    Notes: Abstract: The expression of 34 transmitter-related genes has been examined in the cholinergic neurones of rat striatal brain slices, with the aim of correlating gene expression with functional activity. The mRNAs encoding types I, II/IIA, and III α subunits of the voltage-sensitive sodium channels were detected, suggesting the presence of these three types of sodium channel. Similarly, mRNAs encoding all four α-amino-3-hydroxy-5-methyl-isoxazole-4-propionate (AMPA)-type glutamate receptor subunits and the NR1 and NR2A, 2B, and 2D subunits of the NMDA-type glutamate receptors were detected, suggesting that various combinations of these subunits mediate the cellular response to synaptically released glutamate. Other mRNAs detected included the NK1 and NK3 tachykinin receptors, all four known adenosine receptors, and the GABA-synthesising enzyme glutamate decarboxylase. Subpopulations of these cholinergic neurones have been identified on the basis of the expression of the NK3 tachykinin receptor in 5% and the trkC neurotrophin receptor in 12% of the cells investigated.

    Notes: Abstract Cytokines play a critical role in orchestrating and perpetuating inflammation in asthmatic airways and several specific cytokine and chemokine inhibitors are now in development for the treatment of asthma. Inhibition of IL-4 with soluble IL-4 receptors has shown promising early results in asthma. Anti-IL-5 antibody is very effective at inhibiting peripheral blood and airway eosinophils but does not appear to be effective in symptomatic asthma. Inhibitory cytokines, such as IL-10, interferons, and IL-12 are less promising because systemic delivery produces intolerable side effects. Inhibition of TNF-alpha may be useful in severe asthma. Many chemokines are involved in the inflammatory response of asthma, and small-molecule inhibitors of chemokine receptors are in development. CCR3 antagonists are now in clinical development for the treatment of asthma. Because so many cytokines are involved in asthma, drugs that inhibit the synthesis of multiple cytokines may prove to be more useful. Several such classes of drug are now in clinical development, and the risk of side effects with these nonspecific inhibitors may be reduced by the inhaled route of delivery.

    Notes: As with all systematic models of therapy, cognitive therapy distills a theory to the understanding of particular cases through the case formulation method. This article sets out criteria to evaluate whether cognitive case formulation follows the process of scientific inquiry, and it questions whether the formulation method meets these criteria. In terms of the evidence base for the cognitive theory that underpins cognitive case formulation, the research suggests that although the descriptive elements of cognitive theory are substantiated, the explanatory elements have received less support. In terms of the scientific status of the cognitive case formulation process, current evidence for the reliability of the cognitive case formulation method is modest, at best. There is a striking paucity of research examining the validity of cognitive case formulations or the impact of cognitive case formulation on therapy outcome. Implications for the clinical use of cognitive case formulation within a scientist-practitioner model are discussed, and potential programs of research to evaluate the case formulation method are described.

    Notes: A tandem time-of-flight (TOF-TOF) mass spectrometer comprised of two ion mirrors is described. The first ion mirror, which is a linear-field, single-stage mirror (MS1) with an intermediate collision cell, has been designed to provide the temporal focus necessary for the second, quadratic-field ion mirror (MS2) to function effectively. Due to the wide energy-range focusing capabilities of the quadratic field employed in the second ion mirror all the fragment ions can be collected in one spectrum without the need to step the reflecting working voltage of the MS2. The size of the active area of the microchannel plate detector used in the preliminary experiments was the limiting factor governing the collection efficiently of fragment ions. The use of the first ion mirror to provide temporal focusing of the precursor ion packet at the first focal point of the quadratic mirror used as the MS2 requires no alteration of the focusing conditions for different masses, in contrast to delayed extraction or postsource pulsed focusing. Precursor ions formed by matrix-assisted laser desorption/ionization were mass-selected with an ion gate located before the collision cell and the fragment ions were mass analyzed using the quadratic-field ion mirror. Experimental results demonstrating effective high-energy collision-induced dissociation of polymer and fullerene molecule-ions are presented. © 2002 American Institute of Physics.

    Notes: We report on development of a quantitative x-ray imager (QXI) for the national Inertial Confinement Fusion Program. Included in this development is a study of photocathode response as a function of photon energy, 2–17.5 keV, which is related to diagnostic development on the National Ignition Facility (NIF). The QXI is defined as being a quantative imager due to the repeated characterization. This instrument is systematically checked out, electronically as well as its photocathode x-ray response, both on a direct current and pulsed x-ray sources, before and after its use on a shot campaign. The QXI is a gated x-ray imager1 used for a variety of experiments conducted in the Inertial Confinement Fusion and Radiation Physics Program. The camera was assembled in Los Alamos and has been under development since 1997 and has now become the workhorse framing camera by the program. The electronics were built by Grant Applied Physics of San Fransisco, CA.2 The QXI has been used at the LANL Trident, LLNL Nova, and University of Rochester Laboratory OMEGA laser facilities. The camera consists of a grated microchannel plate (MCP), a phosphor coated fiberoptic faceplate coupled to film for data readout, along with high speed electronic pulsers to drive the x-ray detector. The QXI has both a two-strip and a four-strip detection head and has the ability to individually bias the gain of each of the strips. The timing of the QXI was done at the Trident short pulse laboratory, using 211 nm light. Single strip jitter was looked at as well and determined to be 〈25 ps. Flatfielding of the photocathode across the MCP was done with the Trident main laser with 150 J on a gold disk with a 1 ns. Spatial resolution was determined to be 〈5 μm by using the same laser conditions as before and a backlit 1000 lp/in. grid. The QXI has been used on cylindrical implosion work at the Nova Laser Facility, and on direct-drive cylinder mix and indirect-drive high convergence implosion experiments at OMEGA. Its two-strip module has provided the capability to look at point backlighters, as part of technique development for experiments on the NIF. Its next use will be in March 2000 with its off axis viewer nose at Omega, providing a perpendicular view of Rayleigh–Taylor spike dissipation. © 2001 American Institute of Physics.

    Notes: This paper explores the fundamental limits of the use of quartz tuning forks as force detectors in scanned probe microscopy. It is demonstrated that at room temperature, pressure, and atmosphere these force sensors have a noise floor of 0.62 pN/Hz and exhibit a root mean square Brownian motion of only 0.32 pm. When operated as a shear force sensor both dissipative and reactive forces are detected on approach to the sample. These forces are sufficient to reduce the amplitude of motion of the probe nearly to zero without physically contacting the surface. It is also demonstrated that conventional proportional-integral feedback control yields closed loop responses at least 40 times faster than their open loop response. © 2000 American Institute of Physics.
