with increased arrhythmic risk after myocardial infarction (MI), but the underlying mechanism of QT prolongation and its relation to lethal arrhythmias are unclear. Methods and Results: Ventricular fibrillation (VF) risk was assessed in 12 dogs 1 month after anterior MI during an exercise test coupled with brief circumflex coronary occlusion. Susceptible dogs (n = 5) developed VF during the brief ischemic episode, whereas resistant dogs did not (n = 7). Surface QT interval was measured at rest. Endocardial electroanatomic catheter maps of left ventricular repolarization were obtained in four unique regions identified by echocardiography and compared between groups. Compared to resistant dogs, susceptible dogs were characterized by prolonged surface QT intervals (240 ± 10 msec vs 222 ± 7 msec, P = 0.04). In addition, they had lower baroreflex sensitivity (9.7 ± 1.5 msec/mmHg vs 28 ± 9.8 msec/mmHg, P 〈 0.01) and a tachycardic response to acute ischemia suggesting higher propensity for stronger sympathetic reflexes. Surface QT interval prolongation in susceptible dogs was due to a marked heterogeneity of endocardial left ventricular repolarization (239 ± 42 msec, basal anterior wall vs 197 ± 35, lateral wall; P 〈 0.001). Resistant animals had no regional differences in endocardial repolarization. Conclusion: Sympathetic activation following MI not only produces adverse structural remodeling but also contributes to adverse electrophysiologic remodeling resulting in heterogeneous ventricular repolarization and in a myocardial substrate conducive to lethal reentrant arrhythmias.     Notes: Human beings are essentially vulnerable in the view that their existence qua humans is not given but construed. This vulnerability receives basic protection from the State, expressed in the form of the universal rights all citizens are meant to enjoy. In addition, many individuals fall prey to destitution and deprivation, requiring social action aimed at recognising the specific harms they suffer and providing remedial assistance to palliate or remove their plights.Citizens receive protection against their biologic vulnerability by means of an in rem right to health [care], which is more an attitude of protection than a specific programme. When individuals become susceptible, that is, biologically weak or diseased, they also increase their predisposition to additional harm, and require social actions to treat their demeaned condition. Such assistance takes the form of positive healthcare rights.Research on human beings has been slow to observe that the subjects recruited are susceptible, especially so if research is done in less developed countries. By mislabelling them as vulnerable – a characteristic they share with all humans – sponsors avoid registering the deprivation these people suffer, and the ethical obligation to offer them remedial help.The distinction between vulnerability and susceptibility also marks the difference between being intact but fragile – vulnerable – and being injured and predisposed to compound additional harm – susceptible. Awareness of this difference should give additional force to the rejection of double standards in research ethics.

    Notes: Background:  Epidermolytic hyperkeratosis (EH) is most commonly associated with the diffuse involvement of congenital ichthyosiform erythroderma, but can also be found in a localized pattern. Localized EH is rare, but mucocutaneous lesions have been been identified, most commonly in the mouth.Methods:  We observed a 58-year-old African-American female who noted spots on her genitalia for approximately 2 years. The lesions were increasing in size, darkening, and had become pruritic and sore over the past 6 months.Results:  Physical examination revealed seven scattered, tan to brown, verrucoid papules on the labia and mons pubis, resembling condylomata acuminata or Bowenoid papulosis. Biopsy of a single labial papule revealed epidermal acanthosis, compact hyperkeratotic papillomatosis, perinuclear clear zones, granular keratohyalin clumping, hypergranulosis, and dyskeratosis resulting in intracellular eosinophilic globules, all characteristic of EH.Conclusions:  Because of the rarity of localized genital EH and similar appearance to common diagnoses, clinical confusion may occur without biopsy.

    Notes: Six cheese varieties encompassing a wide compositional and age range were analyzed fresh and after aging to determine correlations between casein proteolysis products, fat content, moisture content, and torsion results. Shear stress, a measure of the strength of the casein matrix, was dependent on the amount of intact αsl-casein. Shear strain, a measure of the cohesiveness of a structure, exhibited a negative correlation with fat content and a positive correlation with moisture content. Shear strain was also correlated with the ratio of moisture to protein, an indication of the interaction between casein particles. Torsion tests provide information on the level of degradation of the casein network in cheese, which can be used by manufacturers to alter storage conditions to optimize the quality of their aged cheese., April 10, 2003

    Notes: Abstract:  The mechanisms underlying induction of UVB-induced immunosuppression are not fully understood, but tumour necrosis factor alpha (TNF-α) is suggested to play a central role. A single base pair polymorphism at position − 308 in the promoter region of the TNF-α gene associated with an enhanced secretion of TNF-α has been identified in humans. We have therefore investigated the association of the − 308 polymorphism with the risk of basal cell carcinoma (BCC) in humans. The frequency of TNF G and TNF A alleles among Caucasian patients with a previous BCC (n= 191) and healthy adults (n= 107) were compared. For the TNF − 308 polymorphism there was no significant association between the genotype or allele frequencies and having a BCC. To determine whether patients with a previous BCC had an increased capacity to secrete TNF-α, mononuclear cells were stimulated with lipopolysaccharide. Mononuclear cells from patients with a previous BCC (n= 15) demonstrated a significantly increased release of TNF-α upon stimulation with lipopolysaccharide (P 〈 0.03) compared with mononuclear cells from age-matched control subjects (n= 16). Further studies of other polymorphisms of the TNF-α gene associated with increased TNF-α production and BCC are necessary.

