    Notes: Zusammenfassung Ein Überblick zeigt Methoden der analytischen Trennung und Bestimmung von aliphatischen Monocarbonsäuren. Die Möglichkeiten der Trennung aliphatischer Monocarbonsäuren an stark basischen Anionenaustauschern in organischen Lösungsmitteln werden theoretisch und praktisch untersucht. Zu diesem Zweck wurden Trialkylpolyvinylbenzyl-ammoniumharze mit verschiedenen Substituenten am quaternären Stickstoffatom dargestellt und nach Ermittlung einiger Kennzahlen für elutionschromato-graphische Trennversuche eingesetzt. Es wurde der bisher in der Literatur nicht beschriebene Versuch unternommen, lediglich durch Mischungen stark polarer und unpolarer Lösungsmittel unter Ausnutzung der Allgemeinadsorption chromatographische Trennungen an Anionenaustauscherharzen zu erreichen. Als am besten geeignetes Elutionsmittel erwies sich ein Gemisch aus Dimethylformamid, Benzol und Wasser. Es konnte eine quantitative Trennung von Säurege-mischen erzielt werden, die bis zu sechs Komponenten der Säuren C2 bis C14 (Essigsäure bis Stearinsäure) enthalten. Die Reihenfolge der Desorption sowie die Güte des Trenneffektes sind abhängig von der Schrumpfung des Harzes im Lösungsmittel. Während am trimethylaminsubstituierten Harz die Säuren beginnend mit Essigsäure in der Reihenfolge ihrer Kohlenstoffzahl austreten (Traubesche Regel), kehrt sich die Reihenfolge beim Triäthylaminharz und anderen längerkettigen Trialkylammoniumaustauscherharzen um. Diese Effekte werden unter Berücksichtigung der besonderen Eigenschaften des Dimethylformamids als eine Überlagerung von Austausch-und Allgemeinadsorption crklärt.

    Notes: All tRNA sequences so far known can be folded into a cloverleaf structure. Physical data and chemical reactions allow us to draw conclusions on secondary (cloverleaf) and tertiary structure. N-oxidation of adenosine to adenosine-1-N-oxide can be done with monoperphthalic acid in non-base-paired regions of polynucleotides and can be followed easily by changes in absorption of ultraviolet light. Thus this method can be used to determine the structure of tRNA's. A fingerprint of the N-oxidation product of tRNAyeastPhe reveals that all adenosine residues are protected except the 3′-terminal adenosine and the three adenosine residues in or adjacent to the anticodon. On this basis a conformation of tRNAyeastPhe is proposed. Similar tertiary structures can be constructed for the other tRNA's. In order to connect tertiary structure of a tRNA and recognition by its aminoacylating enzyme, the rate of aminoacylation, as a function of temperature, was measured. Neither changes in the anticodon nor specific changes at the 3′-terminal adenosine abolish aminoacylation. Single crystals of tRNAyeastPhe were obtained from aqueous solutions upon addition of various organic solvents.

    Notes: Introduction: Cryocatheter techniques have been successfully applied to treat supraventricular tachycardia but there are no reports on their value in treating ventricular tachycardia (VT). We present our initial experience with cryocatheter ablation of right ventricular outflow tract (RVOT) tachycardia. Methods and Results: Cryocatheter ablation was attempted in 14 patients (13 females, age 45.9 ± 12.7 years) who were highly symptomatic due to frequent monomorphic ventricular extrasystole (VES) or nonsustained VT originating within the RVOT. A 9-Fr, 8-mm-tip cryocatheter was used for both mapping and ablation. Cryoablation was started after localizing the arrhythmic focus by pace and activation mapping. Ablation success, defined by complete disappearance of target VES/VT acutely and during a follow-up of 9.3 ± 1.4 weeks, was achieved in 13 of 14 patients. Ablation was successful with local activation times of 35 ± 4 ms, 5.8 ± 3.3 applications, 18.8 ± 7.5 minutes total cryo time, 9.4 ± 4.2 minutes fluoroscopy time, and 66.9 ± 26.1 minutes total procedure time, the latter two measures showing a reduction with number of patients treated. Three patients reported slight pain related to local pressure of the catheter on the RVOT wall. No pain was described related to delivery of cryothermal energy. Conclusions: Initial experience shows that focal VES/VT originating in the RVOT can be successfully treated using cryocatheter ablation. Acute and short term success rates, fluoroscopy times, and duration of procedure are comparable to conventional ablation techniques. A major advantage seems to be the virtual absence of ablation related pain.

    Notes: Introduction: Biventricular pacing has been shown to improve the clinical status of patients with congestive heart failure, but little is known about its influence on ventricular repolarization. The aim of our study was to evaluate the effect of biventricular pacing on ECG markers of ventricular repolarization in patients with congestive heart failure. Methods and Results: Twenty-five patients with congestive heart failure, sinus rhythm (SR), and complete LBBB (6 females; age 61 ± 8 years; NYHA class II–III; echocardiographic ejection fraction 21 ± 5%; QRS ≥ 130 ms) underwent permanent biventricular DDDR pacemaker implantation. A high-resolution 65-lead body-surface ECG recording was performed at baseline and during right-, left-, and biventricular pacing, and the total 65-lead root mean square curve of the QRST complex and the interlead QT dispersion were assessed. The QRS duration was increased during right (RV)- and left ventricular (LV) pacing (127 ± 26% and 117 ± 40%; P 〈 0.05), as compared to SR (100%) and biventricular pacing (93 ± 16%; ns). The QTc interval was increased during RV and LV pacing (112 ± 12% and 114 ± 14%; P 〈 0.05) as compared to SR (100%) or biventricular pacing (99 ± 12%). There was no effect on JT interval during all pacing modes. The Tpeak-end interval was increased during right (120 ± 34%; P 〈 0.01) and LV pacing (113 ± 29%; P 〈 0.05) but decreased during biventricular pacing (81 ± 19%; P 〈 0.01). A similar effect was found for the Tpeak-end integral and the Tpeak amplitude. QT dispersion was increased during right ventricular (129 ± 16 ms; P 〈 0.05) and decreased during biventricular pacing (90 ± 12 ms; P 〈 0.01), as compared to SR (114 ± 22 ms). Conclusions: Using a high-resolution surface ECG, biventricular pacing resulted in a significant reduction of ECG markers of ventricular dispersion of repolarization.