    Notes: The ability of juvenile summer flounder Paralichthys dentatus to utilize dietary lipid as energy, and the effect of dietary lipid on weight gain and body composition was investigated in a 12-week feeding trial. Diets were formulated to provide 55% crude protein from herring meal and casein. Menhaden oil was added to produce diets with 8, 12, 16 or 20% total lipid while providing 16.0 kJ available energy/g dry diet. The diet containing 20% total lipid supplied 16.7 kJ available energy/g dry diet due to the high levels of protein and lipid. An additional diet was included to reproduce currently available commercial diet formulations for flounder, providing 55% crude protein supplied solely from herring meal and 16% total dietary lipid. Juvenile summer flounder (initial weight 23 g) were stocked into triplicate aquaria in a closed, recirculating system maintained at 20 C. Fish were fed 2% of body weight each day divided into two equal feedings. Upon termination of the study, effects of dietary lipid on weight gain, body condition indices, and proximate composition were determined. Weight gain (96–149% of initial weight), feed efficiency ratio values (0.43–0.48). fillet yield, and whole-body composition all were unaffected by dietary lipid level. High levels of dietary lipid did increase the lipid content in the finray muscle, as fish fed diets containing 16 and 20% dietary lipid had significantly higher lipid levels than fish fed the diet containing 8% lipid. No apparent protein sparing effect of lipid was observed. These data indicate that currently available commercial feeds for summer flounder may be over-formulated and show a need for further research to determine specific and accurate nutritional information for this species.

    Notes: Nutrient sensing plays important roles in fungal development in general, and specifically in critical aspects of pathogenicity and virulence, for both animal and plant pathogens. Dimorphic pathogens such as the phytopathogenic smut fungi, Ustilago maydis and Microbotryum violaceum, must switch from a yeast-like to a filamentous form in order to cause disease. Two genes encoding methylammonium permeases (MEPs) were identified from each of these latter fungi and all the encoded proteins were most similar to Mep2p, the high-affinity permease from Saccharomyces cerevisiae that plays a direct role in pseudohyphal or filamentous growth for that organism. This is the first report of MEPs from pathogenic fungi. The two genes from U. maydis and one of the genes from M. violaceum were expressed in diploid S. cerevisiae mutants deleted for all three mep genes (mep1mep2mep3). Each of the heterologous genes could complement the severe growth defect of the S. cerevisiae mutant on low ammonium. Moreover, the U. maydis ump2 gene, initially detected as an upregulated gene in budding cells, was also able to complement the pseudohyphal defect characteristic of the mutant yeast. This gene is thus one of few heterologous MEP genes capable of efficiently restoring pseudohyphal growth in yeast. For U. maydis, disruption of ump2 eliminated the filamentous phenotype of haploid cells on low ammonium, while ump1 disruption only slightly reduced methylamine uptake. The most significant drop in methylamine uptake was seen for the ump2 and the ump1ump2 double mutants. Moreover, when grown in liquid medium, the ump1ump2 double mutant aggregated and sedimented. Also, the importance of a putative site for phosphorylation by protein kinase A was investigated in both Mep2p and Ump2p via site-directed mutagenesis of the respective genes. A mutation predicted to prevent phosphorylation of either protein, still allowed each to provide growth on low ammonium, but eliminated their abilities to provide pseudohyphal growth for the S. cerevisiae triple mutant. These findings allow us to present a model of how ammonium transporters play a role in regulating dimorphic growth in fungi.

    Notes: This study uses information obtained from the advanced manufacturing technology (AMT) literature to develop a conceptual framework that seeks to illustrate the impact of the management information systems (MIS) department on the different facets of AMT adoption. A detailed survey instrument was administered to a cross-section of manufacturing firms in the USA to collect the data required to test five hypotheses relating to the efficacy of this framework. The results of this study indicate that the proposed framework is particularly useful in explaining the role of MIS departments in firms that are attempting to integrate advanced process and information technologies. This finding and other results of this study and their implications are discussed.

    Notes: Enterprise resource planning systems, if implemented successfully, can bestow impressive strategic, operational and information-related benefits to adopting firms. A failed implementation can often spell financial doom. Currently, most of the information about the failures and successes are based on reports on implementations in large manufacturing and service organizations. But enterprise resource planning vendors are now steadily turning their marketing sights on small and medium-sized manufacturers. The time is ripe for researchers to gather, analyze and disseminate information that will help these firms to implement their projects successfully. This research adopts a multiple case study approach to investigate the implementation process in small and midsize manufacturing firms in the US. The research focuses on implementation activities that foster successful installations and are developed using information gleaned from our field studies of four projects. Avenues for future research are also suggested.
